Description
The present invention relates to a ginkgo biloba extract containing a high concentration of ginkgo biloba extract, and more particularly, by containing a high concentration of ginkgo biloba extract as an active ingredient and using a solubilizer at a specific concentration. It can be solubilized even if it contains a high concentration, it ensures the stability of terpene trilactones (TTL), which is particularly decomposed during the manufacturing process, obtains the desired therapeutic effect with only a small dose, and has excellent characteristics in terms of economy. X relates to high concentration-containing injections.
Ginkgo biloba X is typically developed and commercialized in 1971 by Willmar Schwabe AG, Germany, and Beaufour-Ypsen Group, France, and is used as a treatment for brain and peripheral circulatory disorders. The main active ingredients of ginkgo biloba extract are known as ginkgoflavone glycosides (GFG) and terpene trilactones (TTL). TTLs include gingkolides A, B, C and bilobalide. . GFG has antioxidant and free radical scavenging effects. Gingcolides A, B, and C in TTL antagonize platelet activating factor (PAF), decreases blood viscosity due to platelet aggregation inhibition, and inhibits NO (nitric oxide) production. It is known to have an effect of reducing inflammatory response, and bilibaride has an effect of reducing inflammatory response and neuronal cell protection by inhibiting NO production.
However, ginkgo biloba x is low solubility in water, when the preparation of the injection in aqueous solution may cause problems of generation and storage of precipitates due to low solubility, and when injected into the human body may cause vascular obstruction. For this reason, Ginkgo biloba X injections (Tana Minju ™, Yu-yu), which are commercially available in Korea, contain 17.5 mg (0.35 W / V%) of low concentration ginkgo biloba extract in 1 ampule (5 ml). Willmar Schwabe AG also commercializes and sells injectables containing a low concentration of 0.35 W / V% ginkgo biloba extract. Low-dose injections are intended to be administered intramuscularly by intramuscular injection of 5 to 10 ml per day or by mixing 25 to 50 ml into intravenous or intravenous fluids. It is also expensive.
In Korea Patent Registration No. 154984 of Willmar Schwabe AG and Korea Patent Registration No. 76111 of SK Chemical Co., Ltd., Ginkgo leaf extract was dissolved in water to ensure the stability of the active ingredient, especially GFG, in Ginkgo leaf extract. Rapid freezing, and then freeze-dried under reduced pressure to prepare a powder, and separately prepared a solvent to develop a freeze-dried powder injectable form that can be dissolved in the solvent used. These lyophilized powder injections may increase the stability of the active ingredient in the ginkgo biloba extract, but it is inconvenient because it must be dissolved in a separately prepared solvent when used, and bubbles are generated when shaking during the melting process. It is difficult to administer and should be left until foam is gone. In addition, the manufacturing process for freeze drying is uneconomical, leading to an increase in the cost for commercialization from an industrial point of view.
Therefore, it should be easy to use and minimize the pain and discomfort to the patient, while containing high concentrations of ginkgo biloba extract can suppress precipitation, and the high concentration of ginkgo biloba extract containing high concentration of ginkgo biloba that can ensure the stability of TTL as well as GFG. Development is urgent.
Therefore, the present inventors have studied to solve the above problems, as a result, solubilized ginkgo biloba extract as an active ingredient to a high concentration, and ensure the stability of TTL, and also contains a ginkgo biloba extract as an active ingredient and a solubilizer By using the appropriate concentration, the solubility was increased more than 5 times compared to the injection containing the low concentration of ginkgo biloba extract, preventing the deterioration of the stability of TTL during the manufacturing process, and improving the storage stability. The present invention has been completed by developing an injection method containing a high concentration of ginkgo biloba extract and a method for preparing the same, which can provide a desired therapeutic effect as well as economical benefits.
Therefore, an object of the present invention is to provide a ginkgo biloba extract high concentration injection and a method for preparing the same, which prevents the stability of TTL and improves storage stability.
본 발명은 전체 1 ㎖ 용량을 기준으로, 은행엽 엑스를 1.75 ~ 5.83 W/V%, 가용화제로 프로필렌글리콜, 에탄올, 글리세린, 폴리에틸렌글리콜 또는 이의 혼합물을 20 ~ 60 V/V% 함유하며, pH 4 ~ 7.5인 은행엽 엑스 고농도 함유 주사제 및 이의 제조방법을 그 특징으로 한다.The present invention contains ginkgo biloba extract 1.75 ~ 5.83 W / V%, solubilizing agent 20 ~ 60 V / V% propylene glycol, ethanol, glycerin, polyethylene glycol or a mixture thereof, based on the total volume of 1 ml, pH 4 It is characterized by a ginkgo leaf extract containing a high concentration of ~ 7.5 and a preparation method thereof.
