Abstract
본 발명은 마조람(Origanum majorana)으로부터 분리한 천연 항산화제 및 이의 정제방법에 관한 것으로서, 구체적으로는 마조람의 메탄올 추출물로부터 에틸아세테이트로 분획하고, 흡착 크로마토그래피, 크기배제 컬럼 크로마토그래피를 수행하여 항산화 활성, 특히 수퍼옥사이드 라디칼 소거활성을 갖는 물질을 분리하여 제공함으로써, 본 발명의 황산화제 및 정제방법은 기능성식품 또는 보조요법제의 소재로 이용되어 생체내에서 단백질, 핵산, 효소 등의 산화적 손상을 예방 또는 억제할 수 있다.

본 발명은 마조람으로부터 분리한 천연 항산화제 및 이의 정제방법에 관한 것이다.
보다 상세하게는, 마조람의 메탄올 추출물로부터 에틸아세테이트로 분획하고, 흡착 크로마토그래피, 크기배제 컬럼 크로마토그래피를 수행하여 항산화 활성, 특히 수퍼옥사이드 라디칼 소거활성을 갖는 물질을 분리하여 제공한다.
활성산소는 크게 ① 체세포에 의한 생성 : 형질막, 소포체, 핵막 등의 생체막, 미토콘드리아, 세포질, 호중구, 호산구 등(John, G., Scandalios., CSHL Press, New York (1992)), ② 약물 또는 화학물질에 의한 세포내 생성 : 퀴논계 항암제, 니트로퓨란, 알록산(alloxan), 철이온 등(Asada, K. and Takahashi, M., Elsevier, Amsterdam, 227∼235 (1987)), ③ 대기오염물질에 의한 세포내 생성 : NO2, HO·, 담배연기 등과 ④ 광, 방사선 등의 조사(Ledwith, A., Academic Press, N. Y. pp.21∼28 (1977)) 등에 의해 생체내에서 발생된다. 인체내에서 활성산소들은 전자수용체 및 공여체로 이용되거나 GSH 퍼옥시다제, 카탈라제, GSH 리덕타제, 글루타치온-s-트랜스퍼라제, SOD(superoxide dismutase) 등의 소거관련 효소들(Beckman, J. S., et al, Proc Natl Acad Sci, USA, 87: 1620-1624 (1990)), 트랜스페린, 락토페린, 페리니틴 등의 철 이온 킬레이트 물질(Suematsu, M., Gastroenterology, 103, 994-1001 (1992)), 구리 이온 킬레이트 물질인 셀로플라스민(celloplasmin)(Suzuki, H., Pancreas, 8, 465-470 (1993)) 및 일중항산소 소거물질인 β-카로틴(Yamamoto, Y., et al, Arch. Biochem. Biophys., 305, 541-545 (1993)) 등에 의해 소거된다.
그러나 활성산소의 소거와 생성억제 기구가 충분하게 기능을 유지하지 못할 경우 잔여 또는 과잉의 활성산소가 표적 분자를 산화시키게 되며, 표적 분자가 저장 단백질과 같이 큰 장해를 받지 않는 물질인 경우에는 세포 장해현상이 나타나지않지만 DNA의 경우 이의 산화는 변이, 암 등으로 진전되고 효소단백질, 생체막의 지질성분이 산화되면 세포반응, 세포의 항상성(homeostasis) 등이 깨어짐으로써 노인성 치매, 암, 염증, 노화, 동맥경화, 심장질환, 백내장 등의 각종 질환의 발병원인으로 작용하게 된다.
