(a) the subject is identified or monitored to have or maintain a subthreshold anti-uricase antibody level; and/or (b) the treatment dose and/or frequency is determined based on identification or monitoring. and/or (c) the method further comprises one or more steps for identifying or monitoring the subject to obtain anti-uricase antibody levels; and/or (d) the co-administration is and/or (e) a composition comprising polymeric synthetic nanocarriers comprising PLA, PLA-PEG, and rapamycin and a composition comprising uricase together every 2-4 weeks for example, a composition comprising polymeric synthetic nanocarriers comprising PLA, PLA-PEG, and rapamycin and a composition comprising uricase is administered monthly; -PEG and a composition comprising a polymeric synthetic nanocarrier comprising rapamycin at 0.05 mg/kg, 0.08 mg/kg, 0.1 mg/kg, 0.125 mg/kg, 0.15 mg/kg, 0. is administered at a dose of rapamycin of 2 mg/kg, 0.25 mg/kg, or 0.3 mg/kg; and/or (g) a composition comprising uricase is administered at 0.1 mg/kg, 0.2 mg/kg, and/or (h) the uricase is a pegylated uricase, e.g., the pegylated uricase is a pegsiticase or a pegroticase and/or (i) the subject is in need thereof, e.g., the subject (A) has elevated serum uric acid levels; (B) has gout or gout or (C) predicted to have gout flares from treatment with gout therapy,
said composition.

co-administering, to a subject with a subthreshold anti-uricase antibody level (e.g., titer), 1) said composition comprising a polymeric synthetic nanocarrier comprising rapamycin; and 2) a composition comprising pegsiticase. A method wherein the composition comprising polymeric synthetic nanocarriers is administered at a dose of rapamycin between 0.05 mg/kg and 0.3 mg/kg and the dose of the composition comprising pegsiticase is 0 .1 mg/kg to 0.5 mg/kg of pegsiticase, said composition for use in said method;
(a) the subject is identified or monitored as having or maintains an anti-uricase titer below a threshold; and/or (b) treatment dose and/or frequency are determined based on identification or monitoring. and/or (c) the method further comprises one or more steps for identifying or monitoring the subject to obtain anti-uricase antibody levels; and/or (d) the co-administration is and/or (e) a composition comprising a polymeric synthetic nanocarrier comprising rapamycin and a composition comprising pegsiticase are co-administered every 2-4 weeks; and/or (f) a composition comprising a polymeric synthetic nanocarrier comprising rapamycin and a composition comprising pegsiticase is administered monthly; and/or (g) a composition comprising a polymeric synthetic nanocarrier comprising rapamycin. is 0.05 mg/kg, 0.08 mg/kg, 0.1 mg/kg, 0.125 mg/kg, 0.15 mg/kg, 0.2 mg/kg, 0.25 mg/kg, or 0.3 mg/kg and/or (h) a composition comprising pegsiticase is administered at 0.1 mg/kg, 0.2 mg/kg, 0.3 mg/kg, 0.4 mg/kg, or 0.5 mg /kg of pegsiticase; and/or (i) the polymeric synthetic nanocarrier comprises a hydrophobic polyester, optionally the hydrophobic polyester comprises PLA, PLG, PLGA or polycaprolactone; and /or the polymeric synthetic nanocarrier further comprises PEG, e.g., PEG is conjugated to PLA, PLG, PLGA or polycaprolactone, e.g., the polymeric synthetic nanocarrier comprises PLA, PLG, PLGA or and/or (ii) has gout or a gout-related condition; or (iii) appears to have gout flares from treatment with gout therapy. is predicted to be
said composition.
