Abstract
본 발명은 열안정성 펩타이드를 이용한 니신-금 나노입자-환원된 그래핀 옥사이드 나노복합체의 제조방법에 관한 것으로, 본 발명에 따른 제조방법은 니신-금 나노입자-환원된 그래핀 옥사이드 나노복합체를 싱글팟(single pot)으로 매우 짧은 시간에 효율적으로 제조할 수 있으며, 본 발명에 따른 상기 나노복합체는 메모리 장치용 박막 트랜지스터 및 광열 치료 용도로 사용 가능하며, 우수한 전하 전달 특성 및 광열 효과가 있다.
Description
본 발명은 열안정성 펩타이드를 이용한 니신-금 나노입자-환원된 그래핀 옥사이드 나노복합체의 제조방법에 관한 것이다.
지난 20년 동안 흑연(graphite) 및 탄소 나노 튜브와 같은 탄소 물질에 대한 연구가 광범위하게 이루어져 왔다. 그래핀(graphene)은 공액 sp2 탄소로 이루어진 허니콤(벌집) 구조를 갖는 이차원(2-D)의 탄소 물질로서, 독특한 물리적, 화학적, 전기적, 광학적 및 생물학적 특성으로 인하여 과학계 및 다양한 산업 분야에서 주목을 받고 있고, 바이오 센서, 생체분자 캐리어, 태양 전지, 촉매제, 광전자, 배터리, 태양 전지(연료 민감성 또는 유기성) 및 에너지 저장 용도로 사용되고 있다.
그래핀 또는 그래핀 옥사이드(graphene oxide; GO)의 대량 생산을 위한 천연 흑연의 화학적 전환 방법에 대한 연구가 주로 이루어지고 있으며, 그래핀을 합성하기 위한 방법으로 다양한 물리적, 화학적 및 생물학적 방법이 개발되었다. 또한, 그래핀 시트(graphene sheets)를 분리하여 그 특성을 변화시키거나 변화시키지 않고 단일 시트를 형성하기 위한 방법뿐만 아니라 환원된 형태의 그래핀 옥사이드를 합성하기 위한 방법이 개발되고 있다. GO를 환원시키기 위한 물질로 할로겐화 수소산(hydrohalic acid), 하이드라진(hydrazine), p-페닐렌다이아민(p-phenylenediamine), 수소화붕소나트륨(sodium borohydride) 및 하이드로퀴논(hydroquinone)과 같은 화학물질이 사용되어 왔으나, 이를 이용하여 환원된 그래핀 옥사이드(reduced graphene oxide; rGO)는 생물학적 물질에 대한 파괴성 및 독성으로 인해 생물학적인 분야에는 이용되지 않았다. 소니케이션(sonication)은 GO를 박리 및 환원시키는 것으로 밝혀졌으나, 높은 초음파 주파수가 요구되므로 GO의 물리 화학적 특성에 영향을 주는 구조적 손상이 발생할 수 있다. 따라서, 친환경적이고 신속하게 환원된 그래핀을 제조할 수 있는 방법의 개발이 요구되며, 친환경적인 GO 환원을 위하여, 아미노산, 단백질(소혈청알부민), 미생물, 식물추출물, 당류 및 아스코르빈산과 같은 생체분자가 사용되고 있다.
한편, GO의 전기, 열 및 광학 특성을 향상시키기 위하여, 은, 구리, 백금 및 금과 같은 금속 나노입자들을 허니콤 구조에 장식하는 기술이 연구되고 있으며, 특히, 금 나노입자(AuNPs)가 장식된 그래핀 옥사이드(AuNPs-GO) 복합체는 촉매, 센서, 진단 및 치료 분야에서 광범위하게 이용되고 있다. 금속 나노입자가 장식된 그래핀 옥사이드는 독특한 플라즈몬(plasmonic) 특성으로 인해 다양한 광전자(optoelectronic) 용도로 사용되고 있으며, 광전자 장치에서 빛-물질 상호 작용을 향상시키는데 필수적이다. 그래핀에 AuNPs를 결합시키면 플라즈몬 효과가 생겨 광 흡수를 증가시키고 광전자 장치의 성능을 향상시킬 수 있다. 또한, 그래핀의 AuNP 결합에서의 전하 제한(charge confinement)은 전하 트래핑(charge trapping) 및 디-트래핑(de-trapping)으로 인한 메모리 특성(memory characteristics)을 나타내는 전기적 이중 안정성(electrical bi-stability)을 생성할 수 있다.
금속 나노입자와 rGO의 복합체를 형성하기 위한 방법 가운데, 금속 나노입자와 GO를 개별적으로 환원시킨 후 혼합하여 복합체를 형성하는 방법은 공정 시간이 오래걸린다는 단점이 있다. 다른 방법으로 금속 나노입자 전구체 및 GO를 단일 반응으로 환원시키는 방법이 있다. 그러나 AuNPs-rGO 복합체의 신속한 생합성에 초점을 둔 연구는 현재까지 거의 이루어지지 않았다.
이에, 본 발명자들은 금 나노입자(AuNPs)가 도입된 환원된 그래핀 옥사이드(rGO)를 형성하기 위한 GO 및 Au3 +의 신속하고 동시적인 원스텝(one-step) 바이오-환원 방법을 개발하고, 상기 방법으로 제조된 나노복합체의 특성들을 확인하여 본 발명을 완성하였다.
따라서, 본 발명의 목적은 니신-금 나노입자-환원된 그래핀 옥사이드 나노복합체의 제조방법을 제공하는 것이다.
본 발명의 다른 목적은 상기 제조방법으로 제조되는 니신-금 나노입자-환원된 그래핀 옥사이드 나노복합체를 제공하는 것이다.
본 발명의 또 다른 목적은 상기 니신-금 나노입자-환원된 그래핀 옥사이드 나노복합체를 포함하는 비휘발성 메모리 장치, 광 치료용 조성물 및 암 치료용 조성물을 제공하는 것이다.
상기 목적을 달성하기 위하여,
본 발명은 완충액에 금 나노입자 전구체를 포함하는 수용액 및 니신 펩타이드(nisin peptide)를 첨가한 혼합물을 인큐베이팅하는 단계(단계 1); 및 그래핀 옥사이드를 상기 혼합물에 분산시키고 오토클레이빙하는 단계(단계 2);를 포함하는 니신-금 나노입자-환원된 그래핀 옥사이드 나노복합체의 제조방법을 제공한다.
또한, 본 발명은 상기 제조방법으로 제조되는 니신-금 나노입자-환원된 그래핀 옥사이드 나노복합체를 제공한다.
또한, 본 발명은 상기 니신-금 나노입자-환원된 그래핀 옥사이드 나노복합체를 포함하는 비휘발성 메모리 장치를 제공한다.
