Abstract
본 발명은 적외선 반사-흡수 스펙트로스코피를 사용한 암세포의 탐지방법에 관한 것으로서, 보다 상세하게는 적외선 반사-흡수 스펙트로스코피를 사용한 세포 모니터링에 의해 정상 세포와 암세포의 진동 스펙트럼의 차이를 이용한 암세포의 탐지방법에 관한 것이다. 본 발명의 탐지방법을 이용하면 암세포를 비교적 정확하고 손쉽게 미리 탐지할 수 있다.

Description
본 발명은 적외선 스펙트로스코피를 사용한 암세포의 탐지방법에 관한 것으로서, 보다 상세하게는 적외선 반사-흡수 스펙트로스코피를 사용한 세포 모니터링에 의해 정상 세포와 암세포의 진동 스펙트럼의 차이를 이용한 암세포의 탐지방법에 관한 것이다.
세계보건기구(World Health Organization)에 따르면, 암은 전세계적으로 첫 번째 사망원인이다(Holms, J. (2009) Cancer. http://www.who.int/cancer/en/ accessed in 20 January 2009). 폐암은 남성에서 가장 흔하게 진단되는 암 형태의 하나이며, 남성에서 네 번째로 가장 흔한 암과 관련된 사망 원인이다(Ruddon, R.W. (2007) Cancer biology. Oxford University Press, 3-16). 침윤성 폐암의 초기 탐지가 사망률을 감소시키는데 매우 중요하다. 그러나, 구조적 및 생화학적 변화 없이 정상, 전암 증세 및 암성 세포간의 스펙트럼 징후에서 양적인 차이를 측정하는 것은 해결하기 어려웠다.
임상적 판단에서 가장 널리 사용되는 다양한 암을 진단방법중 하나는 병리학자에 의해 특이적 기관에 대한 염색에 의한 조직검사의 현미경적 측정일 수 있다(Dukor, R.K. (2002) Vibrational spectroscopy in the detection of cancer in Handbook of Vibrational Spectroscopy edited by Chalmers, J.M., and Griffiths, P.R., John Wiley & Sons, New York, 5, 3335-3361).
스펙트로스코피적 방법을 사용하여 초기 탐지율을 향상시키기 위한 노력이 있어 왔다(Hirsch, F.R. et. al. (2001) Clin Cancer Res. 7, 5-22). 많은 암중에서 정상 및 악성 조직을 구분하기 위하여, 형광(Torrance, C.J. et. al. (2001) Nature Biotech. 19, 940-945), 라만(Choi, J. et. al. (2005) Biopolymers 77, 264-272), 및 적외선 스펙트로스코피(Dukor, R.K. (2002) Vibrational spectroscopy in the detection of cancer in Handbook of Vibrational Spectroscopy edited by Chalmers, J.M., and Griffiths, P.R., John Wiley & Sons, New York, 5, 3335-3361)와 같은 광학적 스펙트로스코피가 사용되었다. 그러나, 암 분석을 위한 많은 물리적 방법은 아직까지 부정확하거나 매우 비용을 필요로 한다.
적외선 진동 스펙트로스코피는 다양한 생물학적 물질의 구조 및 조성에 관한 의미 있는 정보를 제공할 수 있다(Dukor, R.K. (2002) Vibrational spectroscopy in the detection of cancer in Handbook of Vibrational Spectroscopy edited by Chalmers, J.M., and Griffiths, P.R., John Wiley & Sons, New York, 5, 3335- 3361). FT-IR(Fourier-transform infrared spectroscopy)는 빠른 자동화된-데이터 포착 시간과 저렴한 비용을 갖고 생물의학 과학 분야의 다양한 현상에 적용되어 왔다. 빠른 측정 과정과 높은 입체 해상도 덕분에 FT-IR 기술의 진보는 염증성 및 전암 증세의 세포 상태를 탐지하는 것을 가능케 하였다. 이러한 기술이 지질, 단백질, 탄수화물, 및 인산화된 화합물의 화학 결합의 상태 및 상대적 농도의 연구를 가능케 한다.