라디칼 소거제(radical scavenger)는 시험관내(in vitro)와 생체내(in vivo)에서 산화반응을 강하게 저해하는 것으로 밝혀졌기 때문에 케모프리벤터(chemopreventers)의 후보로서 가장 유력하다. α-토코페롤, 아스코르브산, β-카로틴 등과 간단한 페놀류(phenolics) 같은 체내 라디칼 소거제(dietary radical scavenger)는 최근까지 가장 주목을 받아왔으며(Huang, M. T. and Ferraro, T., Antioxidants and Cancer Prevention, Vol. 2. (1992)), 항산화활성 이외 돌연변이발생 저해활성(antimutagenic), 항암 촉진활성(anti-tumor promoting), 암발생 저해활성(anti-carcinogenesis) 등을 가지는 것으로 보고되고 있다.
높은 항산화 효과와 경제성 때문에 가장 널리 이용되고 있는 합성 항산화제인 부틸레이티드 하이드록시아니솔(butylated hydroxyanisole, 이하 'BHA'로 약칭함), 부틸레이티드 하이드록시톨루엔(butylated hydroxytoluene, 이하 'BHT'로 약칭함), 그리고 터셔리 부틸하이드로퀴논(tertiary butylhydroquinone, 이하 'TBHQ'로 약칭함) 등은 합성 페놀 화합물로 안전성에 논란이 되고, 소비자들의 거부감이 날로 심해지고 있다(신동화, 식품과학과 산업, 30, 1, 1997).
따라서 근래에는 인간이 안전하게 오랫동안 먹어 왔던 식물로부터 항산화 효과가 높은 물질을 분리, 이용하려는 시도가 활발히 이루어지고 있다(김종평, 생물산업, 11, 4, 6-14, 1998). 그 중에서도 여러 향신료들을 첨가하면 식품의 보존성이 증가한다는 사실로부터 향신료의 항산화 성분에 관한 많은 연구가 이루어져 왔다. 최근 향신료로부터 보고된 항산화 활성물질로는 세이지(sage)의 로즈메리-9-에틸에테르(Djarmati, Z., Jankov, R. M., Schwirtlish, E. Djulinac, B. and Djorkjevic, A., J. Am. Oil Chem. Soc., 68,731,1991), 로즈메리(rosemary)의 카노솔(carnosol)(Brieskorn, C. H. Fuch, A., Bredenberg, J. B., Mc Chesney, J. D. and Wenkert, E., J. Org. Chem., 29, 2298, 1964), 로즈메리퀴논(rosemariquinone), 로즈메리디페놀(rosemaridiphenol)(Wu, J. W, Lee, M. H., Ho, C. T. and Chang, S.S., J. Am. Pil Chem. 일반적으로 마조람(Origanum majorana L.)은 소염과에 속하는 다년초로 인도, 아라비아, 이집트가 원산지이며 내한성이 약해 우리 나라에서는 1년생 초본으로 분류하고 있다. 키는 30∼40cm로 자라며 줄기가 네모지고 가지를 많이 치며 빌로드 같은 회록색의 작은 난형의 둥근잎이 대생한다. 6∼8월에 매듭 같은 모양의 흰색 포엽에 하얀 꽃이 피는 것이 특징이며 포기 전체에 상큼하면서도 달콤한 향기가 있고 맛은 약간 쓴 편이며 꽃이 진 후에 갈색의 잔씨가 여문다. 잎은 서양에서 향신료로 많이 쓰여지고 있고 고대에는 요리와 약용 및 목욕재, 화장수, 미이라의 산화방지, 우유의 산패방지 등을 목적으로 사용되어 졌으며, 피부병, 괴혈병에 대한 효과와 진정작용 등이 알려져 있고 차류로도 이용되어 지고 있다. 특히 육류요리의 소스에 널리 이용되고 있다(Franceschi, S., Bidoli, E., La Vecchia, C., Talamini, R., D'Avanzo and Negri, E., Int. J. Cancer, 59, 181-184 (1994)).
