Abstract

PURPOSE:To stably determine adenosine triphosphate (ATP) with a high sensitivity and accuracy, by using hexokinase and pyruvate kinase together, amplifying reaction forming glucose 6-phosphate from D-glucose with a specific chemiluminescence reagent. CONSTITUTION:ATP is determined. In the process, D-glucose which is a substrate is converted into glucose 6-phosphate and the ATP is simultaneously converted into adenosine diphosphate (ADP), which is then converted into ATP with pyruvate kinase. Thereby oxidative-reductive reaction in which the ATP is infinitely rotated in principle to integrate the glucose 6-phosphate that is decomposed with glucose 6-phosphate dehydrogenase to simultaneously convert the nicotinamide adenine dinucleotide phosphate (NADP) into reduced form NADP is used to cause chemiluminescence of 1-methoxy-5-phenazinemethyl sulfate and isoluminol. The ATP is determined by an enzymic amplification method.