적외선 스펙트로스코피를 사용한 암세포를 연구하는데 있어서 가장 큰 문제의 하나는 상피, 섬유성, 지방 조직, 암종성, 및 괴저성 세포 부위를 포함한 세포 성분이 복잡하고 비균질의 형태를 나타낸다는 점이다. 상대적으로 극소수의 진동 스펙트로스코피 도구를 사용한 암세포의 형태를 모니터링하는 보고가 있어 왔지만(Cohenford, M.A. and Rigas. B, (1998) PNAS, 95, 15327-15332 ; Gazi, E. et. al. (2007) Anal. Bioanal. Chem. 387, 1621-1631 ; Hammiche, A. et. al. (2005). Biophys. J. 88, 3699-3706 ; Krishna, C.M., et. al. (2006) Biopolymers 82, 462-470 ; Liu, C. et. al. (2006) J. Luminiscence 119-120, 132-136 ; Maziak, D.E. et. al. (2007) Cancer Detection and Prevention 31, 244-253 ; Mordechai, S. et. al. (2004) J. 이에, 본 발명자들은 스펙트로스코피를 수단으로 암세포를 미리 스크리닝하고자 노력한 결과, 적외선 반사-흡수 스펙트로스코피를 수단으로 폐암 상피 세포의 진동 스펙트럼을 분석하여 인산 및 포스포다이에스테르(phosphodiester) 스트레칭 밴드가 정상 및 초기 기관지ㆍ폐포 암종 세포간에 명확히 구분되는 것을 확인하여 정상 인간의 기관지 상피 세포와 폐암 세포의 구조적 및 세포내 변화를 스크리닝 할 수 있음을 확인함으로써 본 발명을 완성하였다.
본 발명은 스펙트로스코피를 사용하여 정상세포로부터 암세포를 미리 탐지하는 방법을 제공하는 것을 목적으로 한다.
상기 목적을 달성하기 위하여, 본 발명은 적외선 반사-흡수 스펙트로스코피를 사용한 세포 모니터링에 의해 정상 세포와 암세포의 진동 스펙트럼의 차이를 이용한 암세포의 탐지방법을 제공한다.
이하, 본 발명을 상세히 설명한다.
상기 암세포의 탐지방법에 있어서, 상기 적외선 스펙트로스코피는 적외선 반사-흡수: ATR-IR(attenuated total reflection infrared) 스펙트로스코피인 것이 바람직하고, 상기 암세포는 폐 상피 암세포인 것이 바람직하며, 이때 상기 폐 상피 암세포는 NCI-H460 세포(ATCC catalog number: HTB-177) 또는 NCI-H358 세포(ATCC catalog number: CRL-5807)인 것이 보다 바람직하다.
또한, 상기 암세포의 탐지방법에 있어서, 상기 정상 세포와 암세포의 진동 스펙트럼의 차이는 핵산 및 인지질에서 ~970 ㎝-1 에서의 인산 모노에스테르 대칭적 스트레칭 nus(PO3 2-) 밴드 강도 또는 ~1085 ㎝-1 에서의 포스포다이에스테르 대칭적 스트레칭 nus(PO2 -) 밴드 강도 차이인 것이 바람직하다.
ATR-IR(attenuated total reflection infrared) 스펙트로스코피를 수단으로 하여 유효 암세포의 분화를 통한 정상 및 폐 상피 암세포의 진동 스펙트럼 차이를 연구하였다. 정상 세포와 비교하였을 때, 핵산 및 인지질에서 ~970 ㎝-1 에서의 인산 모노에스테르 대칭적 스트레칭 nus(PO3 2-) 밴드 강도와 ~1085 ㎝-1 에서의 포스포다이에스테르 대칭적 스트레칭 nus(PO2 -) 밴드 강도가 다른 진동 밴드에 대하여 암세포에서 상당히 강화되는 것으로 나타났다. 이러한 발견은 암세포에서 보다 광범위한 인산화가 일어남을 제안한다. 이러한 결과는 적외선 스펙트로스코피 도구를 사용하여 폐암세포가 미리 탐지될 수 있음을 증명한다.
