Abstract

PURPOSE:To provide advantageous preparation of a plant having resistance to cucumber mosaic virus (CMV), by forming a gene capable of coding a 3A protein of the CMV having a specific amino acid sequence. CONSTITUTION:RNA3 as a template and a reverse transcriptase are used to synthesize a cDNA. Since the resultant cDNA is a single-stranded DNA, it is used as a template to provide a double-stranded cDNA according to a conventional method. The obtained double-stranded cDNA is then introduced into a vector, such as PUC line or M13mp, and transformed into Escherichia coli HB101, JM103, etc., to carry out cloning and readily afford a clone having the cDNA capable of coding the aimed 3A protein as an inserted DNA. The gene capable of coding the 3A protein is useful for preparing a plant resistant to CMV.
