Description
本発明はシチジンジホスホコリン(以下、
CDP−コリンという)を含有する経口投与に適
した医薬組成物に関する。さらに詳しくは、リン
脂質物質と結合したCDP−コリンを含有し、経
口投与に適した医薬組成物に関する。
CDP−コリンが人間のいくつかの病理学的症
状の治療に用いられていることは知られている。
代表的なCDP−コリンの治療上の特徴は、たと
えばアテローム硬化症に起因する出血、大脳血栓
症および脳障害などのような頭蓋の損傷または内
因性の疾患による意識障害の治療である。そのほ
かパーキンソン病様症候群の治療にも有用である
ことが証明されている。
CDP−コリンの治療学的な作用を説明するに
あたり、CDP−コリンがリン脂質の生合成にお
ける前駆体であるということを考慮に入れておか
なければならない。この点については、CDP−
コリンの生化学的な意味は充分に確認されてお
り、人間に対して1日あたり100〜1000mg投与さ
れる。通常の投与形態は非経口投与であり、一般
に有効成分としてCDP−コリンを5〜25%
(W/V)含む水溶液に処方されたものが投与さ
れる。
実際に使用されているCDP−コリンを含有す
る医薬組成物は、いずれのばあいでも経口投与は
適当でない。というのは、CDP−コリンは吸収
されにくく、さらに胃腸管においてそこに存在す
る酵素により劣化されやすいからである。CDP
−コリンを含有する経口投与剤がないということ
は、CDP−コリン自体の治療学的な用途を大き
く制限しているということがいわれないままに今
日に至つている。
本発明はCDP−コリンまたはその塩を有効成
分として含有してなり、前記の従来の欠点を排除
できる医薬組成物を提供することを目的としてい
る。
すなわち本発明は、リン脂質類を含有する物質
と結合したCDP−コリンまたはその塩を含有す
る経口投与に好適な医薬組成物に関する。本明細
書において結合とは、物理化学的な会合または分
子間の結合(molecular combination)のことを
いう。リン脂質類としては、たとえばレシチン類
などがあげられる。
物理学的会合は、CDP−コリンまたはその塩
の水溶液に固体状態、乳濁液状態または溶液状態
のリン脂質類を加えて混合することにより好適に
えられる。該会合の安定性を改善するためにたん
ぱく様物質および架橋剤(cross−linking
agent)を適当に配合してもよい。たんぱく様物
質としてはたとえば血清アルブミン、卵アルブミ
ンまたは他の食用アルブミンが好ましい。また架
橋剤としてはたとえばホルマリンなどが好まし
い。CDP−コリン/リン脂質類の重量比は1:
20〜1:0.5の範囲が好ましい。
本発明の医薬組成物は、従来治療に用いられて
きた通常のCDP−コリン水溶液以上にきわめて
有用な新規な医薬組成物である。CDP−コリン
の経口投与が可能になるということとは別に、
CDP−コリンの治療学的活性について特筆すべ
き相乗作用が観測された。そのことは、 14Cでラ
ベルされたCDP−コリンを含有する医薬組成物
を用いて明らかにされた。
つぎに実施例をあげて本発明を詳細に説明する
が、本発明はこれらの実施例のみに限定されるも
のではない。
実施例 1
(リン脂質と会合しているCDP−コリンの製
造)
CDP−コリンのナトリウム塩100gを水100ml
に溶かした。別途、撹拌装置を取りつけた容器中
で顆粒状の植物性リン脂質400gを調製した。こ
のリン脂質顆粒を一定速度で撹拌しつつ前記
CDP−コリンのナトリウム塩の水溶液を墳霧し、
ついで該顆粒を真空下もしくは非真空下において
70〜80℃のオーブン中で乾燥した。かくしてえら
れたものは、抽出後異なつた部分から採取したい
ずれのサンプルにおいても同じCDP−コリン含
有率であつた。製剤されたものはCDP−コリン
とリン脂質とが会合しており、理化学的に均一で
かつ経口投与に好適な医薬組成物を調製するのに
適当なものであつた。
CDP−コリン/リン脂質の比率をかえたほか
は前記と同様の手順にしたがつて、2つの化合物
の重量比が1:20〜1:0.5である製剤をえた。
前記のごとく、本発明における製剤の物理化学
的安定性はたんぱく質様物質および架橋剤によつ
て向上せしめることができる。
実施例 2
CDP−コリン100gを血清アルブミン5gを含
有する水100mlに溶かしてえられた水溶液を、実
施例1と同様にしてリン脂質の顆粒400gに噴霧
した。コーテイングされた顆粒に再び撹拌しなが
らホルムアルデヒドを5%含有する水100mlを架
橋剤として噴霧した。えられた物質を最後に実施
