Description
本発明は歯肉炎、歯槽膿漏等の予防薬を含有す
る歯磨。更に詳しくはアミラーゼ酵素を配合した
歯磨に更にアルキルベタインを添加せしめた歯磨
に関する。
アミラーゼ酵素はα−1,4結合でつながつた
D−グルコース単位を3個以上もつポリサツカラ
イドのα−1,4グルコース結合を加水分解する
酵素で、動植物界に広く分布し、ある種の微生物
にも産生される。アミラーゼ酵素はデンプン分解
作用、消炎作用等を有し、消化薬等の医薬品とし
て利用される。又歯料領域においては口腔内疾
患、特に歯肉炎、歯槽膿漏等の予防、進行阻止お
よび治療薬として有用である。
しかしながら、一般に使用されている通常の歯
磨などにアミラーゼ酵素を配合した場合、比較的
高温で長期間保存した時、アミラーゼ酵素が活性
を失い、歯肉炎・歯槽膿漏予防歯磨として効果が
期待できない。
本発明者は歯磨にアミラーゼ酵素を配合した場
合、アミラーゼ酵素の不活性化を防止し、安定化
させるため種々検討した結果、アルキルベタイン
を添加せしむることで所期の目的を達成できるこ
とを見いだした。
従つて本発明の目的はアミラーゼ酵素を配合し
て、歯肉炎・歯槽膿漏等を予防する歯磨におい
て、アミラーゼ酵素の活性を長期間安定化せしめ
た歯磨を提供するものである。
本発明における歯磨とは、練り歯磨、潤性歯
磨、歯ぐきマツサージ用クリームなどを意味し、
研磨剤、湿潤剤、結合剤、発泡剤などは通常使用
されるものが用いられる。
例えば、研磨材としては、炭酸カルシウム歯磨
用第二リン酸カルシウム、不溶性メタリン酸ナト
リウム、ピロリン酸カルシウム、無水珪酸、水酸
化アルミニウムなどが、湿潤剤としてはグリセリ
ン、プロピレングリコール、ポリエチレングリコ
ール、ソルビトールなどが、粘結剤としてはメチ
ルセルロース、カルボキシメチルセルロースナト
リウム、アルギン酸ソーダ、カラギーナンなど
が、発泡剤としてはラウリル硫酸ソーダ、ラウロ
イルサルコシンナトリウム、ポリオキシエチレン
ソルビタン、ポリオキシエチレンノニルフエニル
エーテルなどが用いられる。また本発明の歯磨に
アルミニウムアラントイネート、グリチルリチン
酸塩、塩化ナトリウム、クロルヘキシジン、アズ
レン、ヒノキチオールなどの他の種々の薬効剤を
配合してもよい。
本発明に使用されるアルキルベタインは一般式
(1)または(2)で表される化合物である。

ウリルベタイン、ジメチルオレイルベタイン、ジ
メチルミリスチルベタイン、ジメチルステアリル
ベタイン、メチルエチルラウリルベタイン、メチ
ルn−プロピルラウリルベタインなどがあげられ
る。また一般式(2)で表される化合物は、次の一般
式で表わされるエトキシ化アルキルベタインから
選ばれる。

アルキルベタインの添加量はその種類、各々の
処方、例えば研磨剤、粘結剤、湿潤剤などの種
類・量などによつて異なるが、好ましくは約0.1
〜約20%(重量)さらに好ましくは約1〜約5%
(重量)程度添加する。
次に本発明によつて得られた歯磨は、アミラー
ゼ酵素が安定化され、歯肉炎、歯槽膿漏などの防
止に効果があるが、以下に実験例を示す。
(アミラーゼ酵素の分析法)
歯磨10gを500mlに分散させ、その上澄液1ml
にて2%でんぷん液10mlを加え40゜にて6分間反
応させる。反応後この液0.5mlに0.01%I2−0.02%