이에, 본 발명자들은 상기와 같이 산화적 스트레스에 의한 각종 질병을 예방·치료할 수 있는 식품 첨가물 등으로 이용가능한 천연 항산화물질을 분리하기 위하여 각종 향신채소 추출물을 스크리닝 하던중, 서양 요리에 사용하는 향신료인 마조람의 메탄올 추출물에서 높은 수퍼옥사이드 라디칼 소거능을 확인하고, 이의 메탄올 추출물로부터 에틸아세테이트 분획을 얻고 다시 흡착 크로마토그래피, 크기배제 컬럼 크로마토그래피를 수행하여 수퍼옥사이드 라디칼 소거능을 갖는 물질을 정제함으로써 본 발명을 완성하였다.
본 발명의 목적은 안전성이 확립된 식물로부터 분리하여 생체내에서의 산화적 손상을 예방 또는 억제하는 식품 첨가물 등으로 이용할 수 있는 천연 항산화제를 제공하는 것이다.

상기 목적을 달성하기 위하여, 본 발명은 마조람 추출물을 유효성분으로 하는 항산화제를 제공한다.
이 때, 상기 마조람 추출물은 건조 마조람을 메탄올로 추출하거나, 메탄올 및/또는 클로로포름 및 에틸아세테이트로 순차 추출한 것이 바람직하다.
또한 본 발명은 마조람을 유기용매로 분획하고, 흡착 크로마토그래피, 크기배제 크로마토그래피로 정제한 마초람 추출정제분획을 유효성분으로 하는 항산화제를 제공한다.
또한 본 발명은 마조람을 유기용매로 추출하는 단계; 상기 유기용매 가용분획을 흡착 크로마토그래피, 크기배제 크로마토그래피를 수행하여 정제하는 단계를 포함하는 것을 특징으로 하는 마조람으로부터 항산화제를 정제하는 방법을 제공한다.
이 때 상기 유기용매는 메탄올을 사용하거나 또는 메탄올 및/또는 클로로포름 및 에틸아세테이트를 순차적으로 이용하는 것이 바람직하다. 또한 상기 크로마토그래피는 실리카겔 컬럼, 실리카 얇은막을 이용한 흡착 크로마토그래피와 세파덱스 컬럼을 이용한 크기배제 크로마토그래피를 수행하는 것이 바람직하다.
또한 본 발명은 상기 정제과정에 더하여 소수성 실리카겔 컬럼 크로마토그래피를 수행하는 것을 포함하는 항산화제의 정제방법을 제공한다.
또한 본 발명은 본 발명의 항산화제를 유효성분으로 하는 기능성식품 첨가제 또는 보조요법제로 이용가능한 조성물을 제공한다.
이하 본 발명을 상세히 설명한다.
본 발명은 마조람 추출물을 유효성분으로 하는 항산화제를 제공한다. 본 발명의 항산화제는 특히 수퍼옥사이드 라디칼 소거제로 작용한다.
본 발명자들은 천연 항산화제로 이용가능한 활성물질을 분리하기 위하여, 수십 종의 향신료를 대상으로 항산화 활성 특히, 수퍼옥사이드 라디칼 소거능을 탐색하던 중에 마조람 메탄올 추출물로부터 높은 활성을 발견하였다.
구체적으로 마조람의 건조분말을 실온에서 메탄올로 추출한 후 감압농축하여 메탄올 추출물을 얻었다. 이를 필립 등의 방법으로 수퍼옥사이드 라디칼 소거능을 확인하였다(Phillip C. Chan and Benon H. J. Bielski, J. Biol. Chem., 249(4), 1317-1319 (1974)).
상기 방법은 수퍼옥사이드 음이온 라디칼 발생기질인 크산틴과 라디칼 디텍터인 NADH 및 NADH 산화의 촉매제인 락테이트 디하이드로게나제가 함유된 반응액에 상기 마조람 메탄올 추출물을 넣고 전배양(preincubation)한 다음 수퍼옥사이드 음이온 라디칼 생성효소인 크산틴 옥시다아제를 첨가한 다음 340nm에서 흡광도 감소량을 측정하여 소거활성도(scavenging activity)를 결정한다.
