뉴질랜드 흰토끼의 등 피부에 하기 표 4에 나타낸 조성물을 1일 2회 간격으로 4일간 총 8회 도포를 2회 반복실시 하였다. 물질 도포 후, 홍반 및 가피형성 평점과 부종형성 평점 값을 누적시켜 피부누적자극 지수를 구하였다. 피부누적자극 지수는 표 3에 나타낸 판정 기준에 따라 평가하였고, 그 결과를 하기 표 4에 나타내었다. 결과에 나타낸 자극 지수는 일반적으로 많이 이용되는 Draize의 1차 자극 지수(Primary Irritation Index)의 산출 방법에 따른 것이다(Draize, J.H., Appraisal of the safety of chemical in foods, drugs and cosmetics). 자극완화율은 하기 수학식 1을 사용하여 구하였으며, 그 결과를 하기 표 4에 나타내었다.On the back skin of the New Zealand white rabbit, the composition shown in Table 4 was repeatedly applied twice a total of eight times for four days at intervals of two times a day. 시험에 들어가기 전 한달 동안에 비스테로이드성 항염증제 및 기타 스테로이드 제제를 복용한 경력이 없는 건강한 자원자를 대상으로 페퍼민트 추출물의 레티놀에 의한 피부홍반 유발억제 효과를 CTFA(The Cosmetic Toiletry and Fragrance Association)의 안전성 평가 지침(Safety Testing Guideline, 1981, p3)에 따라 평가하였다. 즉, 15명의 피검자의 전박 안쪽에 하기 표 5의 처방과 같이 제형화한 조성물을 하루에 두번씩 도포하고 (50 ㎕/1.5×1.5 ㎠), 흡수를 돕기 위해 랩(wrap)으로 3시간 동안 싸두었다. 시험 기간은 피검자의 반응 정도와 피부 상태를 고려하여 조정하였으며 최대 3주로 하였다. 검사는 매일 오전 도포 전에 시행하였고, 마지막 검사는 첩포 시행일로부터 8일까지 실시하였다. 자극의 회복 정도를 반영하기 위해 첩포 종료 후에도 계속 평가하였고, 홍반이 완전히 사라지고 색소 침착만 남은 경우에는 점수를 부여하지 않았다. 각 검사 시점에서 하기 표 6의 판정 방법에 따라 자극정도를 평가하였고, 그 합으로부터 상기 수학식 1에 의해 자극완화율을 계산하여 그 결과를 하기 표 7에 나타내었다.A guideline for evaluating the safety of the Peppermint Extract's retinol-induced skin erythema-induced effects in healthy volunteers who had never taken nonsteroidal anti-inflammatory drugs and other steroid preparations for a month before entering the study.
