Abstract

PURPOSE: A composition containing dihydro ergosterol for anti-wrinkling is provided to suppress collagen decomposition, and to improve an anti-wrinkling effect. CONSTITUTION: An anti-wrinkling agent contains dihydro ergosterol glucose as an active ingredient. A composition for anti-wrinkling containing dihydro ergosterol which suppresses collaganase activity. The quantity of collaganase induced by UV rays is reduced by dihydro ergosterol. The expression level of collaganase is concentration-dependently reduced.

Description

Wrinkle improvement composition containing dihydro ergosterol {Composition containing Dihydroergosterol promoting anti-wrinkles}

The present invention relates to a composition having anti-wrinkle action and anti-wrinkle effect containing dihydroergosterol. More specifically, dihydro ergosterol glucose from dihydro ergosterol, dihydro ergosterol glucose, and ergosterol glucose derivatives synthesized with saponin-based compounds was subjected to ultraviolet irradiation to human fibroblasts, followed by treatment with collagenase according to ultraviolet irradiation. The effect of reducing the amount of enzyme expression and inhibiting the degradation of collagen was confirmed. As a result, the present invention relates to active ingredients of cosmetics and functional additives that can help to improve wrinkles.

Collagenase is produced by ultraviolet stimulation (hereinafter referred to as UVB or UVB) in human fibroblasts (Kerwin, JF et al., J. Med. Chem. 1995, 38, 4343-4 362) . In addition, the breakdown of collagen is caused by UVB and wrinkles occur due to the decrease in elasticity of skin cells due to the breakdown of collagen. Collagenase is controlled by the activation of the transcription factor NF-KB through the phosphorylation of MAPK when UV stimulation in fibroblasts. In addition, collagen is known to be enhanced or inhibited by the regulation of smad 3,7 due to the activation of TGF-b. UVB can stimulate skin cells in a variety of ways, especially to break down collagen, a major constituent of the skin layer, and even cause skin cancer. Collagen is present in 70-90% type 1, the free collagen precursor is synthesized into the collagen in the subcutaneous layer. When fibroblasts are formed, they act as substrates between cells. When UV radiation is applied, the expression of collagenase, a collagen degrading enzyme, increases. Increased expression of collagenase promotes the breakdown of collagen, which results in the skin losing its elasticity rapidly. Therefore, functional substances that inhibit the expression of collagenase or increase the synthesis of collagen may help the wrinkle improvement effect due to UV stimulation.

An object of the present invention is to demonstrate a dihydroergosterol in the saponin-based derivatives to demonstrate the efficacy of reducing the expression of collagenase according to ultraviolet stimulation and to provide a material that can help to improve wrinkles.

After UV stimulation, dihydro ergosterol is treated by concentration to check the expression of collagenase to provide a material that can help to improve wrinkles.

The present invention relates to a composition having anti-wrinkle action and anti-wrinkle effect containing dihydroergosterol. Example 1 Collagenase Inhibitory Activity of Saponin-Based Compounds

Collagenase production activity of dihydro ergosterol was measured as follows. Human fibroblasts were treated with ultraviolet rays in an amount of 20 mJ / cm 2 and treated with dihydroergosterol by concentration, incubated for 24 hours, and the cells were separated to separate total RNA. Next, the expression level of collagenase was measured by RT-PCR method, and the amount of collagenase induced by ultraviolet light was confirmed to be reduced by dihydro ergosterol.

1) In order to compare the collagenase expression inhibitory activity of the saponin-based compounds, dihydroergosterol glucose, galactose, xylose, and ergosterol glucose were treated to fibroblasts at concentrations of 10 uM (Fig. 1a).