The present invention will be described in more detail as follows.
The present invention contains a high concentration of ginkgo biloba extract as an active ingredient and the use of a solubilizer in a specific concentration, so that it can be solubilized even if the ginkgo biloba extract contains a high concentration compared to the conventional ginkgo biloba extract containing low concentration, TTL during the manufacturing process The present invention relates to a ginkgo biloba extract containing high concentrations of terpene trilactones, which can achieve the desired therapeutic effect with a smaller dose, and have excellent economical characteristics.
The method for preparing the ginkgo biloba extract high concentration-containing injection according to the present invention will be described in detail as follows.
First, Ginkgo biloba extracts 1.75 to 5.83 W / V% and solubilizer 20 to 60 V / V%, which are the active ingredients, were dissolved in an appropriate amount of water for injection, and then the final product was dissolved. The pH of the mixture is adjusted to 4.0 to 7.5, followed by a sterile membrane filtration process, followed by filling and sealing. At this time, the aim is to reduce the dosage of the injection to a minimum of 1/5 or less, to reduce the 25 ml dosage of the ginkgo biloba x injection currently on sale to 2 ml and to reduce the 50 ml dosage to 3 ml. The preparations were reviewed for ginkgo biloba x 1.75-5.83 W / V%. If an injection is made using less than 20 V / V% of a solubilizer, ginkgo biloba extract may be precipitated when mixed with an infusion solution to instill drip, and the use of an excessive solubilizer may cause side effects such as hemolytic action. Therefore, a high concentration of ginkgo biloba x 5.83 W / V% injection was used up to 60 V / V% to prevent precipitation when mixed with the fluid. In addition, if the pH is adjusted to less than 4.0, the stability of the TTL may be increased, but may be toxic by excessive acidic conditions, and the pH of the blood is 7.5, so that the pH of the injection is 4.0 to 7.5.
Ginkgo biloba extract, an active ingredient, is approved by the Korea Food and Drug Administration for use in foods and medicines. It contains GFG and TTL as indicators. In addition, it is preferable to use propylene glycol, ethanol, glycerin, polyethylene glycol or a mixture thereof as the solubilizer. In addition, conventional pharmaceutically acceptable additives (an analgesic agent, preservative, pH adjuster, stabilizer, etc.) may be further added to the composition forming the composition and component ratio. For example, benzyl alcohol, chlorobutanol, procaine hydrochloride, etc., as a preservative, chimerosal, benzalkonium chloride, cresol, phenol, benzyl alcohol, methyl paraben, propyl paraben, mercury nitrate, butyl paraben, chloro PH adjusting agents, such as cresol and benzenetonium chloride, include buffers, weak acids and salts thereof, strong acids and salts thereof, weak bases and salts thereof, strong bases and salts thereof, and stabilizing agents such as edetic acid and salts thereof, ascorbic acid and ascorbic acid. Corbic acid ester, tocopherol, tocopherol acetate, sodium bisulfite, sodium bisulfite, sodium pyrosulfite, sodium metabisulfite, thiourea, sodium bisulfide and the like are preferably used.
Then, the solubilized injection solution is subjected to only sterile membrane filtration without high pressure steam sterilization to prepare the final injection. The problem of lowering the TTL content of the existing ginkgo biloba extract containing low concentration and the problem of precipitation of the ginkgo biloba extract containing the high concentration of ginkgo biloba extracts was solved by performing the filtration process.
The present invention reduces the dosage to 1/5 or less by increasing the solubility of ginkgo biloba extract by more than five times compared to conventional injections. For example, 25 ml, which is the conventional dosage of commercially available injections, is reduced to 2 ml. To reduce patient discomfort.
As described above, the injection containing a high concentration of ginkgo biloba extract of the present invention solves both the discomfort felt by patients and the problem of solubilization when administering a conventional injection containing a low concentration of ginkgo biloba extract, It is an invention that improved the stability of TTL and room temperature storage stability.
Hereinafter, the present invention will be described in more detail based on the following examples, but the present invention is not limited thereto.
Preparation Example: Preparation of Ginkgo Leaf X
Ginkgo biloba extract was prepared by extracting ginkgo biloba with a mixed solution of methanol or water and acetone or alcohol, adding an inorganic salt, and then extracting it again with an organic solvent, and then extracting and purifying the active active material by removing impurities from the obtained crude extract. .
Examples 1-4: Injection Preparation
The composition of the ginkgo biloba X-containing injection is shown in Table 1 below, and typically, the preparation of the injection of Example 4 is as follows.
To 4 L of water for injection, 1.579 L of 95% ethanol (1.5 L of ethanol) and 3.5 L of propylene glycol are mixed and stirred to homogenize. 14.4 g of methyl-paraben and 1.6 g of propyl-paraben were added to the solution, and the mixture was stirred and dissolved. The final pH of the product was adjusted to NaOH solution to 4 to 7.5 and then water for injection was added to 10 L. Aseptic membrane filtration sterilization was followed by filling into vials.