본 발명자들은 암세포로 폐 상피 암세포를 이용하였는데, 구체적으로는 NCI-H358 및 NCI-H460 세포를 사용하였고, 정상 세포로는 NHBE 세포를 사용하였다. 상기 정상 폐세포와 상피 암세포의 적외선 스펙트럼를 얻었다(도 1 참조). 정상 폐 세포에서는 ~1650 ㎝-1에서의 아미드 I 밴드 및 ~1550 ㎝-1에서의 아미드 Ⅱ 밴드가 강하게 관측되었다(표 1 참조). 폐 상피 암세포에서는 ~970, ~1085, 및 ~1235 ㎝-1에서의 nus(PO3 2-), nus(PO2 -), 및 nuas(PO2 -) 밴드가 강하게 나타난다(표 2 참조). 정상 세포와 비교할 때, 암세포에서는 ~1085 ㎝-1 에서의 핵산 또는 인지질의 대칭적 nus(PO2 -) 및 ~970 ㎝-1 에서의 인산 모노에스테르 대칭적 스트레칭 nus(PO3 2-) 밴드 강도가 강하게 나타났다. 정상 및 암세포에서 900-1290 ㎝-1에는 꽤 많은 스펙트럼 특징적 굴곡이 있다(도 3 참조).
상기에서 살펴본 바와 같이, 본 발명의 탐지방법을 이용하면 암세포를 비교적 정확하고 손쉽게 미리 탐지할 수 있다.
이하, 본 발명을 하기 실시예에 의거하여 보다 상세하게 설명하고자 한다. 단, 하기 실시예는 본 발명을 예시하기 위한 것일 뿐, 본 발명은 하기 실시예에 의해 한정되는 것이 아니고, 본 발명의 기술적 사상을 벗어나지 않는 범위 내에서 치환 및 균등한 타 실시예로 변경할 수 있음은 본 발명이 속하는 기술분야에서 통상의 지식을 가진 자에게 있어서 명백할 것이다.
<실시예 1> 세포 배양 및 적외선 스펙트로스코피를 위한 시료 제조
NCI-H358 및 NCI-H460 세포를 RPMI 1640(Gibco, Grand Island, NY)에서 배양하였다. NHBE 세포는 Cambrex(Walkerville, MD)로부터 구입하였고, 기관지 상피 성장 배지에서 유지시켰다. 모든 세포는 10% 우태아혈청 및 1% 페니실린-스트렙토마이신 항체(Gibco)가 보충되었고, 37℃에서 5% CO2/95% 습도의 대기하에 유지시켰다.
인산 완충 식염수로 대략 5 x 103 세포를 세척하고 상기 세포를 서로 분리하기 위하여 10분 동안 0.25% 트립신-EDTA를 첨가하였다. 10분 동안 1,200 g에서 원심분리한 후에, 상청액을 버렸다. 상기 식염성 시료를 몇분 동안 3,000 rpm에서 원심분리하고 세포 펠릿(pellet)을 마이크로센트로퓨지(microcentrifuge)에 옮기고, 10 분 동안 5,000 rpm에서 원심분리하여 FT-IR 분석을 위한 작은 세포 펠릿을 얻었다(Sindhuphak, R. et. al. (2003) Gynecologic Oncology 90, 10-14).
<실시예 2> 정상 및 폐암 세포의 특성분석
NCI-H460 세포(ATCC catalog number: HTB-177)는 대략 10~20% 폐암을 구성하는 폐의 커다란 세포암을 갖는 환자로부터 기원된다. NCI-H358 세포(ATCC catalog number: CRL-5807)는 카프카스인(Caucasian)의 폐의 초기 기관지ㆍ폐포 암종으로부터 기원되었다. 초기 NHBE(normal human bronchial epithelial; 정상 인간의 기관지 상피) 세포를 정상 폐 세포로 사용하였다. 정상세포는 프로그램된 세포 사멸(apoptosis)이 일어나지만, 암세포는 이러한 세포사멸 없이 계속적으로 증식되고, 이는 인산화 유무의 차이에 의해 일어나는 것으로 여겨지고 있다.
<실시예 3> 적외선 측정
액체 N2 냉각된 HgCdTe 탐지기가 장착된 0.09 ㎝-1 의 최대 해상력을 갖는 FT-IR 스펙트로미터(Thermo Nicolet 6700)를 사용하여 적외선 스펙트럼을 얻었다(Lim, J.K. et. al. (2007) Chem. Lett. 36, 1226-1228). 4 ㎝-1의 해상력을 갖고 1000~3500 ㎝-1의 범위에서 모두 128번의 스캔을 측정하였다. 준비된 폐암 세포 펠릿을 Pike Technology Miracle external reflection accessory의 ZnSe crystal위로 옮겼다. OMNIC v5.1a 소프트웨어를 사용하여 데이터 프로세싱을 수행하였다. SeaSolve software PeakFit version 4.12를 사용하여 스펙트럼 파라미터 및 각 스 펙트럼의 조정을 얻었다.