그 결과 마조람 메탄올 추출물은 높은 항산화활성 특히 수퍼옥사이드 라디칼 소거능을 갖는 것으로 확인되었다.
이에 본 발명자들은 마조람에 포함된 수퍼옥사이드 라디칼 소거능 활성물질을 분리하고자, 마조람을 실온에서 95% 메탄올로 추출한 다음 적당량의 증류수에 분산시킨 후 염산으로 pH를 3으로 맞추고 클로로포름, 에틸아세테이트, 부탄올의 순으로 분획을 실시하여 각 유기용매 가용분획의 수퍼옥사이드 라디칼 소거능을 상기와 같이 확인한 결과 그중 활성이 가장 높은 에틸아세테이트 분획을 얻었다.
상기 추출물을 흡착과 크기배제의 원리로 물질이 분리되는 크로마토그래피를수행하여 그 결과 수퍼옥사이드 라디칼 소거능이 높은 활성물질을 수득하였다. 상기 크로마토그래피의 선택과 실시는 당업자에게 자명한 기술이며, 상기 수퍼옥사이드 라디칼 소거능 활성물질은 구체적인 경로에 비의존적으로 분리가능하다.
구체적으로 본 발명에서는 역상 및 순상 실리카겔 컬럼과 실리카-코팅 얇은막, 세파덱스 컬럼 크로마토그래피를 수행하였다. 상기 역상 실리카겔 컬럼 크로마토그래피는 에틸아세테이트와 메탄올 혼합용액에서 메탄올을 농도를 변화시켜 용출하였으며, 실리카-코팅 얇은막 크로마토그래피는 클로로포름과 메탄올의 10:1(v/v) 혼합용매로 평형화시킨 다음 수행하였다. 또한 세파덱스 칼럼 크로마토그래피는 50% 메탄올로 평형화한 다음 동일 용매로 용출하였고 순상 프렙(Prep) 실리카겔 컬럼 크로마토그래피는 40% 메탄올로 용출하였다.
그 결과 얻어진 활성정제분획을 다시 소수성의 실리카켈을 수지로 하는 컬럼에 주입하고, 메탄올의 농도를 달리하면서 용출하여 순도높은 수퍼옥사이드 라디칼 소거물질을 분리하였다.
상기 과정으로 분리된 활성물질은 각각 M-EA-1-T3b 및 M-EA-1-T4-2-Ⅱb로 도 1에 나타내었다.
상기의 방법으로 분리한 각 단계별 분획으로 상기 마조람 메탄올 추출물에서와 같은 방법으로 수퍼옥사이드 라디칼 소거능을 확인하였다. 그 결과 본 발명의 활성정제분획과 활성정제물질은 탁월한 소거능을 나타내었다(표 1 참조).
또한 본 발명의 활성정제물질의 지질과산화물 생성 억제활성을 측정하기 위하여, 리놀렌산에 본 발명의 활성정제물질을 가하고 UV를 조사하여 반응시킨 다음티오바비튜린산(thiobarbituric acid)을 첨가하여 이의 발색정도를 532nm에서의 흡광도를 측정하여 확인하고 흡광도 감소량을 근거로 저해율(inhibition rate)을 결정하였다. 또한 FeSO4와 아스코르브산에 의한 펜톤(Fenton)반응을 이용한 지질과산화물 생성 억제활성을 측정하였는데, 이는 상기 반응액에 FeSO4와 아스코르브산을 첨가하여 반응시킨 다음 흡광도를 확인하여 저해율을 결정하였다.
그 결과 본 발명의 활성정제물질은 기존에 천연 항산화제로 사용되는 아스코르브산, 토코페롤, 에피칼로카테킨 갈레이트(epigallocatechin gallate, 이하 'EGCg'로 약칭함), 퀴에르세틴, 에피카테킨, 루틴 등과 유사한 지질과산화물 생성 억제활성을 나타내었다(도 2 참조).