Solubilizers were used to solubilize ginkgo biloba extracts from 1.75 to 5.83 W / V%. The solubilizer used at this time is 20 ~ 60 V / V%. Injectable pH of the present invention was adjusted to 4 ~ 7.5. Example 3 and Example 4 reduced the dose corresponding to 25 ml of commercially available formulations to 2 ml.
Total TTL content in
Conventional ginkgo biloba extracts containing low concentrations were measured only GFG as the main active ingredient, and the TTL content was not evaluated. Therefore, the total TTL content in commercially available ginkgo biloba extracts was measured and evaluated. For the analysis, HPLC was used, and TTL components were measured for gingkolides A, B, C, and bilobalide.
Commercially available ginkgo biloba extracts have a low total TTL content of 2.56% and no bilobalide.
Test Example 1: Stability Check
Conventional ginkgo biloba extracts containing low concentrations were prepared by solubilizing ginkgo biloba extracts, followed by aseptic membrane filtration sterilization and autoclaving. Autoclaving alone is sufficient for sterilization, but sterile membrane filtration is added to ensure sterilization and sterile membrane filtration is not essential.
따라서, 멸균 제조공정 중의 GFG(gingkoflavone glycosides)와 TTL(terpene trilactones)의 함량 변화를 알아보기 위하여 다음과 같은 실험을 실시하였다.Therefore, the following experiments were conducted to determine the changes in the content of GFG (gingkoflavone glycosides) and TTL (terpene trilactones) during the sterilization process.
The ginkgo biloba extract was solubilized in the composition shown in Table 3, and then the contents of GFG and TTL were measured before and after autoclaving (121 ° C., 15 minutes) and aseptic membrane filtration sterilization (0.2 μm).
Assay, GFG measured quercetin, kaempferol, and isorhamnetin using HPLC, and TTL measured gingcolides A, B, C and bilobalide.
According to the results of Table 4, when high-pressure steam sterilization (121 ℃, 15 minutes), the content of TTL was observed to decrease, in particular, the content of bilobaride was observed. It was found that the use of only a sterile membrane filtration sterilization process (0.2 μm) without sterilizing a high pressure steam sterilization process as a sterilization process was a manufacturing process capable of suppressing a decrease in the content of TTL.
Test Example 2: Confirmation of room temperature storage stability
In countries with four seasons, such as Korea, ginkgo biloba may be deposited from the injection during the seasons, such as winter. In order to observe the phenomenon of ginkgo leaf extract at room temperature, especially at low temperature, the following experiment was conducted.
Injectables containing high concentrations of ginkgo biloba extract were left at 4 ° C. for 2 weeks and precipitation was observed.
According to the results of Table 5, precipitation was observed when the aseptic membrane filtration process was not performed, but was not precipitated when the sterile membrane filtration process was performed. That is, by performing a sterile membrane filtration process it can effectively suppress the precipitation during storage at room temperature it can be made a product with improved storage stability.
Comparative Example 2 Preparation of Injection and Confirmation of Room Stability
In order to evaluate the change in the content of GFG and TTL during the injection preparation process and the low temperature storage stability according to the use of the solubilizer and the surfactant, a comparative example containing a high concentration of ginkgo biloba extract was prepared. Precipitation was observed when left at 4 ° C. for 2 weeks.
Solutol HS15 is a surfactant sold by BASF, and it is a solubilizing agent commonly used in the industry because it is known to have solubilizing ability and has the least side effects such as hemolytic action and blood histamine secretion. . Solutol HS15 could be used to reduce the amount of solubilizer, but precipitation was observed in both cases with and without aseptic membrane filtration when left at 4 ° C for 2 weeks. However, when the aseptic membrane filtration sterilization process (0.2 ㎛) was able to suppress the decrease in the content of TTL.
Conventional ginkgo biloba extracts containing low concentrations have been combined with autoclave filtration and autoclaving to ensure sterilization during the manufacturing process, but sterile membrane filtration is not essential. In the case of the ginkgo biloba extract containing high concentration of Solutol HS15 as a surfactant in the comparative example, aseptic membrane filtration process could not solve the precipitation problem at room temperature.
As a matter of example, propylene glycol, ethanol, glycerin, polyethylene glycol, or a mixture thereof is used as a solubilizer to improve the precipitation problem at room temperature storage by aseptic membrane filtration process.
As described above, the ginkgo biloba X-concentrated injection according to the present invention solves all the problems caused by the existing low-concentration injection (patient pain, discomfort, TTL stability, precipitation at room temperature, etc.) It is expected that the availability of the award is very high.