또한, 본 발명은 상기 니신-금 나노입자-환원된 그래핀 옥사이드 나노복합체를 포함하는 광 치료용 조성물을 제공한다.
또한, 본 발명은 상기 니신-금 나노입자-환원된 그래핀 옥사이드 나노복합체를 포함하는 암 치료용 조성물을 제공한다.
본 발명에 따른 니신 펩타이드를 이용한 제조방법은 니신-금 나노입자-환원된 그래핀 옥사이드 나노복합체를 싱글팟(single pot)으로 매우 짧은 시간에 효율적으로 제조할 수 있으며, 본 발명에 따른 상기 나노복합체는 메모리 장치용 박막 트랜지스터 및 광열 치료 용도로 사용 가능하며, 우수한 전하 전달 특성 및 광열 효과가 있다.

이하, 본 발명을 상세히 설명한다.
본 발명에서는 매우 짧은 시간에 높은 온도에서 GO를 환원시키기 위한 바이오환원제(bioreductants)로서 열안정성 니신 펩타이드(nisin peptide)를 사용하였다.
본 발명의 상기 니신(nisin)은 121℃에서 15분간 활성을 나타내며, -20℃에서 안정한 상태를 나타내는 열안정성 펩타이드로, 34개의 아미노산(ITSISLCTPGCKTGA- LMGCNMKTATCHCSIHVSK)으로 이루어져 있으며, 우유-기반 제품 및 야채로부터 분리되는 락토코커스 락티스(Lactococcus lactis) 아종 락티스(lactis)로부터 생산되는 란티바이오틱(lantibiotic)이다. 또한, 니신은 식품 방부제로 사용되는 FDA에서 승인받은 항균성 펩타이드이며, 그램 양성 미생물에 대한 광범위한 항균 활성을 갖는다.
본 발명자들은 오토클레이브(autoclave)에서 열안정성 항균성 니신 펩타이드(nisin peptide)를 사용하여 니신-금 나노입자-환원된 그래핀 옥사이드(nisin-gold nanoparticles-reduced graphene oxide; NAuNP-rGO) 나노 복합체를 원스텝(one-step)으로 합성하였다(실시예 2 참조). 본 발명의 NAuNP-rGO 나노 복합체의 합성에는 생체 의료 분야에 사용되지 않는 독성 화학물질이나 가혹한 반응조건은 이용되지 않았다. AuNPs 및 rGO는 싱글팟(single pot)으로 매우 짧은 시간에 형성되었고, 15분 동안의 반응으로부터 분산된 NAuNPs와 결합된 rGO를 수득하여 본 발명을 완성하였다.
따라서, 본 발명은 완충액에 금 나노입자(gold nanoparticles; AuNPs) 전구체를 포함하는 수용액 및 니신 펩타이드(nisin peptide)를 첨가한 혼합물을 인큐베이팅(incubating)하는 단계(단계 1); 및 그래핀 옥사이드(graphene oxide; GO)를 상기 혼합물에 분산시키고 오토클레이빙(autoclaving)하는 단계(단계 2);를 포함하는 니신-금 나노입자-환원된 그래핀 옥사이드 나노복합체(nisin-gold nanoparticles-reduced graphene oxide; NAu-rGO)의 제조방법을 제공함에 특징이 있다.
본 발명에 따른 제조방법에 있어서, 상기 단계 1의 금 나노입자 전구체는 사염화금소수산(HAuCl4), 삼염화금산(AuCl3), 소듐사염화금산(NaAuCl4) 등 금 이온을 제공할 수 있는 것이면 제한 없이 사용할 수 있다. 바람직하게는, 사염화금소수산(HAuCl4), 삼염화금산(AuCl3), 및 소듐사염화금산(NaAuCl4)로 이루어지는 군으로부터 선택되는 1종인 것일 수 있다. 본 발명의 일실시예에서는, HAuCl4·H2O 수용액을 사용하였다.
본 발명에 따른 제조방법에 있어서, 상기 단계 1의 완충액은 인산 완충액(phosphate buffer), 아세테이트 완충액(acetate buffer), 시트레이트 완충액(citrate buffer), 인산완충식염수(Phosphate buffered saline) 등을 사용할 수 있고, 바람직하게는 인산 완충액을 사용하는 것일 수 있다.
본 발명에 따른 제조방법에 있어서, 상기 단계 1의 인큐베이팅은 15 내지 35℃의 온도에서 1 내지 3시간 동안 수행되는 것일 수 있다. 바람직하게는 상온에서 2시간 동안 수행되는 것일 수 있다.
본 발명에 따른 제조방법에 있어서, 상기 단계 2의 그래핀 옥사이드는 1시간 동안 소니케이션(sonication) 처리하여 분산시키는 것일 수 있다.
본 발명에 따른 제조방법에 있어서, 상기 단계 2의 오토클레이빙은 10 내지 20 lbs 압력하에 100 내지 150℃의 온도에서 1 내지 60분 동안 수행되는 것일 수 있다. 바람직하게는, 15 lbs 압력하에 121℃의 온도에서 15분 동안 수행되는 것일 수 있다.
본 발명에 따른 제조방법에 있어서, 상기 단계 1의 니신 펩타이드는 금 나노입자 전구체 및 그래핀 옥사이드를 동시에 환원시키는 환원제의 역할을 하는 것에 특징이 있다. 본 발명의 일실시예에 따르면, 상기 니신 펩타이드에 의해 GO 및 Au3+가 동시에 환원반응이 일어나 그래핀 옥사이드(GO)를 환원된 그래핀 옥사이드(rGO)로, 금 나노입자 전구체(HAuCl4·H2O)를 금 나노입자(AuNPs)로 환원시켜, 최종적으로 니신 및 AuNPs가 rGO에 장식된(decorated) 나노복합체(NAu-rGO)가 합성되었다.
또한, 본 발명은 본 발명의 제조방법에 따른 니신-금 나노입자-환원된 그래핀 옥사이드 나노복합체를 제공함에 특징이 있다.
본 발명에 따른 상기 나노복합체는 근적외선(near infrared; NIR) 영역의 빛을 흡수하여 발열하는 것일 수 있다. 본 발명의 나노복합체를 함유한 배지에 근적외선 레이저를 1 내지 10분간 조사하였을 때, 배지의 온도가 약 40 내지 70℃로 상승하였으며, 금나노입자 및 환원된 그래핀 옥사이드의 흡광도가 누적됨에 따라 광열 변환 효율이 매우 향상되는 효과가 있음을 확인하였다(실험예 3 참조).
또한, 본 발명은 본 발명에 따른 니신-금 나노입자-환원된 그래핀 옥사이드 나노복합체를 포함하는 비휘발성 메모리 장치(non-volatile memory device)를 제공함에 특징이 있다.