<3-1> 정상 폐세포의 적외선 스펙트럼
도 1(a)는 정상 폐세포의 중간-적외선 스펙트럼을 나타낸다. 몇 가지 진동적 특징중에서 주로 펩티드 주사슬의 C=O 스트레칭 진동 밴드에 기인된 ~1650 ㎝-1에서의 아미드 I 밴드가 가장 특징적으로 나타난다. CN 스트레칭과 N-H 그룹의 평면(in-plane) 벤딩의 커플링에 주로 기인된 ~1550 ㎝-1에서의 아미드 Ⅱ 밴드, 또한 적외선 스펙트럼에서 상당히 강하게 관측되었다. ~1460 ㎝-1 및 ~1400 ㎝-1에서의 2가지 진동 밴드는 CH3의 평면 벤딩 진동으로 설명될 수 있다. 한편, ~1235 ㎝-1 및 ~1085 ㎝-1에서의 진동 밴드는 PO2 - 그룹의 스트레칭 모드에 속한다고 판단된다. 970 ㎝-1 에서의 약한 진동 밴드는 PO3 - 그룹으로 설명될 수 있다. 본 발명자들에 의한 이러한 할당은 주로 이전의 보고에 기초한 것이고(Meurens, M. et. al. (1996) Vib. Spectrosc. 10, 341-346 ; Zenobi, M.C. et. al. (2008) Spectrochim. Acta Part A. 70, 270-276), 표 1에 요약하여 기재하였다.

<3-2> 폐암 세포의 적외선 스펙트럼
폐암 세포의 적외선 스펙트럼은 도 1(b) 및 (c)에 제시된 것과 같이 기록된다. 대부분의 경우에 스펙트럼은 유사하게 보이고, 이전의 연구로부터 통계적 분석이 필요로 된다. 각각 ~970, ~1085, 및 ~1235 ㎝-1에서의 nus(PO3 2-), nus(PO2 -), 및 nuas(PO2 -) 밴드가 본 발명이 제시한 다른 진동 모드와 비교하여 2 종류의 암세포에서 크게 강화된 것으로 나타난 것을 주목할 필요가 있다. 이들 밴드는 폐의 초기 기관지ㆍ폐포 암종으로부터 유발된 NCI-H358 세포에서 매우 증진된 것으로 확인되었다. 이것은 DNA 또는 부피 밀도에서 구조적 변화에 기인될 수 있다. 앞서의 FT-IR 연구에서, 대부분의 스펙트럼은 매우 유사하게 보였고 통계적 분석을 필요로 하였다. 본 발명에서는 몇 가지 진동 밴드의 직접적 관찰에 의해서 스펙트럼 차이가 명확히 관찰될 수 있음은 주목할 만하다. 아미드 I 밴드에 대한 상대적인 강도를 표 2에 요약하여 기재하였다. 정상 세포와 비교할 때, ~1085 ㎝-1 에서의 핵산 또는 인질질의 대칭적 nus(PO2 -) 및 ~970 ㎝-1 에서의 인산 모노에스테르 대칭적 스트레칭 nus(PO3 2-) 밴드 강도는 암세포에서 강화되는 것으로 나타났다. 흡수 (nus(PO3 2-)/흡수 (아미드 I) 또는 흡수 (nus(PO2 2-)/흡수 (아미드 I)의 상대적 강도 비율이 정상 세포로부터 폐암 세포를 구분하는데 사용될 수 있다. 도 2는 몇 가지 진동 밴드 강도의 3가지 폐암 세포에 대한 히스토그램을 나타낸다.