생체내(in vivo)상에서의 본 발명의 활성정제물질의 수퍼옥사이드 라디칼 소거능을 확인하기 위하여, 본 발명에서는 분화시킨 HL-60 세포주(ATCC)에 본 발명의 활성정제물질을 첨가하고 반응시킨 다음, O2-·생성을 유도하는 TPA(12-O-tetradecanoylphorbol-13-acetate) 용액을 가한후 시토크롬 c를 첨가하여 혼합한 후 배양하였다. 다음에 반응을 정지시키고 O2-·생성에 의한 시토크롬 c의 환원정도를 측정하였다.
그 결과, 본 발명의 활성정제물질은 콩으로부터 분리한 이소플라보노이드형 항암제인 제니스테인과 유사한 활성을 나타내었다(도 3 참조).
또한 본 발명에서는 생체내(in vivo)상에서의 본 발명의 활성정제물질의 과산화수소 형성 저해활성을 확인하기 위하여, 분화시킨 HL-60 세포주에 디클로로플로로레신 디아세테이트(dichloroflurorecin diacetate, 이하 'DCFH-DA'로 약칭함)를 가하여 반응시킨 다음 본 발명의 활성정제물질을 첨가하고 배양하였다. 다음에 O2-·생성을 유도하는 TPA 용액을 가한후 반응시키고 플로우사이토미터(flowcytometer)로 DCF-형광의 형광세기를 측정하였다.
그 결과 본 발명의 활성정제물질이 효과적인 과산화수소 형성 저해활성을 가짐을 확인하였다(도 4 참조).
또한 본 발명의 활성정제물질이 5,5-디메틸-1-피롤린 N-옥사이드(5,5-dimethyl-1-pyrroline N-oxide, 이하 'DMPO'로 약칭함)의 존재하의 하이포크산틴-크산틴 옥시다제 시스템에서 항산화활성을 나타내는지를 ESR을 측정하여 확인하였다. 그 결과 본 발명의 활성정제물질이 OH 피크를 감소시키는 것을 확인하였다.
이하 실시예에 의하여 본 발명을 더욱 상세히 설명한다. 하기 실시예는 본 발명을 예시하는 것으로 본 발명의 내용이 이에 한정되는 것은 아니다.
실시예 1: 마조람 메탄올 추출물의 수퍼옥사이드 라디칼 소거능 확인
마조람의 메탄올 추출물이 수퍼옥사이드 라디칼 소거능을 갖는지 여부를 확인하기 위하여 하기와 같이 필립 등의 방법을 수행하였다.
반응계는 1M 소디움 포스페이트 버퍼(pH 7.0) 50㎕, 수퍼옥사이드 음이온 라디칼 발생기질인 0.1mM 크산틴 250㎕, 라디칼 디텍터인 1mM NADH 150㎕, NADH 산화의 촉매제인 락테이트 디하이드로게나제(Boehringer Mannheim) 30㎕ 및 금속이온들의 영향을 제거하기 위한 0.5 mM EDTA 100㎕를 넣은 반응액에 시료를 50㎕ 첨가한 후 혼합하였다. 이 용액을 23℃에서 30초간 전배양한 다음 수퍼옥사이드 음이온 라디칼 생성효소인 크산틴 옥시다제 10㎕를 첨가한 후 340nm에서 1분간의 흡광도 감소량을 추적하였다.
대조구는 시료대신 시료를 녹인 용매(Methanol)를 50㎕ 첨가하여 측정하였으며, 반응계의 총부피는 상기의 반응액에 물을 첨가하여 1,000㎕로 하였다. 반응계의 각 구성성분의 최종농도는 소디움 포스페이트 버퍼(pH 7.0) 50mM, EDTA 0.05mM, NADH 0.15mM, 크산틴 0.025mM이 되게 하였다. 한편 크산틴 옥시다제와 락테이트 디하이드로게나제는 340nm에서의 분당 흡광도 변화가 0.150정도 되도록 농도를 조절하여 첨가하였다.
