인간의 질병연구에 있어서 임상실험은 없어서는 안 될 필수 단계이다. 임상실험이 가장 효과적인 실험 방법임에는 틀림 없지만, 여러 가지 위험한 요소들과 윤리적 문제들 때문에 극히 제한적으로만 가능한 것이 현실이다. 이를 대체하기 위해 여러 동물모델들이 사용되고 있지만 인체와는 상이하기 때문에 동물실험에 성공을 한다고 하더라도 인체와 동일하게 적용 되리란 확신을 얻기가 힘들다. 따라서 인체와 비슷한 조건을 가지는 동물모델의 제작은 암이나 에이즈 등의 불치병 치료 연구에 있어서 커다란 발전을 가져올 것이라 사료되어 우수한 동물모델 개발은 계속 활발히 진행 중이다. 그 중에서도 인간의 유전자와 97% 유사한 Mouse는 암 연구에 있어서 매우 광범위하게 사용되고 있다. 특히, 1960년대에 흉선이 없는 Nude mouse가 활성화된 T-림프구의 결핍으로 면역거부 반응이 약하다는 것이 알려지면서, 암세포를 Nude Mouse에 이식하는 방법의 연구가 활성 되었다. 그리고, 1983년에는 Bosma에 의해 Severe Combined Immunodeficiency (SCID) Mouse가 보고되었는데, SCID Mouse는 림프구의 분화과정에 있어 B세포와 T세포의 항체(Antigen)을 인식하는 부위인 V(D)J를 조합하는데 관련된 DNA의 이중 가닥 파손 수리 활성(Break Repair Activity)이 결여되어 B세포와 T세포의 기능을 제대로 하지 못하는 모델이다. 이러한 B와 T세포 기능의 결함 때문에 SCID Mouse는 사람의 세포를 받아들이는데 거의 거부반응이 없으므로 SCID Mouse는 암을 비롯한 면역질환, 장기이식 및 감염 등 여러 분야의 연구에 널리 이용되고 있다. Non Obese Diabetic-Severe Combined Immunodeficiency (NOD/SCID) Mice는 선천적인 면역(Innate Immunity)이 결핍된 NOD Mouse와 후천적인 면역(Acquired Immunity)이 결핍된 SCID Mouse에서 유래된 Mouse Model로 NOD Mouse의 기능적 특징(Phenotype)인 자연 독성 세포(Natural Killer Cell)의 활성 저하, 단핵백혈구(Monocytes)의 사이토카인(Cytokine)으로의 분비기능장애, 보체 제5성분(Complement5, C5)의 활성 결여 등을 나타내며 SCID Mouse의 기능적 특징(Phenotype)인 B세포와 T세포의 기능 결여도 동시에 나타낸다. 인간말초혈액단핵구(Human Peripheral Blood Mononuclear Cell, huPBMC)는 Lymphocyte나 Monocyte 같은 원형의 Nucleus를 갖는 Blood Cell로 외부 물질에 의한 감염에 적응하거나 저항하는 Immune System에서 중요한 역할을 하는 세포이다. 이것은 Ficoll-Paque를 이용하여 전혈(Whole Blood)로부터 분리가 가능하며 CD4+ Cell 등을 포함하고 있기 때문에 HIV Research에 사용가능하며 암 연구에 있어서도 인체와 비슷한 면역체계를 제공할 수 있다는 특징을 보유하고 있는 세포이다.