본 발명에 따른 상기 비휘발성 메모리 장치는 상기 나노복합체가 박막 트랜지스터(thin film transistor; TFT) 내의 유전체 층에 배치되는 것일 수 있다. 구체적으로, 본 발명에 따른 상기 나노복합체가 ZnO 반도체 층(semiconductor layer) 및 SiO2 유전체 층(dielectric lyer) 계면에 삽입된 형태의 박막 트랜지스터(TFT)를 포함하는 비휘발성 메모리 장치 일 수 있다. 본 발명의 일실시예에 따르면, 상기 박막 트랜지스터(TFT)는 Si 기판 위의 SiO2 유전체 층(dielectric lyer)에 상기 나노복합체를 코팅하고, ZnO 반도체 층(semiconductor layer)을 코팅하여 제조되는 것일 수 있다.
rGO는 우수한 전기적 및 광학적 특성으로 인해 광전자 용도로 광범위하게 사용되고 있다. 그래핀 및 rGO의 페르미 레벨(Fermi level) 및 일 함수는 화학적인 도핑을 통해 조절할 수 있으며, 고효율의 화학적으로 도핑된 태양 전지와 같은 소자 응용 분야에서 이용되고 있다. 따라서 rGO는 TFT 메모리 소자에서 전하 캐리어 트래핑 동작을 효과적으로 조작할 수 있는 멀티 밴드 구조를 제공할 수 있는 물질로 기대되고 있다.
본 발명의 일실시예에서는 NAuNP-rGO 나노복합체의 전기화학적 응용 가능성을 평가 위해 메모리 장치(memory device)용 박막 트랜지스터(Thin film transistor; TFT)에 적용하여 NAu-rGO 나노 복합체를 유전체층에 배치한 ZnO TFT를 제조하였고, 우수한 전하 전달 특성 효과를 나타냄을 확인하였다(실험예 2 참조).
또한, 본 발명은 본 발명에 따른 니신-금 나노입자-환원된 그래핀 옥사이드 나노복합체를 포함하는 광열(photothermal) 치료용 약학적 조성물을 제공함에 특징이 있다.
본 발명의 용어, 광열 치료(photothermal therapy; PTT)는 주로 근적외선 레이저를 조사하여 다양한 질환을 치료하는 것을 의미하며, 상기 PTT로 치료가능한 질환으로는 암 등이 있다. 이는 전자기파를 조사하여 광감음 물질을 여기상태로 전환시켰을 때, 이로부터 방출되는 진동 에너지 즉, 열에 의해 표적된 세포를 사멸시키는 것이다. 광열 치료는 낮은 에너지를 갖는 장파장의 빛을 이용하므로 다른 세포및 조직에 덜 유해하하며, 최소 침습적 치료가 가능하다.
예컨대, 상기 조성물을 종양부위로 빠르고 정확하게 전달하기 위하여 상기 광감응제 조성물은 종양 표적 물질을 추가로 포함할 수 있다. 예컨대, 상기 분지된 나노 구조물의 표면에 종양 세포에서 특이적으로 발현되는 단백질 등에 특이적으로 결합하는 리간드, 폴리펩티드 등을 결합시켜 사용할 수 있으나, 이에 제한되지 않는다.
본 발명의 나노복합체는 750nm 이상의 근적외선을 조사하여 광열효과를 나타낼 수 있으므로, 생체 내 주입하여 비침습적 광열 치료에 광감응 물질로 사용하기에 적합하다. 예컨대, 가시광선 등의 비교적 단파장의 빛을 이용하여 광열효과를 나타내는 입자는 체외에서 광원으로 조사시 체내의 환부로 빛의 투과가 불가능하여 내시경이나 광섬유를 이용하여 환부에 조사해야 하는 등의 번거로움이 있으나, 750 nm 이상의 근적외선을 이용하는 경우 광감응제를 환부에 주사하고 체외에서 광원을 조사하여 PTT를 수행할 수 있으므로 비침습적인 암치료가 가능한 장점이 있다.
본 발명의 일실시예에서는 니신 펩타이드를 이용하여 합성된 NAu-rGO 나노복합체의 생의학적 및 생화학적 적용 가능성을 입증하기 위하여, GO, NAuNPs 및 NAu-rGO 나노복합체와 함께 배양한 유방암 세포(MCF7 세포)에 800nm 다이오드 레이저(전력 = 0.5 W/cm2)로 NIR 방사선을 조사하여 여러 시간에 걸쳐 PTT 효과(in vitro)를 분석한 결과, rGO의 존재로 인하여 AuNPs의 광열효과가 향상되며, 효과적으로 암 세포를 사멸시킬 수 있음을 확인하였다(실험예 3 참조).
또한, 본 발명은 본 발명에 따른 니신-금 나노입자-환원된 그래핀 옥사이드 나노복합체를 포함하는 암 치료용 약학적 조성물을 제공함에 특징이 있다.

Hummers 및 Offeman(J. Am. Chem. Soc., 1958, 80 (6), pp 1339-1339)의 방법을 이용하여 천연 흑연(graphite)을 산화시켜 그래핀 옥사이드(graphene oxide; GO)를 제조하였다. 갈색의 GO 수용액(4mg/mL)을 하기 실험예에서 사용하였고, 균질한 그래핀 시트를 얻기 위해 각각의 실험에서 소니케이션(sonication) 처리를 수행하여 사용하였다.

니신-금 나노입자-환원된 그래핀 옥사이드(nisin-gold nanoparticles-reduced graphene oxide; NAu-rGO) 나노복합체(nanocomposite)는 열안정성 니신(nisin) 펩타이드(2.5% [w/w], 약 1×106IU/g)에 의한 환원반응을 이용하여 합성하였다.
단계 1: 인산 완충액(phosphate buffer, pH 6.8)에 0.5mM HAuCl4·H2O 수용액 및 0.5mg/mL의 니신 펩타이드를 첨가하여 25℃의 온도에서 2시간동안 인큐베이팅하였다.
단계 2: 제조예 1의 그래핀 옥사이드(GO, 0.8 mg/mL)를 1시간 동안 소니케이션 처리하여 상기 단계 1의 혼합물에 분산시킨 뒤, 15 lbs 압력하에 121℃에서 15분 동안 오토클레이빙하여 니신-금 나노입자-환원된 그래핀 옥사이드(NAu-rGO)를 제조하였다.
NAu-rGO의 합성은 UV-Vis 분광법에 의해 모니터되었다.

나노복합체를 포함하는 TFT 기반 비휘발성 메모리 장치의 제조
실시예 1에서 제조된 NAu-rGO 나노복합체의 비휘발성 메모리 소자용 전하 트래핑 매체로의 이용 가능성을 확인하기 위하여, NAu-rGO 나노복합체가 유전체층에 배치된 ZnO 박막 트랜지스터(thin film transistor; TFT) 기반의 비휘발성 메모리 소자를 제조하였다.
