(1) 세포배양(1) Cell culture
염증 활성 실험에 사용된 뮤린 대식세포주(murine macrophage cell line)인 RAW264.7은 한국세포주은행(Korean Cell Line Bank, KCLB)로부터 분양 받아 사용하였다. 1% 항생제(Gibco, USA)과 10% FBS(fetal bovine serum, Gibco, USA)이 함유된 DMEM 배지(Dulbecco’s modified Eagle’s minimal essential medium) 배지를 사용하여 37℃, 5% CO2 항온기에서 배양하였으며, 3일에 한 번씩 계대 배양 하였다. LPS(Lipopolysaccharide)는 시그마-알드리치(Sigma-Aldrich, St. Louis, MO, USA) 로부터 구입하여 사용하였다.RAW264.7, a murine macrophage cell line used in inflammatory activity experiments, was purchased from Korean Cell Line Bank (KCLB). (2) 일산화질소(Nitric oxide, NO) 측정(2) Nitric oxide (NO) measurement
24 웰 플레이트에 RAW264.7 세포를 웰당 1.5×105 cells로 분주하고 18시간 전 배양 후, 시료와 LPS 1 ㎍/ml를 동시 처리하여 24시간 배양하였다. 생성된 일산화질소의 양은 그리스 시약(griess reagent, Sigma)를 이용하여 세포배양액 중에 존재하는 NO2- 형태로 측정하였다. 세포배양 상등액 100 ㎕와 그리스 시약(griess reagent) 100 ㎕를 혼합하여 96 웰 플레이트에서 10분 동안 반응시킨 후 540 nm에서 흡광도를 측정하였다. 결과를 시료 무처리군 대비 백분율로 도 1에 나타내었다. 도 1에서 확인되듯이, 대체로 처리 농도에 비례하여 NO 생성을 억제하였다.RAW264.7 cells were plated at 1.5 × 10 5 cells / well in a 24-well plate and cultured for 18 hours. 24 웰 플레이트에 RAW264.7 세포를 웰당 1.5×105 cells로 분주하고 24시간 배양 후, LPS를 1 1 ㎍/ml로 처리하여 염증성 사이토카인의 생성을 자극하고 동시에 시료를 농도 별로 처리하였다. 24시간 배양 후 원심분리(12,000 rpm, 3 min)하여 얻어진 상층액의 염증성 사이토카인을 ELISA 키트(R&D systems, USA)를 이용하여 측정하였다. 결과를 시료 무처리군 대비 백분율로 도 2에 나타내었다. 도 2에서 확인되듯이, 순비기나무 열매 정유 추출물은 염증성 사이토카인의 생성 억제 활성을 나타내지 못하였으나 그 잎 정유 추출물은 대체로 처리 농도에 비례하여 높은 억제 활성을 보였다.RAW264.7 cells were plated at a rate of 1.5 × 10 5 cells / well in a 24-well plate, cultured for 24 hours, treated with 1 μg / ml of LPS to stimulate the production of inflammatory cytokines, and treated at the same time. 여드름균인 프로피오니박테리움 아크네스 CCARM 9009(Propionibacterium acnes CCARM 9009), 표피포도상구균인 스타필로코쿠스 에피더미스 CCARM 3709(Staphylococcus epidermidis CCARM 3709) 및 황색포도상구균인 스타틸로코쿠스 아우레우스 CCARM 0027(Staphylococcus aureus CCARM 0027)를 항생제 내성 균주은행(CCARM, Culture Collection of Antimicrobial Resistant Microbes)으로부터 분양받아 사용하였다. 균주를 배양하기 위한 배지로 LB 아가 배지(luriabertani agar medium), TSA 배지(tryptic soy agar medium) 및 GAM 배지(gifu anaerobic medium)을 디프코라사(Difco La; Sparks, MD. USA)에서 구입하여 사용하였다. 스타필로코쿠스 에피더미스 및스타틸로코쿠스 아우레우스의 경우 37℃ 호기 조건에서 배양 하였고 24시간 마다 계대 배양을 실시하였다. 프로피오니박테리움 아크네스는 37℃ 혐기성 조건에서 배양 하였으며 24~48시간 마다 계대 배양하여 실험에 사용 하였다.The LB agar medium, tryptic soy agar medium and gifu anaerobic medium were purchased from Difco La (Sparks, MD, USA) as a medium for culturing the strain. 최소억제농도(MIC, minimum inhibitory concentration)는 미생물의 성장을 막는 항생물질의 최소농도로, 미생물에 대한 MIC값이 낮으면 낮을수록 시료물질은 그 미생물에 대한 감수성이 높다고 할 수 있다. MIC 측정은 액체배지희석법(broth dilution method)을 변형하여 사용하였다. 96 웰 플레이트에 시료를 포함하는 배지를 100 ㎕ 넣어준 후, 균 현탁액의 농도를 2×105 CFU/mL가 되도록 조절하여 100 ㎕씩 넣어주었다. 스타필로코쿠스 에피더미스 및스타틸로코쿠스 아우레우스는 37℃에서 24시간 배양하였으며, 프로피오니박테리움 아크네스는 37℃, 혐기성 조건에서 48시간 배양하였다. 이후 균의 증식이 나타나지 않는 최소농도를 확인하여 결과를 아래의 표 1에 나타내었다. 아래 표 1에서 확인되듯이 순비기나무 잎 정유 추출물이 그 열매 정유 추출물에 비하여 세가지 균주에 모두에 대해 항균 활성이 높았으며, 특히 프로피오니박테리움 아크네스에 높은 항균 활성을 나타내었다. MIC값이 낮다고 해서 그 농도에서 균이 모두 사멸한 것은 아니며, 시료의 영향으로 균의 성장을 방해하여 균이 자라지 않아 죽은 것처럼 보일 수 있기 때문에 최소사멸농도(MBC, minimum bactericidal concentration)는 MIC값이 나타난 그 이상의 농도의 시료 배양액들을 하드배지(1.5% agar)에 처리하여 콜로니를 형성하는지 확인하였다.1.5% 아가를 포함하는 배지에 MIC를 진행한 시료 배양액을 스트리킹(streaking)하여 배양하였다.스타필로코쿠스 에피더미스 및스타틸로코쿠스 아우레우스는 37℃에서 24시간 배양하였으며, 프로피오니박테리움 아크네스는 37℃, 혐기성 조건에서 48시간 배양하여, 콜로니가 형성되지 않는 최소농도를 확인하였다. 결과를 상기 표 1에 함께 나타내었다. 상기 MIC 결과와 유사하게 순비기나무 잎 정유 추출물이 그 열매 정유 추출물에 비하여 세가지 균주에 모두에 대해 항균 활성이 높았으며, 특히 프로피오니박테리움 아크네스에 높은 항균 활성을 나타내었다.
