Abstract

PURPOSE:To efficiently sort a microorganism, capable of decomposing lignin and suitable for saccharifying wood, producing paper pulp, etc., with a high accuracy, by cultivating a microorganism in a culture medium containing wood meal and guaiacol and examining coloration of reddish brown color. CONSTITUTION:(A) Preferably 0.1-0.5% wood meal having preferably a particle size passing through a 20 mesh screen prepared by drying and finely cutting wood, such as Japanese beech or Japanese oak, (B) preferably 0.1-1.0mM guaiacol, such as a dry distillation product of guaiacum polymer, and (C) preferably 1-2% agar and used to prepare a culture medium. (D) Wood-decaying fungi separated from a sample in the natural world, such as decayed wood, are then inoculated and cultivated to judge and sort the aimed strain by the presence or absence of a reddish brown colored zone.