例1と同様にして乾燥して本発明に用いる製剤を
えた。
試験例
(経口投与によるリン脂質と会合した 14Cでラ
ベルされたCDP−コリンの吸収およびその分
布状態)
最初のCDP−コリン水溶液中に 14Cでラベル
されたCDP−コリン(以下、 14C−CDP−コリ
ンという)を加えたほかは実施例1と同様に処方
して本発明に用いる製剤をえた。ラベルした炭素
原子は、コリン部位のメチル基の炭素原子であ
る。一般に 14Cの放射性物質濃度はCDP−コリ
ン1gあたり50μキユリーであつた。
(A) 14C−CDP−コリンの水溶液および 14C−
CDP−コリンとリン脂質との処方の水性懸濁
液を胃管栄養(gastric gavage)により平均体
重200gを有するラツトに投与した。投与量は
コリン50mgに相当する量であつた。異なつた時
間間隔で一定量ずつ採血し、実験を開始してか
ら24時間後にはラツトを解剖した。種々の臓器
または組織における前もつて決めておいた容量
または重量の血清中の放射性物質濃度を測定し
た。血清中の濃度の時間変化の結果を第1図に
示し、臓器における濃度を第1表に示す。

本発明の医薬組成物の治療学的に利点は苦痛
を伴なわない投与法を提供することができると
いう利点にとどまらず、血清中に高レベルの放
射性物質濃度が保持されていることからわかる
ように、1回の投与のみで長時間薬理作用が持
続されるという利点をも有しているのである。
(B) 体重約2Kgの2匹のマカクザル
(cynomolgus)を用いてつぎの試験を行なつ
た。
放射性物質濃度が0.1μキユリー/mgの 14C−
CDP−コリンを25mg/Kg体重(2.5μキユリー/
Kg体重に相当)静脈内に一度に投与した。つい
で投与時および投与後5分、10分、15分、30
分、60分、2時間、4時間、7時間、24時間お
よび30時間目に採血し、それぞれの血清中のデ
イー・ピー・エム(d.p.m.)濃度を測定した。
血清中のデイー・ピー・エム濃度−時間曲線の
グラフから、生体からのみかけの脱離速度定数
(β値)および生物的半減期が計算できる。こ
れらの値は、同じサルにおける全デイー・ピ
ー・エム濃度−時間曲線を推定するために必要
な30時間以降の血清中濃度−時間曲線下面積
(以下、AUCという)を計算するのに用いられ
る。投与から30時間までのAUCはトラペジア
ム法(trapezium methods)によつて計算さ
れる。
この試験終了後同じサルに本発明の組成物を2
つの異なつた投与量で投与した。対照として
CDP−コリンを本発明の組成物の投与量に対応
する量経口投与した(対照投与)。各試験はそれ
ぞれ10〜15日間の間隔をおいて行なつた。CDP
−コリンの量で表わした投与量は200mg/Kg体重
および400mg/Kg体重であり、それぞれ2μキユリ
ー/Kg体重および4μキユリー/Kg体重の放射性
物質濃度に相当する。各試験の順序はランダムに
行ない、2匹の動物に重複して行なつた。投与後
30時間を経過するまで一連の検査用血液を採血
し、それぞれの血清中のデイー・ピー・エム濃度
を測定した。
1つのデイー・ピー・エム濃度−時間曲線に基
づいて投与から30時間以内のAUCを計算するこ
とができる。
静脈内投与も含めた各投与後、0〜24時間、24
〜48時間および48〜72時間の各期間ごとに尿を集
め、尿に排出されるCDP−コリンの割合を調べ
た。
第2表および第3表にそれらの結果を示す。ま
た第4表に2匹のサルを用いてえられた結果(第
2表および第3表)の平均および相対標準偏差を
示す。第2〜4表において本発明の組成物は、ア
ソニツク(ASONIC)として表示してある。

静脈内投与したばあい、○は 14C−CDP−コリ
ンを200mg/Kg体重(2μキユリー/Kg体重)を含
有する本発明の組成物を経口投与したばあい、●
は 14C−CDP−コリンを400mg/Kg体重(4μキユ
リー/Kg体重)を含有する本発明の組成物を経口
投与したばあい、△は 14C−CDP−コリンを200
mg/Kg体重(2μキユリー/Kg体重)経口投与し
たばあい、▲は 14C−CDP−コリンを400mg/Kg
体重(4μキユリー/Kg体重)経口投与したばあ
いの結果である。
第2〜4表および第2〜5図から明らかなごと
く、本発明の組成物を経口投与したばあい、 14C
−CDP−コリンの顕著な吸収を示している。す
なわち本発明の組成物を投与したばあい、最高濃
度点に達する時間が対照投与によるそれに比べて
早く、しかもその最高濃度は2種の投与量のいず
れにおいても対照投与( 14C−CDP−コリン単
