PURPOSE: Provided is an antioxidant comprising 3,4,5-trihydroxybenzaldehyde as an active ingredient, which has an improved free radical elimination power than that of existing material, and excellent anti-acidification of pig fat and palm oil, and thus can be widely used as an antioxidant such as additives for fat and fatty oil processed foods. 뱀무에서 추출한 메탄올 엑기스를 클로로포름과 에틸아세테이트 및 증류수로 순차용매분획한 다음, 각 분획에 대하여 디페닐피크릴히드라질(diphenylpicrylhydrazyl, 이하, `DPPH`라 함)을 이용하여 자유라디칼 소거력을 평가하여 항산화 활성을 조사한 결과, 에틸아세테이트 분획에서항산화 활성을 확인하였다. 상기 항산화 활성의 유효성분을 순수하게 분리하기 위하여 세파덱스 LH-20 컬럼(50 x 5cm)으로 8ml씩 300개의 분획을 확보한 다음, 이를 HPLC(Supelcosil LC-18, 250 x 21.2mm, 5㎛, wavelength 320nm)를 이용하여 정제하였다. 상기 정제된 물질에 대하여 IR, NMR, 질량분석기 등의 분석기기를 이용하여 분석한 결과, 유효성분이 3,4,5-트리히드록시벤즈알데히드(3,4,5-trihydroxybenzaldehyde, THBA) 임을 확인하였다(참조: 도 1-1, 도 1-2 및 도 1-3).Methanol extract extracted from bamboo radish was extracted sequentially with chloroform, ethyl acetate and distilled water, and then free radical scavenging ability was evaluated for each fraction by using diphenylpicrylhydrazyl (hereinafter referred to as' DPPH`). 전술한 DPPH의 소거력을 추적하여 분리·정제한 상기 3,4,5-트리히드록시벤즈알데히드에 대하여 표준 항산화물질과 함께 항산화력을 비교평가하여, 그 결과를 표 1에 정리하였다. 표준 항산화물질로서는 동일 용도로 당업계에 공지된 BHA, BHT, α-토코페롤, 로즈마린산 등이 사용되었다. 상기 표준 항산화물질들과 실시예 1에서 분리·동정한 3,4,5-트리히드록시벤즈알데히드가 일정한 농도로 용해된 에탄올 용액(10mg/50ml)을, 특정한 농도(10mg/50ml)의 DPPH/에탄올 용액에 천천히 적가하면서, 광도계(spectrophotometer)를 이용하여 450nm파장에서 DPPH 용액의 흡광도를 50%로 감소시키는데 필요한 상기 항산화물질들이 용해된 에탄올 용액의 양을 측정하여, 이를 활력지수로 하였다.The 3,4,5-trihydroxybenzaldehyde separated and purified by tracking the scavenging power of the above-described DPPH was compared with the standard antioxidants, and the antioxidant power was evaluated.
