또한, β-hexosaminidase 측정 결과 도 11에서 도시된 바와 같이 실험물질의 RBL-2H3 비만세포의 탈과립에 미치는 효과를 확인하기 위해 IgE에 감작된 비만세포에서 방출된 히스타민의 양을 측정하는 것이 일반적이나 비만세포에서 방출하는 히스타민의 농도는 낮고 측정하는 과정이 복잡하여 β-hexosaminidase 농도를 측정 하였다. β-hexosaminidase는 비만세포나 호염구의 히스타민이 저장된 과립 안에 존재하며 알레르기 반응을 통해 세포 밖으로 방출되므로 탈과립의 지표로 알레르기 억제물질의 측정에 이용된다. IPEE가 β-hexosaminidase 생성량에 미치는 영향을 RBL-2H3 세포에서 확인한 결과, 도 11과 같이 DNP-IgE와 DNP-HSA에 의해 감작된 control군에서 분비된 β-hexosaminidase 억제능을 0%로 볼 때 IPEE는 500, 250, 125 μ농도에서 83.05 ±6.6, 60.69 ±3.08, 52.58 ±3.08%의 억제효과를 확인하였다. 실험물질의 농도가 증가하면서 β-hexosaminidase 분비량이 감소하는 양상을 나타내었다.In addition, as shown in Figure 11 as a result of β-hexosaminidase measurement, it is common to measure the amount of histamine released from mast cells sensitized to IgE to confirm the effect of the test substance on degranulation of RBL-2H3 mast cells. RAW 264.7 cell을 이용한 항염증 평가(세포독성평가 )결과 도 13에서 도시된 바와 같이 IPEE은 31.25∼500 μ에서 모두 80% 이상의 높은 세포 생존율을 나타냈으며 최고 농도인 500 μ에서 82.6%를 나타내었다. 따라서 이후 실험에서는 세포 생존 및 증식에 영향을 미치지 않는 농도범위인 500 μ이하 농도에서 수행하였다. 삭제delete 삭제delete 미성숙 감의 꼭지 부분을 제거하면서 흐르는 물에 세척을 하는 세척단계(S100); 와,
상기 세척한 미성숙 감을 1 내지 3cm 크기로 절단을 하는 절단단계(S200); 와,
상기 절단한 미성숙 감 100kg을 용기 외부에 전열 화덕(60)이 구비된 상부용기(10)에 투입을 하고, 입자와 미립자의 이물질을 2단계에 걸쳐 여과시킬 수 있는 여과필터(41,42)가 일체형으로 고정된 필터아세이(40)를 유출이 되지 않도록 상부용기(10)에 결합 고정하여 내용물의 유출을 방지하는 유출방지단계(S300); 와,
추출액이 임시 저장되면서 상등액이 배출되도록 하는 배출구(21)를 구비하는 하부용기(20)를 상부용기(10)의 저면에 고정하되 결합링크(30)를 이용하여 밀폐가 되도록 결합고정시키는 상/하부용기 결합고정단계(S400); 와,
상기 상/하부용기 결합 고정후 상부용기의 외부에 구비된 전열 화덕(60)에 전원을 인가하여 600 내지 700℃ 조건에서 5 내지 7일 동안 가열을 하는 가열단계(S500); 와,
