Abstract
Description
도 2는 트랜스펙션(transfection)된 옥수수 엽육 원형질체에서 OsWRKY67-GFP 융합 단백질의 세포 내 위치를 나타낸다. 엽록소 자가형광과 OsABF1-RFP는 엽록체 및 핵의 마커로서 각각 사용되었다. GFP(녹색) 및 RFP(적색) 형광으로부터 구별하기 위해, 위색(false color)(청색)이 엽록소 자가형광에 대해 사용되었다.
도 3은 OsWRKY67 활성화 돌연변이체 및 야생형 동진(Dongjin) 잎에 벼 도열병균(M. oryzae) PO6-6을 접종한 후의 저항성 반응 결과를 나타낸다. (A) 벼 도열병균 PO6-6로 접종된 야생형 동진, OsWRKY67-D1 및 OsWRKY67-D2 활성화 돌연변이 식물체의 질환 반응. 병증은 접종 후 10일에 보여준다. 내성 대조군 RIL260을 이용하였다. (B) 벼 도열병균 PO6-6로 접종된 야생형 동진, OsWRKY67-D1 및 OsWRKY67-D2 활성화 돌연변이 식물체의 잎 병변의 길이. 도 5는 OsWRKY67 활성화 돌연변이체 및 야생형 동진(Dongjin) 잎에 벼 흰잎마름병균 PX099를 접종한 후의 저항성 반응 결과를 나타낸다. (A) 벼 흰잎마름병균 PXO99로 접종된 야생형 동진, OsWRKY67-D1 및 OsWRKY67-D2 활성화 돌연변이 식물체의 질환 반응. 식물은 5주령에 접종했고 병증은 접종 후 10일에 보여준다. (B) 야생형 및 OsWRKY67 활성화 돌연변이체의 병변의 길이. 접종 후 0, 5, 10, 및 14일에 측정되었고, 세 잎의 평균값으로 나타내었다. 막대기는 SD값을 나타낸다. (C) 흰잎마름병균 성장곡선. 성장정도는 CFU/잎 콜로니 형성 단위를 log에 의한 정도로 나타내었고, 야생형 동진, OsWRKY67-D1 및 OsWRKY67-D2에서 유사한 성장 단계의 세 잎에 대해 측정하였다. 막대기는 SD값을 나타낸다.
도 6은 벼 도열병균 접종 후 OsWRKY67 활성화 돌연변이체 및 야생형 동진(Dongjin)에서 방어 관련 유전자들(OsWRKY67, PR1a, PR1b, PR4, PR10a 및 PR10b)의 발현 패턴을 RT-PCR 분석 결과를 통하여 나타낸다. 상기 유전자들의 발현은 야생형 동진, OsWRKY67-D1 및 OsWRKY67-D2 활성화 돌연변이에 벼 도열병균 PO6-6 처리 0, 12 및 24시간 후 돌연변이체 및 야생형 식물에서 분석되었다. OsUbq5은 내부 대조군으로 사용되었다.
도 7은 벼 도열병균 접종 후 OsWRKY67 활성화 돌연변이체 및 야생형 동진(Dongjin)에서 방어 관련 유전자들(EDS1, PAD4, ICS1, CHS1, PAL2, PAL3, PAL5, PAL6, PAL8 및 PAL9)의 발현 패턴을 RT-PCR 분석 결과를 통하여 나타낸다.
도 8은 OsWRKY67 활성화 돌연변이체 및 야생형 동진(Dongjin)에서 키틴(A) 또는 flg22(B) 처리 후 유도되는 ROS 반응 결과를 나타낸다. 오차 막대는 표준오차를 나타낸다(n=3).
도 9는 OsWRKY67 활성화 돌연변이체에서의 유전자 온톨로지(GO) 분석 결과를 나타낸다.
도 10은 생물학적 스트레스에서의 맵맨(Mapman) 분석을 나타낸다. 적색은 OsWRKY67 활성화 돌연변이체에서의 상향조절을 가리킨다.
도 11은 벼 도열병균 접종 후 OsWRKY67 활성화 돌연변이체 및 야생형 동진(Dongjin)에서 R 유전자들의 발현 패턴을 RT-PCR 분석 결과를 통하여 나타낸다. 6가지 R 유전자들의 발현은 야생형 동진, OsWRKY67-D1 및 OsWRKY67-D2 활성화 돌연변이에 벼 도열병균 PO6-6 처리 0, 12 및 24시간 후 돌연변이체 및 야생형 식물에서 분석되었다. OsUbq5은 내부 대조군으로 사용되었다.

Claims (13)
- 서열번호 2의 아미노산 서열로 이루어진 벼(Oryza sativa) 유래의 OsWRKY67 단백질을 코딩하는 유전자를 포함하는 재조합 벡터를 식물세포에 형질전환시켜 OsWRKY67 유전자를 과발현하는 단계를 포함하는 비형질전환체에 비해 식물체의 병 저항성을 증가시키는 방법.
- 삭제
- 제1항에 있어서, 상기 병 저항성은 도열병 또는 흰잎마름병에 대한 저항성인 것을 특징으로 하는 비형질전환체에 비해 식물체의 병 저항성을 증가시키는 방법.
- 서열번호 2의 아미노산 서열로 이루어진 벼 유래의 OsWRKY67 단백질을 코딩하는 유전자를 포함하는 재조합 벡터를 식물세포에 형질전환하는 단계; 및