먼저, 아연 아세틸아세토네이트 하이드레이트(Sigma Aldrich)를 2-메톡시에탄올에 용해시켜 0.1M 용액을 제조한 뒤, 준비된 전구물질(HAuCl4·H2O 및 GO)을 2시간 동안 교반하여 완전히 용해 시켰다. 메모리 장치 제조를 위해, B-도핑된 p형 Si 웨이퍼(0.001 내지 0.003Ω-cm의 저항률)를 게이트 전극(gate electrode)으로 사용하였고, 300nm의 열성장된 SiO2를 유전체층(dielectric layer)으로 사용하였다. 스핀코팅 전에, SiO2/Si 기판을 아세톤 및 메탄올에서 각각 10분 동안 초음파(ultrasonically) 세척한 후, 5분 동안 탈이온수로 헹구었다. 실시예 1에서 제조된 NAu-rGO 나노복합체 용액을 SiO2/Si 기판 위에 2000rpm으로 20초 동안 스핀 코팅한 뒤, 반도체층(semiconductor layer)으로서 ZnO 용액을 4000rpm에서 40초 동안 스핀 코팅하여 25nm 이하의 두께를 갖는 필름 층을 제조하였다. 상기 필름을 200℃에서 10분간 핫플레이트상에서 건조시켜 용매를 증발시킨 다음, 400℃의 튜브로(tube furnace)에서 1시간 동안 어닐링하여 전구체를 분해 및 산화시켰다. 마지막으로 Al 전극을 쉐도우 마스크를 사용하여 활성층 위에 열적으로 성장시키고 채널 너비 및 길이를 각각 1000μm 및 100μm로 유지하도록 하였다. 최종 소스 및 드레인 전극은 약 100nm 두께였다.
NAu-rGO 나노복합체 층을 SiO2 및 ZnO 층 사이에 도입하여 제조된 NAu-rGO 나노복합체 기반 ZnO 박막 트랜지스터(thin film transistor; TFT) 메모리 장치의 구조는 도 6a에 나타난 바와 같다.

1-1. 나노복합체의 UV-
vis
스펙트럼 분석
그래핀 옥사이드(GO)의 환원 및 니신-금 나노입자-환원된 그래핀 옥사이드(NAu-rGO) 나노복합체의 형성은 UV-Vis 분광법을 이용하여 분석하였다. GO, 환원된 GO(rGO) 및 NAu-rGO 나노복합체의 UV-Vis 흡수 스펙트럼을 UV 1800 분광광도계(spectrophotometer, Shimadzu, Kyoto, Japan)로 측정하여, 도 2a에 나타내었다. GO는 C=C 결합의 π→π* 전이로 인해 230nm에서 최대 흡수를 보였고, C=O 결합의 n→π* 전이로 인하여 ~300nm의 흡광도에서 흡수를 보였다(도 2a; 청색, GO). 환원 반응이 감소한 후 ~270nm의 흡광도에서 레드 시프팅(red-shifting)이 관측되으며, 이는 rGO가 형성되었음을 의미한다(도 2a; 적색, rGO). 상기 실시예 1에서 GO 및 Au3+의 동시 환원을 통해 형성된 NAu-rGO 나노복합체는 UV-Vis 분광기를 통해 분석한 결과, rGO에 대한 ~270nm 및 NAuNPs에 대한 ~530nm에서의 흡수 피크가 확인되었으며, 이는 환원된 그래핀 옥사이드 시트의 표면에 NAuNPs가 형성되었음을 나타낸다(도 2a; 흑색, NAu-rGO).
1-2. 나노복합체의 구조 및 전기적 특성 분석
GO, rGO 및 NAu-rGO 나노복합체의 결정 구조 분석 및 GO와 rGO의 층간 거리를 결정하기 위하여 X-선 회절(X-ray diffraction; XRD) 분석을 실시하여 도 2b에 나타내었다. X-선 회절(X-ray diffraction; XRD) 분석은 Cu Kα 방사선(λ-0.15418nm)을 갖는 X-선 회절계(X-ray diffractometer, Bruker, Billerica, MA, USA)를 사용하여 수행하였다. GO의 10.30°에서의 2θ 회절 피크는 0.85nm의 상대적 층간 거리를 나타내었고, 이것은 산화된 흑연 층의 존재를 의미한다. 열안정성 펩타이드에 의한 환원 반응 후, rGO는 25.6°에서 넓은 회절 피크를 나타내어, GO가 성공적으로 환원되었음을 확인하였다. NAu-rGO 나노복합체의 XRD 패턴은 약 38.20°, 44.48°, 64.58° 및 77.60°에서 2θ 회절 피크가 관측되었고, 이는 각각 엘리멘탈 NAuNPs의 입방 구조의 (111), (200), (220) 및 (311) 결정 평면을 나타내며, 이를 통해 복합체에 NAuNPs가 형성되었음을 확인하였다(도 2b). XRD 패턴은 JCPDS 카드 No 004-0784와 비교하였다.
rGO 및 NAu-rGO 나노복합체의 구조적 및 전기적 특성을 분석하기 위하여 라만 분광법(Raman spectroscopy)을 실시하였다. 라만 스펙트럼은 레이저 532nm가 장착된 레니쇼 공초점 현미경 라만 시스템(Renishaw confocal microscope Raman system, Gloucestershire, UK)을 통해 확인하였다. 도 2c는 GO 분말(흑색) 및 NAu-rGO 나노복합체(적색)의 라만 스펙트럼이다. 그래핀에서 G 피크와 2D 피크는 sp2-혼성화된 탄소-탄소 결합을 나타낸다. G 피크는 ~1593cm-1 에서, D 피크는 ~1345cm-1에서 나타났으며, 이들 피크는 각각 탄소 원자의 A1g 모드와 E2g 모드에 해당한다. 전자 결합 상태의 변화는 D 피크 및 G 피크의 라만 세기의 비(ID/IG ratio)의 변화에 따라 평가하였다. 환원 전의 ID/IG 비는 GO의 경우 0.9, NAu-rGO 나노복합체의 경우 1.22로, 환원 반응의 마지막 단계에서 sp2 도메인의 크기가 감소함을 보여주었다. 그래핀 시트는 환원 과정에서 붕괴되어 다수의 sp2 도메인이 생성됨에 따라, 환원 과정이 진행된 것을 확인하였다. 또한, NAu-rGO 나노복합체에서 2D 피크(~2681cm-1) 및 S3 피크(~2931cm-1)의 증가된 강도는 개선된 흑연화를 나타내며, NAu-rGO 나노 복합체에 그래핀 시트가 거의 존재하지 않음을 확인하였다.
