Abstract

PURPOSE:To obtain a monoclonal antibody to be specifically reacted with human osteosarcoma useful with respect to medicine and clinical diagnosis, by cloning an immunocyte of mouse. CONSTITUTION:An animal such as mouse is immunized against an antigen (e.g. isolated antigen, antigen-containing tumor cell or homogenate thereof) to be recognized by a monoclonal antibody and an antibody-forming cell is obtained from an immunized animal. Then the cell is fused with a myeloma cell to give a fused cell, which is screened to give a fused cell to produce a monoclonal antibody. Then the fused cell is cloned and cultivated in a medium to give a supernatant liquid of the culture. The supernatant liquid is separated and purified to give a monoclonal antibody such as mouse monoclonal antibody 2H10 or 2D3 to be specifically reacted with human osteosarcoma.
