Abstract
PURPOSE:To analyze a sample such as serum contg. a material to be examined precisely with high sensitivity at a low dilution rate by incorporating a free SH group into antibodies or part of the antibodies or antigens to be conjugated. CONSTITUTION:Lipid contg. a halogenated alkyl group is first synthesized by bringing halogenated acetic acid, etc., into reaction with suitable amino group- contg. lipid in the presence of N-hydroxysuccinimide, etc. Phospholipid and glycolipid contg. the suitable halogenated alkyl group and further, a suitable amt. of cholesterol and other lipids are dissolved and mixed together with a solvent in and with said lipid. After the solvent is sucked away, an aq. soln. contg. a labeling material is added to the mixture under shaking to prepare the liquid suspension of liposome. On the other hand, the free SH group is imparted to the antibodies, or part of the antibodies or the antigens by subjecting the same to an enzyme treatment, reduction treatment, etc. The halogenated alkyl group of liposome and the SH group thereof react with each other and the chemical conjugation takes place if these antibodies or part of the antibodies or antigens and the liquid suspension of liposome are mixed under suitable conditions. The desired reagent for analysis is thus obtd.