1-3. X-선 광전자 분광 분석
X-선 광전자 분광(X-ray photoelectron spectroscopy; XPS) 분석은 Al Kα를 조사원으로 사용하여 PHI 5000 Versa Probe(ULVAC PHI, 치가 사키, 일본) 분광계로 수행하였다.
도 3a는 GO 및 NAu-rGO 나노복합체의 XPS 측량 스펙트럼을 보여 주며 C1s 및 O1s의 특성 피크를 보여준다. 조사 스펙트럼으로부터 또한 NAu-rGO 나노복합체에서 Au4f 및 N1s 원소의 존재를 확인하였고, GO에 비해 NAu-rGO 나노복합체에서 O1s 피크가 현저히 약화되었고, C1s 관련 피크는 더 우세한 것으로 나타났다. Au4f 피크는 Au3 +가 Au°로 환원된 후 NAu-rGO 나노복합체의 XPS 스펙트럼에서 관찰되었다. 또한, NAu-rGO 나노복합체에서의 N1s의 존재는 rGO의 표면상의 펩타이드를 나타낸다. 도 3b는 83.6eV 및 86.9eV를 중심으로 하는 금속성 Au0 특유의 XPS 스펙트럼을 나타내며, 이를 통해 NAu-rGO 나노복합체의 표면상에 금 나노입자(AuNPs)가 형성되었음을 확인하였다.
GO 및 NAu-rGO 나노복합체에서의 탄소-산소 결합 배열의 기여는 C1s 영역의 고해상도 XPS 스펙트럼을 통해 분석하였고, 각각 도 3c 및 도 3d에 나타내었다. GO의 C1s로부터의 고해상도 스펙트럼에서 2개의 주성분으로 ~284.56eV에서 C=C/C-C 및 ~286.61eV에서 C-O 피크가 확인되었고, 그리고 하나의 미량성분으로 288.1eV에서 C=O 피크가 나타났다(도 3c). 열안정성 니신 펩타이드에 의한 환원 후, 산소와 결합되는 작용기(히드록실기 및 에폭시기)와 관련된 C-O 피크의 강도는 현저하게 감소하였고, C-C 결합은 고에너지 영역에서 단일 피크로서 지배적으로 남아 있었다 (도 3d). 이러한 결과로부터 열안정성 펩타이드의 구조가 환원 과정에서 GO의 구조를 크게 변화시켰음을 확인할 수 있었다.
1-4. 나노복합체의 형태 분석
전계-방출 주사전자현미경 이미지(Field-emission scanning electron microscope image)는 Carl Zeiss 200 FEG FE-SEM(Jena, Germany)를 통해 확인하였고, 투과전자현미경(Transmission electron microscopy; TEM) 및 고해상도 TEM (high-resolution TEM; HR-TEM) 분석은 JEM-3010(JEOL, Tokyo, Japan)을 사용하여 가속 전압 300kV에서 수행되었다.
도 4a 및 4b는 rGO 및 NAu-rGO 나노복합체의 FE-SEM 현미경 사진을 도시한다. rGO는 맑은 표면을 갖는 것으로 나타났으나(도 4a), NAu-rGO 나노복합체는 AuNPs의 존재로 인하여 거친 표면을 갖는 것으로 나타났다(도 4b). rGO의 표면에 존재하는 NAuNPs는 확대된 FE-SEM 현미경 사진으로부터 더 분명하게 확인할 수 있었다(도 4b; Inset).
도 4c 및 도 4d는 제조된 NAu-rGO 나노복합체의 HR-TEM 현미경 사진을 도시한다. AuNPs는 rGO의 표면에 분산되어 거의 응집되지 않았으나 다분산 형태로 존재하는 것으로 나타났다(그림 4d; Inset). 모든 AuNPs가 rGO의 표면에 존재하고 표면으로부터 돌출되지 않는 것으로 나타났으며, 이러한 결과로부터 니신 펩타이드를 통해 rGO 및 NAuNPs 사이에 매우 강한 상호 작용이 존재함을 확인할 수 있었다. AuNPs의 평균 직경은 25nm로 XRD 분석에 의해 얻어진 크기와 일치하였다.
에너지 분산형 X선 분석법(Energy-dispersive X-ray spectroscopy; EDS)을 통해 NAu-rGO 나노복합체를 분석한 결과, 엘리멘탈 금 금속의 특정 신호를 나타내는 것을 확인하여, rGO 표면에 형성된 나노 입자가 구체형의 NAuNPs임을 확인하였다(도 5).

2> 비휘발성 TFT 메모리 특성 평가
2-1.
ZnO
TFT 메모리 장치의 전하 전달 특성 분석
실시예 2에 기재된 방법으로 NAu-rGO 나노복합체 층을 SiO2 및 ZnO 층 사이에 도입하여 NAu-rGO 나노복합체 기반 ZnO 박막 트랜지스터(thin film transistor; TFT) 메모리 장치를 제조하였다(도 6a). 비휘발성 TFT 메모리 특성을 평가하기 위하여, 전류-전압(Current-voltage; I-V)은 프로브 스테이션(probe station)이 연결된 반도체 분석기(semiconductor analyzer, Keithley 4200, Tektronix, Beaverton, OR, USA)를 사용하여 측정하였다.
상기 비교예 3에서 제조된 ZnO 및 SiO2 계면에 rGO가 삽입된 ZnO TFT 메모리 소자의 전달 특성[드레인 전류(Drain current; IDS) vs. 게이트 전압(gate voltage; VGS)]을 분석한 결과, 이중 스윕(sweep) 모드에서 작은 히스테리시스 윈도우(hysteresis window)를 보였다(도 6b). 상기 실시예 2에서 제조된 ZnO 반도체 및 SiO2 유전체 층 사이에 NAu-rGO 나노복합체가 삽입된 ZnO TFT의 다양한 드레인 전압(drain voltage; VDS)에서의 전달 특성을 분석한 결과, 정방향(-40V~120V)에서 역방향(120V~-40V) 전압 방향으로 VGS를 스위핑(sweeping)함으로써 시계방향(clockwise)으로 더 넓은 히스테리시스의 IDS-VGS 곡선이 나타났다(도 6c). 또한, 10V에서 30V로 드레인 전압이 커질수록 상기 NAu-rGO 나노복합체를 포함한 ZnO TFT의 히스테리시스가 더 넓어짐에 따라(도 6c), ZnO 반도체 및 SiO2 유전체층 사이의 전하 트래핑 매체에 더 많은 전하 캐리어(charge carriers)가 트랩된다는 것을 알 수 있었다. 이러한 결과는 NAu-rGO 나노복합체를 포함한 ZnO TFT의 큰 히스테리시스가 순방향 및 역방향 전압 스위프 동안의 NAu-rGO 나노복합체의 전자(electron)/홀(hole) 트래핑에 기인한다는 것을 보여준다. 상기 비교예 4에서 제조된 rGO 또는 NAu-rGO 나노복합체를 포함하지 않는 ZnO TFT(도 6d)는 매우 낮은 히스테리시스 특성을 나타내었으며, 따라서, NAu-rGO 나노복합체를 포함하는 ZnO TFT가 ZnO 및 SiO2 유전체층 사이의 계면보다는 NAu-rGO 나노복합체의 정전기(electrostatic) 전하(charging) 및 방전(discharging)에 의해서 트래핑이 일어난다는 것을 확인할 수 있었다.