수퍼옥사이드 음이온 라디칼에 대한 소거활성(scavenging activity)은 다음의 식에 의해 측정하였으며, NADH의 산화를 50% 억제하는 시료의 양을 IC50으로 계산하였다.
실시예 2: 마조람으로부터 수퍼옥사이드 라디칼 소거물질의 정제
천일건조하여 분쇄한 마조람을 실온에서 메탄올로 3회 반복 추출한 후 감압농축하여 메탄올 가용분획을 얻었다. 메탄올 가용분획을 적당량의 증류수에 분산시킨 후 pH를 3으로 맞추고 클로로포름, 에틸아세테이트, 부탄올로 각각 분획한 후 유기용매 가용분획과 물 가용분획으로 분리하였다. 수퍼옥사이드 라디칼에 대해 높은 소거활성을 보인 에틸아세테이트층을 클로로포름으로 평형화된 실리카겔 컬럼(6.0 × 68 cm)에 시료를 주입하고 흡착되지 않은 성분을 클로로포름으로 세척한 후 용매의 극성도를 높여가며(0%, 2%, 5%, 10%, 20%, 100% 메탄올 순으로 단계별 용출) 각 분획 당 bed volume의 2배씩 용출하여 총 12개의 분획을 얻었다. 활성분획을 농축한 후 목적물질이 잘 분리될 수 있는 조건인 클로로포름 : 메탄올 = 10 : 1의 전개용매 조건으로 평형화된 실리카-코팅 TLC를 실시하여 활성분획을 각각 얻었다.
상기 활성분획중 하나를 50% 메탄올로 평형화된 세파덱스 LH-20 칼럼(2.5 × 38.5 cm)에 주입하고 동일 용매로 0.5 ml/분의 속도로 용출하여 활성이 있는 부분만을 농축하였다. 이를 C18 컬럼(analytic HPLC)에 주입하여 95% 메탄올 조건에서 본 발명의 활성정제물질 M-EA-1-T3b을 얻었다.
또 다른 활성분획은 40% 메탄올로 평형화된 C18 컬럼(preparative HPLC, 19 × 30 cm)에 주입하고 동일 용매로 10 ml/분의 유속으로 용출하여 활성이 있는 부분만을 농축하고 다시 C18 컬럼(analytic HPLC)에 주입하여 1% 아세토나이트릴, 3% 메탄올 조건에서 활성정제물질 M-EA-1-T4-2-Ⅱb를 얻었다.(도1 참조).

실시예 3: 본 발명의 활성정제분획의 지질과산화물 생성 억제능 측정
지질과산화물 생성 억제활성은 0.8%(W/V) 소디움 라우릴 설페이트 수용액에 리놀렌산(linoleic acid)이 0.1%(W/V) 되도록 분산시킨 기질용액 0.8㎖에 50mM 트리스 버퍼(pH 7.0) 1.0mL 및 0.1mM EDTA 0.1㎖을 혼합하고, 메탄올에 녹인 본 발명의 활성정제물질과 기존의 각종 항산화제를 0.1㎖ 가하여 총반응액의 부피를 2.0㎖이 되도록 하였다. 이 반응액을 페트리디쉬상에서 조사거리를 30 cm로 하여 UV(30 W)를 60분 동안 조사하고 반응액 1㎖을 취하여 0.44M 트리클로로아세트산(trichloroacetic acid) 0.5㎖, 0.8%(W/V) 2-티오바비튜린산(thiobarbituric acid) 0.5㎖을 가한 후 100℃에서 15분간 발색시켜 532nm에서 흡광도를 측정하였다. 측정된 흡광도 증가에 대한 각각 반응군의 흡광도 증가의 비로 지질과산화물 생성 억제능의 비율로 계산하였다.
