Abstract
천산룡의 기원 식물은 부채마지만, 근연종인 단풍마도 구별 없이 천상룡으로 유통되고 있는 실정으로, 도 4 및 5에 개시한 바와 같이, 부채마와 단풍마의 외형은 거의 유사하다. 특히, 천산룡은 절단하여 건조된 것이 유통되므로 이들을 유관으로 구별하는 것은 매우 어려운 일이다(도 6).
따라서 본 발명은 유통과정에서 육안으로 식별이 어려운 천산룡 약재인 부채마와 단풍마를 감별하는 방법으로, 사프라닌과 패스트 그린 염색 시약을 이용한 이중염색법 또는 고속액체크로마토그래피법으로 손쉽고 빠르게 구별하는 방법을 제시하였다.
Description
도 2는 부채마의 고속액체크로마토그래피 분석 결과이다. (A)는 표준품 디오스신(dioscin)이고, (B) 내지 (D)는 동일 조건으로 추출한 천산룡 유통품(부채마) 추출액(20㎕)의 분리 결과이다.

- 삭제
- (a) 부채마 또는 단풍마로 의심되는 시료의 내부조직을 사프라닌(safranine)과 패스트 그린(fast-green)의 염색약으로 이중염색하는 단계; 및
(b) 상기 염색된 조직으로부터 주피(periderm)가 사프라닌(safranine) 염색약에 의해 붉은색으로 염색되며, 코르크층(phellem)만으로 이루어진 것은 단풍마로 판별하고, 주피가 패스트 그린(fast-green) 염색약에 의해 푸른색으로 염색되고, 코르크층(phellem), 코르크형성층(phellogen) 및 코르크피층(phelloderm)으로 이루어진 것은 부채마로 판별하는 단계;를 포함하는 것을 특징으로 하는 부채마와 단풍마의 감별방법. - 삭제
- 제2항에 있어서, 상기 부채마 또는 단풍마로 의심되는 시료는 사프라닌(safranine)과 패스트 그린(fast-green)의 염색약으로 이중염색하기 전에
(a) 건조하여 절단한 후, 끓는 물에 8 내지 15분간 전처리하는 단계;
(b) 상기 단계 (a) 이후에, 10%, 20%, 35%, 50%, 75% 및 100%의 TBA(tert-Butyl alcohol)를 순차적으로 처리하여 조직을 탈수하는 단계;
(c) 상기 단계 (b)에서 탈수된 조직에 파라핀을 포매하는 단계;
(d) 상기 포매한 조직을 10~20㎛의 두께로 절단하고 알부민 용액을 이용하여 슬라이드에 접착하는 단계; 및
(e) 상기 슬라이드에 접착된 조직을 자일렌과 알코올에 순차적으로 침지하여 파라핀을 제거하는 단계;를 포함하여 제조한 것을 특징으로 하는 부채마와 단풍마의 감별 방법. - 삭제
