Claims (13)
- L-쓰레오닌을 생산할 수 있고, iclR 유전자가 불활성화된 미생물.
- 제1항에 있어서, 엔테로박테리아세 (Enterobacteriaceae) 과에 속하는 미생물.
- 제2항에 있어서, 대장균인 미생물.
- 제3항에 있어서, 상기 대장균이 L-메티오닌 유사체에 대한 내성을 가지며, 메티오닌 영양요구성, L-쓰레오닌 유사체에 대한 내성, 이소루이신 리키형 요구성, L-라이신 유사체에 대한 내성 및 α-아미노부티릭산 내성을 가짐을 특징으로 하는 미생물.
- 제4항에 있어서, L-메티오닌 유사체가 D,L-에티오닌, 노르루이신, α-메틸메티오닌 또는 L-메티오닌-D,L-설폭시민임을 특징으로 하는 미생물.
- 제4항에 있어서, L-쓰레오닌 유사체가 α-아미노-β-히드록시 발레릭산 또는D,L-쓰레오닌 히드록사메이트임을 특징으로 하는 미생물.
- 제4항에 있어서, L-라이신 유사체가 S- (2-아미노에틸)-L-시스테인 또는δ-메틸-L-라이신임을 특징으로 하는 미생물.
- 제1항에 있어서, 염색체 상의 pckA 및 aspA 유전자가 불활성화된 미생물.
- 제1항에 있어서, 내재적 포스포에놀 피루베이트 카르복실레이즈 (ppc) 유전자와 thrA, thrB 및 thrC 유전자 외에 추가로 1 카피 이상의 포스포에놀 피루베이트 카르복실레이즈 (ppc) 유전자와 thrA, thrB 및 thrC 유전자가 염색체 DNA 중에 삽입된 것을 특징으로 하는 미생물.
- 제1항에 있어서, 대장균 (Escherichia coli) FTR0627 (KCCM-10545), 대장균 (Escherichia coli) FTR3627 (KCCM-10537), 및 대장균 (Escherichia coli) FTR0808 (KCCM-10546)로 이루어지는 군으로부터 선택되는 미생물.
- 불활성화된 iclR 유전자 또는 그의 DNA 단편을 제조하고 L-쓰레오닌을 생산할 수 있는 미생물에 도입시켜 상기 미생물의 염색체 상에 존재하는 iclR 유전자와 재조합시키고, iclR 유전자가 불활성화된 미생물을 선별하는 것을 특징으로 하는, L-쓰레오닌의 생산용 미생물을 제조하는 방법.
- 제11항에 있어서, 상기 불활성화된 iclR 유전자 또는 그의 DNA 절편이 항생제 마커 (loxpKAN)를 포함한 카세트를 iclR 유전자 내부에 삽입하여 제조된 것임을 특징으로 하는 방법.
- 제1항 내지 제10항에 따른 미생물을 배양하고, 그 배양물로부터 L-쓰레오닌을 분리하는 것을 특징으로 하는, L-쓰레오닌을 생산하는 방법.