한편, 도 6c에서 확인된 NAu-rGO 나노복합체를 포함하는 ZnO TFT의 전달 특성은 기존의 다양한 AuNPs 및 rGO 기반 나노-플로팅(nano floating) 게이트 유전체에 대해 확인된 것보다 더 넓은 히스테리시스 윈도우를 보였다(J. Phys. D: Appl . Phys. 2008, 41;135111-135115 및 Biomacromolecules, 2015, 16;2766-2775).
게이트 전압을 0V에서 40V까지 변화시키면서 SiO2/NAu-rGO 나노복합체 유전체 스택 메모리 TFT를 사용한 ZnO 반도체 층의 방출 특성을 확인한 결과, 전형적인 n-형 트랜지스터 특성을 나타내는 NAu-rGO를 포함하는 TFT의 활성 채널의 핀치 오프(pinching-off)로 인해 게이트 전압이 증가함에 따라 드레인 전류(IDS)가 증가하였고 포화 상태에 도달하였다(도 6e).
NAO-rGO 나노복합체가 ZnO 및 SiO2 계면에 삽입된 ZnO TFT 비휘발성 메모리 소자의 전기적 파라미터를 확인하기 위하여, 상기 소자의 전달 특성을 표준 MOSFET 방정식에 적용하여 계산하였다. 포화이동도(saturated mobility)는 0.52cm2/V.s로 나타났으며, 순방향 및 역방향 임계 전압(threshold voltage)은 각각 14.52V 및 45.52V이었고, 온/오프(on/off) 전류비(current ratio)는 103 범위인 것으로 확인되었다.
2-2.
ZnO
TFT 메모리 장치의 스위칭 거동 및 전하 이동 분석
NAu-rGO 나노복합체 기반 ZnO 메모리 TFT의 스위칭 거동을 확인하기 위해, ±50V의 게이트 바이어스(gate bias)를 기록(writing) 및 소거(erasing) 공정에 적용하였다. 전달 특성을 나타낸 곡선에서 정전압(positive voltage) 바이어스 조건(기록 프로세스)하에 정전압 임계 전압 시프트(threshold voltage shift; ΔVth)가 관측되었으며, 기록 프로그래밍 시간의 증가에 따라 전달 곡선의 양방향 이동(positive shift)이 증가되었다(도 7a). +50V에서 0.1s 동안 기록 프로그래밍한 후, ΔVth는 0.1s의 프로그래밍 시간의 22.25V에서 0.01ms의 프로그래밍 시간의 61.30V로 증가했다(도 7a). 도 7b는 소거 프로세스의 전달 곡선을 도시한다. -50V에서 1초 동안 소거 프로그래밍한 후, 소스, 드레인 및 채널 영역이 접지됨에 따라 음의 소거 게이트 펄스가 1초의 소거시간 동안 인가되었다. -50V 게이트 바이어스(소거 프로세스) 적용시, 전달 곡선은 약 40-50V만큼 음방향으로 이동했다. 상기와 같은 NAu-rGO 나노복합체 기반 ZnO 메모리 TFT의 가역적 임계 전압 시프트 거동은 NAu-rGO 나노복합체의 캐리어 트래핑 및 디-트래핑에 의한 것으로 볼 수 있으며, 결과적으로 NAu-rGO 나노복합체가 메모리 소자 적용을 위한 전하 트랩 층으로 사용될 수 있음을 시사한다.
도 7c는 ZnO 반도체 및 SiO2 유전체층의 계면에 NAu-rGO 나노복합체가 내장된 ZnO-TFT 기반 비휘발성 메모리 소자의 전하 트랩 및 디-트랩 프로세스의 간단한 설명을 개략적인 밴드 다이어그램의 형태로 나타낸 것이다. 기억 프로그래밍 단계에서 정전압을 인가할 경우, 채널에 축적된 전자(electron)가 NAu-rGO 나노복합체로 전달되며, 이때 대부분의 전자가 NAu-rGO 나노복합체에 트랩되고, 트랩된 전자들은 얕은 준위(shallow level) 상태에서 깊은 준위(deep level) 상태로 이동된다(도 7d). 화학적으로 합성된 NAu-rGO 나노복합체에서의 전하 트래핑 에너지 준위는 구조적 결함 및 NAu-rGO, 채널 층 및 게이트 유전체의 일 함수에 기인할 수 있다. 화학적으로 합성된 NAu-rGO 나노복합체에서 많은 수의 구조적 결함이 형성되며 이는 관련 장치에서 전하 트래핑의 주요 원인이 된다. 또한, rGO와 NAu-rGO 나노복합체 사이에 존재하는 산소 기능기(oxygen groups)는 전하 캐리어를 효율적으로 포획(trap)한다. 따라서, 부가적인 트랩 준위(trapping level)가 NAu-rGO 나노복합체에 존재한다. 소거 프로그래밍 단계에서 얕은 준위에 머무르는 전자가 먼저 디-트랩되어 채널로 전달되며, 그 다음 깊은 준위 상태의 전자가 디-트랩된다(도 7e).

ATCC(Manassas, VA, USA)에서 인간 자궁경부암 세포(HeLa), 인간 섬유아세포 세포(L929) 및 인간 유방 선암종 세포(MCF7)를 구입하여 시그마(St. Louis, MO, USA)로부터 수득한 10% 소태아혈청 및 항생제(100g/mL 페니실린 및 100㎍/㎖ 스트렙토마이신)가 포함된 McCoy's 5A 배지에서 계대배양하였다. 25-cm2 조직 배양 플라스크에서 계대배양이 완료된 HeLa, L929 및 MCF7 세포를 농축하고 37℃의 CO2(5 %) 가습 분위기하의 배양기에서 유지시켰다. 90%의 세포포화상태(confluence)가 된 후, 인산염완충식염수(Dulbecco's phosphate-buffered saline, DPBS)를 사용하여 HeLa, L929 및 MCF7 세포를 세척하였다. 2mL의 트립신-EDTA 용액(0.25 %)을 사용하여 HeLa, L929 및 MCF7 세포를 분리한 다음, 최종적으로 10mL의 완전배지(complete medium)에 분산시켰다.