정상 및 폐암 세포에 대한 900-1290 ㎝-1에서의 파장수에서의 적외선 스펙트럼의 확장된 시각을 도 3에 나타낸다. 도 3에 제시한 것과 같이, 정상 및 암세포에서 900-1290 ㎝-1에는 꽤 많은 스펙트럼 특징 커브 조정이 있었다. ~970 및 ~1085 ㎝-1에서의 밴드, nus(PO3 2-) 및 nus(PO2 -)에 추가하여, 또한, 916, 1029, 1056, 1118, 및 1155 ㎝-1에서 밴드가 관찰되었다. 이들 밴드들은 탄수화물 또는 당연결체(glycoconjugate)와 연관될 수 있다. 추가하여, ~1235 ㎝-1에서 nuas(PO2 -) 밴드는 1221 및 1244 ㎝-1의 2 밴드로 나누어질 수 있다. 이들 2 밴드중 하나는 CH2 모드의 비틀림(torsional) 모드로 설명될 수 있다. 이들 밴드들은 또한 도 3에 제시한 바와 같이, 정상 또는 암세포와는 약간 다른 것이 확인되었다. 1030 및 1155 ㎝-1에서의 밴드가 정상 세포에서는 상대적으로 보다 증진된 것을 주목하여야 한다. 이전의 보고에 따르면, 발암(carcinogenesis) 동안에 글리코겐 수준이 감소이 감소되므로 이들의 강도가 감소되는 결과를 가져온다고 여겨진다(Wong, P.T.T, et. al. (1991) PNAS 88, 10988-10992). 그러나, 명확히 ~970 및 ~1085 ㎝-1에서의 밴드, nus(PO3 2-) 및 nus(PO2 -)가 정상 및 암세포에서 뚜렷한 차이를 잘 제공하는 것이 당연하게 보인다.
본 발명자들은 적외선 반사분광법(ATR-IR ; attenuated total reflection infrared) 스펙트스코피를 수단으로 정상 및 유방암 세포의 진동 스펙트럼을 조사하였다. 상기 결과는 적외선 스펙트로스코피 도구를 사용하여 폐암 세포가 미리 스크린될 수 있음을 증명하므로, 현재 본 발명자들은 폐암세포의 다른 스펙트럼 특징을 설명하려고 시도하고 있다. 단백질 인산화는 많은 세포 조절 과정에 관여된다(Hunter, T., and Karin, M. (1992) Cell 70, 375-387). 예를 들면, 종양 억제자인 p53의 인산화가 p53 활성을 조절하는데 중요하다(Ko, L.J., and Prives C. (1996) Genes and Development. 10, 1054-1072). 종양 세포에서, p53의 인산화 변형을 포함한 조절 완화(deregulation)가 종양세포에서 빈번히 관찰되고 종양발생과 연관된다고 알려져 있다(Zhang, W. et. al. (1994) Cancer Research 54, 4448-4453 ; Bode, A.M, and Dong, Z. (2004) Nat. Rev. Cancer 4, 793-805). 도 4는 DNA 또는 지질내의 포스포다이에스테르(PO2 -) 및 인산(PO3 2-) 작용 그룹을 설명하고 있다.
한편, 본 발명의 구체적 범위는 상기 기술한 실시예 보다는 특허청구범위에 의하여 한정지어지며, 특허청구 범위의 의미와 범위 및 그 등가적 개념으로 도출되는 모든 변경 및 변형된 형태를 본 발명의 범위로 포함하여 해석하여야 한다.
도 1은 정상 및 폐암 세포의 적외선 스펙트럼이고(이때, (a)는 정상 세포, (b)는 NHBE 및 암세포 및 (c)는 NCI-H460 세포이다. 각 시료는 스펙트럼의 반복성을 확인하기 위하여 두 번씩 얻어졌다. ~1650 ㎝-1에서 아미드 I 밴드의 강도에 대하여 상기 강도를 표준화시켰다),

Claims (5)
- 적외선 반사-흡수 스펙트로스코피를 사용하여 각각 대조군과 실험군으로 선정한 세포 내의 핵산 또는 인지질에서, 인산 모노에스테르 대칭적 스트레칭 nus(PO3 2-) 밴드 강도 또는 포스포다이에스테르 대칭적 스트레칭 nus(PO2 -) 밴드 강도를 측정하고, 대조군과 실험군의 상기 강도 데이타들을 서로 대비함으로써 대조군으로 선정한 세포와 실험군으로 선정한 세포내의 인산화 정도를 비교하는 방법.
- 제 1항에 있어서, 상기 적외선 반사-흡수 스펙트로스코피는 ATR-IR(attenuated total reflection infrared) 스펙트로스코피인 것을 특징으로 하는 방법.
- 제 1항에 있어서, 인산 모노에스테르 대칭적 스트레칭 nus(PO3 2-) 밴드 강도는 970 ㎝-1 에서 측정하는 것을 특징으로 하는 방법.
