Abstract

PURPOSE:To obtain a large amount of an active human lysozyme by recombinant DNA method, by using DNA prepared by bonding DNA to code a specific signal peptide of to 5' end of DNA to code a human lysozyme. CONSTITUTION:DNA which has leucine at fourth of amino acid sequence of signal peptide of glair lysozyme replaced with phenylalanine, isoleucine at sixth with leucine and valine at eighth with alanine, (i) selects a codon having high use frequency in yeast, (ii) uses sequence of a yeast PGK gene in the upper stream of ATG to raise an amount of manifestation and (iii) determines nucleotide sequence to enable construction of hybrid signal and codes a signal sequence shown by the formula is prepared. The DNA is bonded to DNA which is chemically synthesized based on the amino acid sequence of human lysozyme and codes a human lysozyme having TaqI site in the vicinity of N end. A cell transformed by using the prepared DNA is cultivated and human lysozyme is used and accumulated in the culture mixture.