3-2. 세포 독성 분석
NAu-rGO 나노복합체의 생의학적 적용을 위해 자궁경부암세포(HeLa 세포주) 및 섬유아세포 세포(L929 세포주)를 NAu-rGO 나노복합체 존재하에서 24시간 및 48시간 동안 성장시켰고, MTT 분석법을 이용하여 상기 두 세포에 대한 세포 독성 분석을 수행하였다. 구체적으로, 세포(2×105 세포/mL)를 3-(4,5-디메틸티아졸-2-일)-2,5-디페닐테트라졸륨 브로마이드(MTT) 용액에서 배양하고, EZ-CYTOX-Enhanced Cell Viability Assay Kit를 사용하여 MTT의 미토콘드리아 산화에 대한 비색계를 관찰하였다. 세포를 농도를 달리한 상기 실시예 1의 NAu-rGO 나노복합체(10, 20, 40, 60, 80 및 100μg/mL)와 함께 DMEM(Dulbecco 's modified Eagle Medium)에서 배양하였다. 24시간 및 48시간 동안 배양한 후, NAu-rGO 나노복합체를 포함한 배지를 제거하고 새로 제조한 10μL의 MTT 용액을 첨가하였다. 세포를 4시간 더 배양한 뒤, 100μL의 디메틸설폭사이드를 처리하였다. 마지막으로, ELISA 플레이트 리더(BIORAD, USA)를 사용하여 각 웰(well)에 대한 405nm에서의 흡광도를 측정하였다. 세포의 생존율은 MTT 분석 방법을 통해 정량화하였다.
그 결과, 두 세포 모두에서 용량 의존적인 세포 독성이 관찰되었으나, 40μg/mL 농도까지 두 세포 모두 24시간 및 48시간 배양시 90% 이상의 생존률을 보이는 것으로 나타났다(도 8a 및 8b). 농도의 추가 증가는 두 세포주 모두에 대해 증가 된 독성을 나타냈다. 따라서, 10μg/mL 농도의 NAu-rGO 나노복합체를 사용하여 분석을 수행하였으며, 대조군과 동일한 농도의 GO 및 NAuNPs와 비교하였다.
3-3.
근적외선
영역에서의 흡광도 분석
광열 활성을 분석하기에 앞서, 상기 제조예 1, 비교예 1 및 실시예 1에서 제조된 GO, rGO 및 NAu-rGO 나노복합체에 대한 근적외선 영역에서의 흡광도를 조사하기 위해 UV-Vis-NIR 분광 분석을 수행하였다. 그 결과, NAu-rGO 나노복합체는 AuNPs의 특징적인 흡광도인 ~540nm에서 흡광도를 나타내지만 800nm에서의 흡광도는 나타내지 않았다(도 9).
이는 상기 TEM 현미경 사진(도 4c 및 4d)에서 확인된 바와 같이 rGO에 다양한 크기의 NAuNPs가 형성됨에 따라, 가시광선 및 근적외선 영역의 흡광도가 나타난 것임을 알 수 있었다. 이와 유사한 현상으로서, 실리카 나노입자가 층상 이중 수산화물(LDHs)로 장식된 금 나노구체가 NIR 영역에서 흡광도를 나타내지 않았지만 808nm에서 광열 활성을 나타내는 것을 Komarala et al과 Sharma 등에 의해 확인된 바 있다.

광열
치료 효과 분석(
in-vitro
)
광열 전환 효능(in-vitro)은 0.5W/cm2의 전력 밀도를 가진 800nm 적외선 다이오드 레이저(Changchun New Industries Optoelectronics Technology Co., Ltd., Changchun, China)와 온도계(UNI-T 1310, UNI-T Electronic Corp., Dongguan, China)를 사용하여 분석하였다. 10μg/mL의 GO(제조예 1), NAuNPs(비교예 2) 및 NAu-rGO 나노복합체(실시예 1)와 함께 24시간 동안 배양한 MCF7 유방암 세포를 1 내지 5분 동안 800nm 다이오드 레이저(전력 = 0.5W/cm2)에 노출시키고 24시간 동안 성장시켰다. 레이저 처리 2시간 및 24시간 후의 세포 생존력을 MTT 분석법을 이용하여 정량화하였다.
10μg/mL의 GO, NAuNPs 및 NAu-rGO 나노복합체를 각각 함유한 세포 배지에 근적외선 레이저(레이저 출력 밀도 0.5W/cm2)를 조사하여 GO, NAuNPs 및 NAu-rGO 나노 복합체의 광열 변환 효율을 측정한 결과, 레이저 조사 2분 이내에 NAu-rGO 나노복합체가 처리된 배지의 온도가 49℃로 급격히 상승 한 반면, 동일한 조사 조건하에서 GO 및 NAuNPs를 처리한 배지의 온도는 각각 25℃ 및 38℃로 관측되었다(도 10a). 따라서, GO는 레이저 조사시 매우 낮은 에너지 변환 효율을 갖는 것으로 나타났고, GO 시트에 NAuNPs를 장식함에 따라, 구형 NAuNPs가 GO의 광열 특성을 향상시키는 촉매로서 기능하였음을 확인하였다. 또한, NAuNPs의 광열 변환 효율 또한 NAu-rGO 나노복합체 형성에 의하여 향상된 것으로 나타났다. 이러한 NAu-rGO 나노복합체의 향상된 광열 활성은 상기 실험예 3-3 및 도 9에서 확인된 NIR 영역의 NAuNPs 및 rGO의 흡광도의 누적 효과에 의한 것으로 볼 수 있다. AuNPs는 나노복합체에서 근적외선 레이저 조사시 NAuNPs의 이동성에 관여하는 NAu-rGO 나노복합체의 rGO의 결함 및 빈자리에 부착하는 경향이 있으며, 결과적으로 구형 NAuNPs의 모양 변형이 발생하지 않게 된다. 또한, 근적외선 레이저 조사는 rGO의 탈산소화를 유발했으며, 이는 rGO의 열전도도 및 열전달 능력을 증가시켰고, 808nm에서 NAu-rGO 나노복합체의 광열 수렴 특성을 향상시켰다.