또한 FeSO4와 아스코르브산에 의한 펜톤 반응을 이용한 지질과산화물 생성 억제활성의 측정은 0.8%(W/V) 소디움 라우릴 설페이트 수용액에 리놀렌산이 0.1%(W/V) 되도록 분산시킨 기질용액 0.8㎖에 50mM 트리스 버퍼(pH 7.0) 1.0㎖ 및 0.1mM EDTA 0.1㎖을 혼합하고, 메탄올에 녹인 본 발명의 활성정제물질과 기존의 각종 항산화제를 0.1㎖을 가하여 총반응액의 부피를 2.0㎖이 되도록 하였다. 이 반응액에 2mM 아스코르브산과 0.2mM FeSO4를 각각 0.1㎖을 가하여 37℃에서 60분 반응시키고, 이 반응액 1㎖을 취한 후 0.44M 트리클로로아세트산 0.5㎖, 0.8%(W/V) 2-티오바비튜린산 0.5㎖을 가한 후 100℃에서 15분간 발색시키고 532nm에서 흡광도를 측정하였다. 하기식과 같이 측정된 흡광도 증가에 대한 각각 반응군의 흡광도 증가의 비로 지질과산화물 생성 억제능의 비율을 계산하였다.
그 결과 본 발명의 활성정제물질은 기존에 천연 항산화제로 사용되는 아스코르브산, 토코페롤, EGCg, 퀴에르세틴, 에피카테킨, 루틴 등과 유사한 지질과산화물 생성 억제활성을 나타내었다(도 2 참조).
실시예 4: HL-60 세포에서의 수퍼옥사이드 음이온 라디칼 생성 저해활성 측정
분화시킨 HL-60 세포주(ATCC)를 PBS를 사용하여 세척하고 1,000rpm에서 5분간 원심분리를 2회 반복하여 세포를 깨끗이 세척한 후 PBS에 재현탁하여 세포농도가 1.0 × 106 세포/㎖이 되도록 한 다음 시료 50 ㎕가 첨가되어 있는 마이크로튜브에 세포현탁액을 1㎖씩 분주하였다. 37℃에서 15분간 배양하여 시료와 분화된 HL-60 세포를 반응시키고 세포외부에 있는 물질을 제거하기 위해 PBS로 2회 세척한 후 다시 PBS 1㎖로 재현탁하여 37℃에서 5분간 배양하였다. O2-·생성을 유도하기 위해 TPA용액을 5㎕ 첨가(final conc. 100 nM)하고, O2-·생성을 유도하지 않는 군은 TPA대신 EtOH을 첨가한 다음 90초 후 O2-·의 디텍터인 시토크롬 c 25㎕(final conc. 38μM)를 첨가하여 혼합한 후 37℃에서 15분간 배양하였다. 반응정지를 위해 냉수에서 5분간 정치한 다음 6,500rpm에서 2분간 원심분리 후 상층액을 취해 550nm에서 O2-·생성에 의한 시토크롬 c의 환원정도를 측정하였다(Markert, M., Andrews, P. C. and Babior, B. M., Methods Enzymol., 105, 358-365 (1984)).
수퍼 옥사이드 라디칼의 정량은 하기 식에 의해 산출하였다. 본 발명의 활성정제물질 M-EA-1-T3b의 IC50(㎍/㎖)는 50.75㎍/㎖으로 제니스테인(47.71㎍/㎖)과 유사한 활성을 나타내었다(도3 참조).
