Abstract

PURPOSE:To easily determine pyruvic acid with a stable dry chemistry reagent, by using a novel enzyme having high thermal stability and capable of converting pyruvic acid into acetylphosphoric acid, CO2 and a reduced-type electron acceptor in the presence of phosphoric acid and an electron acceptor. CONSTITUTION:A specimen containing pyruvic acid is added with (A) phosphoric acid, (B) an electron acceptor (e.g. phenazine methosulfate), (C) a formazan dye (e.g. nitrotetrazolium blue) and (D) a novel pyruvic acid dehydrogenase catalyzing the reaction of formula and stable at pH 6.3 and 50 deg.C for 15min, developing the color of the produced reduced-type electron acceptor with the formazan dye, etc., and the extinction of the reaction system is determined to determine the amount of pyruvic acid in the specimen.