근적외선 조사 실험을 수행하여 유방암 세포(MCF7 세포주)에 대한 GO, NAuNPs 및 NAu-rGO 나노복합체의 광열 전환 효율을 MTT 분석법을 이용하여 분석하였다. 입자와 세포의 효과적인 상호작용이 나타날 수 있도록 MCF7 세포를 10μg/mL의 GO, NAuNPs 및 NAu-rGO 나노복합체와 함께 각각 배양한 다음 5분 동안 근적외선을 조사하였다. 근적외선 레이저 조사 후 2시간 동안 배양한 뒤, 세포생존율을 MTT 분석법으로 측정하였다. 10μg/mL의 NAu-rGO 나노복합체와 함께 배양한 MCF7 세포는 근적외선을 5분 동안 조사하고 2시간 배양한 후 50% 생존만을 보인 반면, GO와 함께 배양한 MCF7 세포는 동일한 반응 조건하에서 90% 이상의 생존율을 보였다(도 10b). 또한, 이들 세포를 24시간 동안 배양하고 세포의 생존율을 계산한 결과, NAu-rGO 나노복합체가 처리된 세포에서 24시간 배양 후 20% 미만의 생존율을 나타냈고, 이와는 대조적으로, 동일한 반응 조건하에서 GO 및 NAuNPs를 처리한 MCF7 세포는 각각 80% 및 45% 이상의 생존율이 관찰되었다(도 10c). 따라서, NAu-rGO 나노복합체는 GO 및 NAuNPs보다 개선되고 효과적인 광열 전환 효능을 나타내었으며, 생의학 분야에 적용될 수 있음을 확인하였다.
3-5.
암 세포
사멸 효과 분석(
in vitro
)
NAu-rGO 나노복합체의 암 치료 효과를 입증하기 위해, 무처리한 MCF7 세포 또는 GO, NAuNPs 및 NAu-rGO 나노복합체와 함께 배양된 MCF7 세포에 실험예 3-3의 800nm 다이오드 레이저를 5분간 조사한 후, 살아있는 세포와 죽은 세포에 각각 청색, 적색 및 녹색 형광을 생성시키는 형광 라벨 4',6-디아미디노-페닐린돌(4',6-diamidino-2-phenylindole; DAPI), 프로피디움 아이오딘화물(propidium iodide; PI) 및 플루오레세인 디아세테이트(fluorescein diacetate; FDA) 염료를 이용하여 정성 생체/사멸 세포 분석을 수행하였다(도 11). 세포는 공초점 레이저 스캐닝 현미경(CLSM)하에서 시각화하여 관찰되었다. PI 염료는 고분자의 염료로, 살아있는 세포는 손상되지 않은 세포막 때문에 PI 염료를 흡수할 수 없으나, 괴사성 또는 초기 세포 사멸 세포는 세포막이 손상되어 PI를 흡수할 수 있다. 따라서 살아있는 세포핵을 염색하기 위해 FDA 염료(녹색)를 사용했고, 죽은 세포는 PI(적색)를, 살아 있는 세포 및 죽은 세포의 핵은 DAPI(청색)를 사용하여 염색하였다.
GO, NAuNPs 및 NAu-rGO 나노복합체를 처리하지 않은 MCF7 세포는 24시간 배양 후에 청색 및 밝은 녹색 형광을 나타냈으며, 적색 형광은 나타나지 않아 입증된 원상태 그대로의 형태를 나타내었다(도 11a 내지 11d). GO를 처리한 MCF7 세포는 소수의 세포가 적색 형광을 나타내었고, 많은 수의 세포가 녹색 형광을 나타냄에 따라, GO의 광열 활성에 의한 영향은 적은 것으로 확인되었다(도 11e 내지 11h). 상기 MTT 분석 결과에서 NAuNPs가 MCF7 세포를 50% 이상을 사멸시킨 것과 마찬가지로(도 10c), NAuNPs를 처리한 MCF7 세포에서는 적색 형광 세포의 수가 증가하고 녹색 형광 세포 수가 감소하여 MCF7 세포에 대해 우수한 광열 활성을 보였다(도 11i 내지 도11l). 그러나 NAu-rGO 나노복합체를 처리한 MCF7 세포에 대한 CLSM 분석에서 우수한 광열 활성이 나타났으며(도 11m 내지 11p), 일부 녹생 형광이 나타나는 NAuNPs를 처리한 MCF7세포(도 11i 내지 도11l)에 비해 녹색 형광이 거의 나타나지 않았으므로, NAu-rGO 나노복합체가 NAuNPs보다 현저하게 우수한 광열 변환 효능을 갖는 것을 확인할 수 있었다(도 11m 내지 11p).
결과적으로, 상기 MTT 분석 및 CLSM 분석을 통해 상기 실시예 1에서 제조한 NAu-rGO 나노복합체가 근적외선 광열치료(PTT) 제제로서 높은 잠재력을 가지며 포유류 세포에 대한 세포 독성이 낮아 항균제 및 약물 전달체와 같은 다양한 생의학 및 약학적 용도로 사용될 수 있음을 확인하였다.

L-시스테인, 소혈청 알부민, 호르몬 등의 다양한 생물학적 환원 물질을 이용한 방법들은 알칼리 조건하에서 이루어지거나 매우 긴 반응 시간(최소 2.5시간)을 필요로 하는 반면, 상기 실시예 1의 방법은 중성 조건하에서 매우 짧은 시간(15분)에 환원이 가능하며, ID/IG 비율(ratio) 역시 가장 큰 것으로 나타났다. 따라서, 본 발명의 니신 펩타이드를 이용한 방법이 종래의 방법들보다 더 효율적이고 신속한 방법임을 확인하였다.
이제까지 본 발명에 대하여 그 바람직한 실시예들을 중심으로 살펴보았다. 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자는 본 발명이 본 발명의 본질적인 특성에서 벗어나지 않는 범위에서 변형된 형태로 구현될 수 있음을 이해할 수 있을 것이다. 그러므로 개시된 실시예들은 한정적인 관점이 아니라 설명적인 관점에서 고려되어야 한다. 본 발명의 범위는 전술한 설명이 아니라 특허 청구범위에 나타나 있으며, 그와 동등한 범위 내에 있는 모든 차이점은 본 발명에 포함된 것으로 해석되어야 할 것이다.
Claims (13)
- 완충액에 금 나노입자 전구체를 포함하는 수용액 및 니신 펩타이드(nisin peptide)를 첨가한 혼합물을 인큐베이팅하는 단계(단계 1); 및
그래핀 옥사이드를 상기 혼합물에 분산시키고 오토클레이빙하는 단계(단계 2);를 포함하고,
상기 니신 펩타이드는 금 나노입자 전구체 및 그래핀 옥사이드를 동시에 환원시키는 환원제의 역할을 하는 것인, 니신-금 나노입자-환원된 그래핀 옥사이드 나노복합체의 제조방법. - 제1항에 있어서,
상기 단계 1의 금 나노입자 전구체는 사염화금소수산(HAuCl4), 삼염화금산(AuCl3), 및 소듐사염화금산(NaAuCl4)로 이루어지는 군으로부터 선택되는 1종인 것인 제조방법. - 삭제