独投与)よりも明らかに高い。とくに投与量が低
いときにその違いがはつきりと現われている(第
5図参照)。
本発明の医薬組成物の絶対的な生物学的利用能
はサルにおいて50%以上であることが立証され
た。すなわちつぎの各CDP−コリン投与量での
F値はそれぞれつぎのとおりであつた。
200mg/Kg体重→F=0.5109
400mg/Kg体重→F=0.7781
一方CDP−コリンを単独で投与したとき、各
投与量における絶対的な生物学的利用能はかなり
低いことがわかつた。すなわち、
200mg/Kg体重→F=0.2143
400mg/Kg体重→F=0.6174
このように本発明の医薬組成物は広範囲で吸収
され、さらに投与量に伴なつて吸収の割合も明ら
かに増加する。
以上の試験結果から、本発明の医薬組成物は静
脈内におけるCDP−コリンを比較したばあい、
すぐれた絶対的な生物学的利用能を有している。
本発明における絶対的な生物学的利用能は、低投
与量においては50%以上であり、さらに高投与量
においては80%近くまであつた。一方、ラツトを
用いた試験では、投与量のほとんど全量の吸収が
みられた。
本発明の医薬組成物においては、速度論的なま
たは経口投与における生物学的利用能がいちじる
しく改善された。というのは、吸収がより迅速に
起りかつ低投与量においてはほとんど吸収されて
しまうからである。さらにはサルにおける吸収の
割合は、CDP−コリンの単独投与量で観察され

The results are for oral administration of body weight (4μ cuyries/Kg body weight). As is clear from Tables 2 to 4 and Figures 2 to 5, when the composition of the present invention is orally administered, 14 C
- Shows significant absorption of CDP-choline. That is, when the composition of the present invention is administered, the time to reach the maximum concentration point is faster than that when the control administration is performed, and the maximum concentration is lower than that of the control administration ( 14C -CDP-choline) at any of the two dosages. clearly higher than that of single administration). The difference is especially obvious when the dose is low (see Figure 5). The absolute bioavailability of the pharmaceutical compositions of the invention was demonstrated to be greater than 50% in monkeys. The absolute bioavailability in the present invention was more than 50% at low doses and even close to 80% at high doses. On the other hand, in tests using rats, almost the entire dose was absorbed. In the pharmaceutical composition of the present invention, the kinetic or oral bioavailability is significantly improved. This is because absorption occurs more rapidly and is mostly absorbed at low doses. Furthermore, the rate of absorption in monkeys was higher than that observed with a single dose of CDP-choline.
