1. 시험목적1. Purpose of Testing

국보싸이언스(주)에서 의뢰한 시트랄 5% 제제의 털진드기 및 참진드기에 대한 살비효력을 검정하고자 한다.We would like to test the saliva efficacy against hairy ticks and mites of citral 5% preparation commissioned by National Science Cycle Co., Ltd.

2. 시험방법2. Test method

가. 시험동물end. Test animal

1) 활순털진드기(Leptotrombidium scutellare) 유충1) Leptotrombidium scutellare ) larva

나. 채집장소I. Collection place

1) 활순털진드기 유충: 2017년 11월 6일 - 14일에 전남 곡성군 옥과면 소룡길과 나주시 다시면 주변에서 등줄쥐를 포획하여 털진드기를 채집하였다.1) Bullwing mite larvae: On November 6th - 14th, 2017, weedworms were caught in the vicinity of Sowong-gil and Naju-si in Ogye-myeon, Geosung-gun, Chonnam Province.

2) 작은소피참진드기 자충: 2017년 11월 8일 - 11일에 제주특별자치도 서귀포시 동흥동 미악산(솔오름)과 회수동 인근에서 채집하였다.2) Small Sphinx mites: On November 8-11, 2017, the mosquitoes were collected from Myeongsan (Sol-Oum) in Donggu-dong, Seogwipo-si, Jeju Special Self-Governing Province, and near Hoesu-dong.

1) 활순털진드기: 전날 오후에 밭뚝 주변 풀숲에 미끼먹이가 들어있는 sherman trap을 설치하고, 다음날 포획된 등줄쥐(Apodemus agrarius)를 수거하여 기생하고 있는 털진드기를 채집하였다.1) Sweet white mite: On the previous afternoon, a sherman trap containing bait food was placed in the grass around the paddy field , and the parasitic mite was collected by collecting the trapped Apodemus agrarius the next day.

2) 작은소피참진드기: 초지 위에 천 끌기 방법으로 작은소피참진드기 자충을 채집하였다.2) Small Sophora japonica: A small sphagnum mite was collected by pulling on a grass.

라. 시험동물 확보la. Obtain test animals

1) 털진드기 유충 [도 2 참조]1) Hairy tick larva [see FIG. 2]

① 땅콩버터에 오트밀을 2:1로 섞어 등줄쥐의 유인미끼로 사용하였다.① Peanut butter and 2: 1 oatmeal mixed as a bait for the mankind was used.

② 셔먼 트랩에 유인미끼를 약 2g 정도 넣고 등줄쥐가 서식할만한 풀숲에 17:00 경에 설치하였다.② Approximately 2g of manned bait in the shaman trap was installed around 17:00 in a grassy area where the lizard was inhabited.

③ 다음날 08:00 - 09:00 경 포획된 트랩을 수거하여 털진드기 유충의 숙주인 등줄쥐를 확보하였다. 포획되지 않은 트랩은 장소를 이동하여 재설치하고 연속적으로 등줄쥐를 포획하였다.③ The next day, 08:00 - 09:00, trapped traps were picked up to secure the host of the mite larva. Untrapped traps were moved and repositioned to capture the backbone in succession.

2) 작은소피참진드기 자충2) Small sphyx mite tick

① 야생동물이 많이 서식하는 지역이나 가축을 방목하는 지역에 특히 작은소피참진드기가 많이 분포하므로 제주도를 채집지역으로 선택하였다.① Cheju Island has been selected as a gathering area because there are a lot of small sphagnum mites especially in wild animals and grazing areas.

② 작은소피참진드기는 풀끝에 올라와 지나가는 숙주동물로 이동하기 때문에 천끌기법으로 작은소피참진드기를 채집하였다.② Small Sphinx mites climb to the end of the pool and move to the passing host animal.

③ 가로와 세로의 길이가 1 m인 하얀 천의 한쪽 면에 같은 길이의 막대를 고정하고 고정된 막대 양쪽 끝을 줄로 연결하여 초지에서 쉽게 끌고 다닐 수 있게 제작하였다.③ A white cloth with a length of 1 m and a length of 1 m is fixed on one side of the same length rod, and both ends of the fixed rod are connected by a line so that they can be easily dragged from the grass.

길이 10 cm × 폭 5 cm의 크기로 자른 여과지에 시트랄 5% 제제를 0.25 mL(50 mL/m2 기준) 적하 처리하고, 실내에서 용매를 증발시킨 후 여과지를 지름 5 cm 크기로 오려냈다. 오려낸 여과지는 지름 5 cm 페트리디쉬 바닥면에 붙이고 네일아트용 붓으로 진드기를 10마리 넣어준 후 계속 확인하여 치사되는 개체를 관찰하고 치사율을 산출하였다.The filter paper was cut into a size of 5 cm in diameter after being dripped with 0.25 mL (based on 50 mL / m 2 ) of the citral 5% preparation on the filter paper cut into a size of 10 cm × 5 cm in width and the solvent was evaporated in the room. ① 털진드기 시험과 동일한 방법으로 실시하였으며, 작은소피참진드기는 육안으로 활동력을 확인하여 1회 시험에 10 마리를 넣어 주었고, 총 3회 실시하였다.(1) The same method as that of the hair mite test was carried out. Ten mites of the small sphagnum mite were visually examined and put into the test for a total of 3 times.

① 90 x 15 mm 페트리디쉬에 90 mm 필터페이퍼를 깔고 제작한 얇은 핀으로 집먼지진드기를 접종하였다.① House dust mite was inoculated with a thin pin made of 90 x 15 mm Petri dish and 90 mm filter paper.

② 대조군(Control, 메탄올), 시트랄 5.0% 2종류의 액체를 자사 100 mL 플라스틱 용기로 5회씩 필터페이퍼에 분사하였다.② Control (control, methanol) and citral 5.0% Two kinds of liquids were sprayed into the filter paper five times with their 100 mL plastic containers.

집먼지진드기에 시트랄 5.0%를 분사한 후 체색변화를 관찰한 결과가 도 4에 나타나 있다.FIG. 4 shows the results of observing changes in body color after spraying 5.0% citral into house dust mite.
