Claims (7)
- 시료 상에 제각기 다양하게 형성된 서로 다른 세균들 중 특정세균의 집합체인 특정세균 집단이 따로 분리되는 제 1 단계;상기 특정세균 집단에 속한 특정세균에 대항하는 항체가 포함된 형광나노입자 결합물이 상기 특정세균 집단에 투입됨에 따라, 상기 특정세균이 상기 형광나노입자 결합물과 부착되고, 상기 형광나노입자 결합물을 부착한 특정세균이 표지(標識)되는 제 2 단계;상기 특정세균과 미부착된 형광나노입자 결합물이 제거되는 제 3 단계; 및상기 특정세균에 부착된 상기 형광나노입자 결합물로부터 인지되는 형광의 세기가 기설치된 세균양 측정 장치에 의해 측정됨으로 말미암아 상기 특정세균의 양(量)도 상기 세균양 측정 장치를 통해 예측되는 제 4 단계를 포함하며,상기 형광나노입자 결합물은 10 내지 80 nmol을 갖는 형광나노입자(QD)의 표면에 생체분자인 아비딘층을 형성시킨 후 상기 아비딘층에 바이오틴이 결합된 항체를 반응시켜 형성되며,상기 제 2 단계 중 상기 특정세균과 상기 형광나노입자 결합물 간 부착 과정은 상기 특정세균이 상기 바이오틴이 결합된 항체와 우선 반응하고 상기 아비딘층이 상기 바이오틴과 반응함에 의해 실시되는 것을 특징으로 하는 세균양 측정 방법.
- 제 1 항에 있어서, 상기 제 1 단계는,상기 특정세균 집단이 상기 특정세균의 항체가 고정된 2 내지 3㎛의 자석입자에 의해 분리 형성되는 단계를 포함하는 것을 특징으로 하는 세균양 측정 방법.
- 제 1 항에 있어서, 상기 제 1 단계는,상기 특정세균의 항체가 형성된 자석입자가 상기 시료에 투입되는 단계;항원항체 반응에 의해 상기 자석입자가 상기 시료 속의 특정세균에 부착되는 단계; 및상기 자석입자가 강자성체에 의해 끌려옴에 따라 상기 특정세균이 상기 특정세균 집단에 속하게 되는 단계를 포함하는 것을 특징으로 하는 세균양 측정 방법.
- 제 1 항에 있어서,상기 제 2 단계 중 상기 형광나노입자 결합물이 상기 특정세균 집단에 투입되는 과정에 이용되는 상기 형광나노입자 결합물은,자외선이 조사되면 형광을 발생시키는 상기 형광나노입자 표면에 상기 특정세균의 항체를 고정화시켜 생성되는 것을 특징으로 하는 세균양 측정 방법.
- 삭제
- 제 1 항에 있어서, 제 3 단계는,상기 형광나노입자 결합물이 투입된 특정세균 집단을 표준인산용액으로 세척한 후, 강자성체를 이용한 다른 한차례 분리 과정을 통해 상기 특정세균에 미부착된 형광나노입자 결합물을 제거하는 단계를 더 포함하는 것을 특징으로 하는 세균양 측정 방법.
- 제 1 항에 있어서, 제 4 단계는,상기 세균양 측정 장치가 기구비된 LED(Light Emitting Diode)램프를 이용해 450nm 파장을 갖는 자외선을 생성시켜 상기 세균 집단에 조사하는 단계; 및상기 세균양 측정 장치가 기구비된 실리콘 PMT(Photo Multiplier Tube)를 이용하여 상기 형광나노입자 결합물의 형광나노입자부터 발생되는 형광의 세기를 측정하는 단계를 포함하는 세균양 측정 방법.
