Example 1 Preparation of Korean Herb Extract
Hanhubak was washed with purified water and dried, and then, 200 g of granulated Hanhubak powder was added to 1 liter of an aqueous 70% ethanol solution, boiled for 12 hours in an extractor equipped with a cooling condenser, filtered, and filtered through a 300 mesh filter cloth. Example 2 Preparation of Hanfu Park and Seomoktae Complex Extract
After washing Seomoktae with purified water and drying, 200g of granulated Seomoktae powder was put in 1 liter of 70% ethanol aqueous solution, boiled for 12 hours in an extractor equipped with a cooling condenser, filtered with 300 mesh filter cloth, and the filtrate was filtered at 4-15 ° C. [Test Example 1] Wrinkle improvement effect
The collagenase expression inhibitory effect (producing ability) of each of the extracts obtained in Examples 1, 2 and 3 was measured as follows by using retinoic acid known to inhibit collagenase expression as a positive control group. First, human fibroblasts were placed in a 96-well microtiter plate containing Dulbecco's Modified Eagle's Media (DMEM) medium containing 2.5% fetal bovine serum to 5,000 cells / well. [Test Example 2] Retinoic acid receptor alpha, gamma (RAR-α / γ) transcriptional activity effect
Retinoic acid receptor transcriptional activity was measured as follows for each of the extracts obtained in Examples 1, 2, and 3 using retinoic acid known to activate the retinoic acid receptor as a positive control group. First, the monkey kidney epithelial cell line CV-1 cells were inoculated into 24 well plates at a concentration of 4 × 104 cells / well, followed by incubation for 12-16 hours, before transfection.