실시예 5 : HL-60 세포내에서의 과산화수소 형성 저해활성 측정
세포내 H2O2 형성저해활성의 검토는 플로우사이토미트리 분석법(flowcytometry assay)을 이용하였다. 즉 분화시킨 HL-60 세포주를 PBS를 사용, 세척한 후 1,000rpm에서 5분간 원심분리를 2회 반복하고 세포를 깨끗이 세척하여 PBS에 재현탁한 후 세포농도가 1.0 × 106 세포/㎖이 되도록 한 다음 DCFH-DA가 5㎕(final conc. 5 μM)씩 분주되어 있는 마이크로튜브에 세포현탁액을 1㎖씩 분주하였다. 37℃에서 15분간 배양을 통해 DCFH-DA와 분화된 HL-60 세포를 반응시킨 다음 시료를 첨가하여 15분간 배양하였다. O2-·생성을 유도하기 위해 TPA용액을 5㎕ 첨가(final conc. 100 nM)하고, O2-·생성을 유도하지 않는 군은 TPA대신 EtOH을 첨가한 후 37℃에서 10분간 배양하였다. 반응정지를 위해 EDTA 50㎕(final conc. 10 mM)를 첨가한 다음 PBS로 세포를 2회 세척한 후 700㎕의 PBS에 재현탁하여 플로우사이토미터로 DCF-형광의 형광세기(fluoroscence intensity)를 측정하였다. 플로우사이토미터로 DCF-형광 측정시의 여기(exitation)와 방출(emision) 파장은 각각 488와 525nm로 하였다.
본 발명의 활성정제물질 M-EA-1-T3b를 25, 50㎍/㎖의 농도로 적용한 결과 각각 HPP = 18.63%, 23.32%, IE = 74.04%, 63.37%로 나타나 활성정제분획의 효과적인ROOHs 형성 저해활성을 관찰할 수 있었다(도4 참조).
실시예 6: 본 발명의 활성정제물질의 ESR 측정
5mM 하이포크산틴 40㎕, 1mM 디에틸렌트리아민펜타아세트산 (diethylenetriaminepentaacetic acid, DETAPAC) 20㎕, 9M DMPO 20㎕, 50mM PBS(pH 7.4∼7.8) 140㎕를 넣은 반응액에 디메틀 술폭사이드(dimethyl sulfoxide)에 용해된 시료 20㎕를 첨가한 후 이 용액에 475 mU/㎖ 크산틴 옥시다제 40㎕를 첨가하여 측정하였다. 대조구는 시료대신 DMSO 20㎕를 넣어 측정하였다.
그 결과 본 발명의 활성정제물질 M-EA-1-T3b이 첨가된 경우 OH 피크가 감소되는 것을 확인하였다(도 5 참조).
상기에서 살펴본 바와 같이, 본 발명의 마조람으로부터 분리한 수퍼옥사이드 라디칼 소거제 및 그 정제방법은 마조람의 메탄올 추출물로부터 항산화활성 특히, 라디칼 소거활성을 갖는 천연물질을 분리함으로써 이를 이용하면 생체내에서 산화적 손상에 의한 성인병, 고령화병 등의 질환을 예방 또는 억제하는 다기능 천연 항산화제가 포함된 기능성식품을 개발할 수 있다.

- 마조람을 메탄올, 클로로포름, 에틸아세테이트로 순차적으로 추출하고, 흡착 크로마토그래피, 크기배재 크로마토그래피로 정제하여 얻어진 마조람 추출물을 유효성분으로 함유하는 기능성식품 첨가물로 이용가능한 조성물.
- 마조람을 메탄올, 클로로포름 및 에틸에세테이트로 순차 추출하는 단계와;상기 추출 분획을 흡착 크로마토그래피, 크기배제 크로마토그래피를 수행하여 정제하는 단계를 포함하며, 상기 흡착 크로마토그래피는 고정상으로 실리카겔 컬럼을 이용하고 이동상으로 농도구배 에틸아세테이트 메탄올 혼합용매나 아세토나이트릴 메탄올 혼합용매를 사용하고, 상기 크기배제 크로마토그래피는 50% 메탄올로 평형화된 세파덱스 LH-20 컬럼을 이용하는 것을 특징으로 하는 마조람으로부터 수퍼옥사이드 라디칼 소거제를 정제하는 방법.
