Abstract

PURPOSE:To analyze an unstable fluorescent derivative in a stable state while suppressing the lowering of sensitivity, by guiding vitamin K in a specimen to the derivative held to a reaction column and subsequently changing over a valve to guide the derivative to an analytical column. CONSTITUTION:An introducing liquid/washing liquid sump 11 and a reaction liquid sump 12 are connected to a change-over valve 15 through a liquid feed pump 13. Further, a mobile phase liquid sump 16 is connected to the change-over valve 15 through a liquid feed pump 17 and a pre-column 18. Furthermore, a reaction column 14 and the analytical column 3 connected to a fluorescence detector are connected to the change-over valve 15. Then, by changing over the change-over valve 15 to a broken line side, a derivative reaction flow passage and an analytical flow passage are formed and the change-over valve 15 is changed over to a solid line side to incorporate the reaction column 14 in the analytical flow passage. By this constitution, the lowering of sensitivity due to dilution and the rising of a blank value due to the existence of a reagent are prevented and a fluorescent derivative can be analyzed in a stable state.
