Description
本発明はびやくだん(sandalwood)の特徴を
有しかつ嗅覚用試薬として利用性を有する新規な
α−ヒドロキシアルキル−4−t−アルキルシク
ロヘキサンに関するものである;本発明はまたこ
の化合物を含有する香料組成物に関するものであ
る。
香料組成物に極めて望ましいニユアンスを付す
る多くの他の天然材料と同様に、東インド諸島び
やくだん油(East Indian Sandalwood Oil)は
天然生成物特有の変動を受け易い。その高いコス
トと限定された供給の故に香料は必然的にこの価
値ある成分の使用が限定される。
従つて、びやくだん芳香を有する合成材料を供
するために努力が続けられている。α−及びβ−
サンタロールは所望の主な芳香であるびやくだん
油の成分であることは公知であるが、これらの化
合物の合成に対し何ら工業的に利用できる方法が
ない。要するに、経済的でありかつ容易に入手し
得るびやくだん芳香を有する任意の材料が香料組
成物における天然のびやくだん油の置換物又は一
部置換部として即時のそしてかなりの商業的関心
がもたれる。
本発明に従つて式:
(式中R1及びR3はC1-3−アルキル基であり、R2は
メチル又はエチルでありそしてR1+R2+R3の合
計中の炭素原子の総数は6を越えない)のα−ヒ
ドロキシアルキル−4−t−アルキルシクロヘキ
サンが供される。
本発明の化合物は種々の方法で製造できる。
各々の場合で好適である方法は通常には必要な出
発材料のコストと入手性により決定される。

According to the invention the formula: α of (wherein R 1 and R 3 are C 1-3 -alkyl groups, R 2 is methyl or ethyl, and the total number of carbon atoms in the sum of R 1 + R 2 + R 3 does not exceed 6) -Hydroxyalkyl-4-t-alkylcyclohexane is provided. The method which is preferred in each case will usually be determined by the cost and availability of the necessary starting materials. Thus, the new compound has the formula:

を、R3を供給するオルガノ金属誘導体と反応さ
せることにより製造できる。
この新規な化合物及び必要な出発材料は一般に
種々の方法により適当なt−アルキルベンゼン又
は4−t−アルキルシクロヘキサノンから製造で
き、そのあるものを下記のチヤートに示す。あ
る場合には、チヤートに示すもののような中間
体はこれらが市販されている時に利用され又は当
業者に公知の別法により容易に製造される。
チヤートの経路Aに示すように、t−アルキ
ルベンゼンはフリーデルクラフツ型アルカノイル
化を介してアルカノイル化して所望の4−t−ア
ルキルアルカノイルベンゾンaから本質的にな
る組成物を供する。次にこのように製造されたケ
トンは好適な触媒の存在で水素添加されて所望の
アルコールを供する。
経路Bに示すように4−t−アルキルシクロヘ
キサノンは適当なアセチリド、R4−C≡CMe
〔R4=H、CH3−、C2H5−、MeはLi、Na又はK
である〕と反応してエチルアルコールを供し、こ
れは公知のルーペ転位(即ちギ酸の影響下で)を
介して適当なアルカノイルシクロヘキセンに転換
される。次に4−t−アルキル−1−アルカノイ
ルシクロヘキセンは1段階又は2段階の何れかで
水素添加されて所望のアルコールを供する。
経路Cは、4−t−アルキル−シクロヘキサン
は4−t−アルキルシクロヘキセンに転換する方
法を示すが、最初にアルコールに還元して次に脱
水してオレフインを供する。このオレフインは次
の公知のコンダコフ反応を介して4−t−アルキ
ル−1−アルカノイルシクロヘキセンcを含む
組成物へ転換され、次にこのシクロヘキセンは前
記のように所望のアルコールに転換される。経
路Dは、クロル酢酸のエステルを使用する時には
4−t−アルキルシクロヘキサノンをアルデヒド
d(R5=H)に、又は高級α−クロル酸を使
用する時にはケトンd(R5=R3)に転換する周
知のダルツエンの反応を利用する別法を示す。こ
のように製造されたケトンはカルボニル基を適当
に還元して(即ちケトンをアルコールに還元する
ために適した還元剤を使用することにより)所望
のアルコールへ転換できる。ダルツエンの反応
のアルデヒド生成物は適当な、即ちR1-3−アルキ
ル供給オルガノ金属誘導体、例えばアルキルリチ
ウム試薬又はグリニヤール試薬、例えばR3Li、
R3MgCl、R3MgBr、R3MgJとアルデヒドを反応
させることにより所望のアルコールへ転換でき
る。
前記のように、本発明のアルコールは幾つかの
方法で製造できる。本発明で中間体として必要と
される1−アルカノイル−4−t−アルキルベン
ゼン、1−アルカノイル−4−t−アルキルシク
ロヘキセン、1−アルカノイル−4−t−アルキ
ルシクロヘキサン、及び4−t−アルキルシクロ
ヘキシルカルボキシアルデヒド(1−ホルミル−
4−t−アルキルシクロヘキサン)誘導体の多く
は当業者に公知であり、又はチヤートに示され
るもののように当業者に周知の化学反応を利用す
ることにより製造できる。例えば:1−アセチル
−4−t−ブチルシクロヘキセンはエム・エス・
ニユーマン(M.S.Newman)等のJ.Am.Chem.
Soc.82、4098(1960)に報告され;1−ホルミル
−4−t−ブチルシクロヘキサンはビー・クロス
(B.Cross)等のJ.Chem.Soc.、3895(1960)に報
告され;4−t−ブチルアセトフエノンはジエ
イ・シー・バトラー(J.C.Butler)等のJ.Am.
Chem.Soc.、76、1906(1954)に報告されてい
る。
チヤートの方法Aにおいて、水素化分解を最
少にするためにロジウム又はルテニウム触媒の何
れかを使用して所望のアルコールに1−アルカ
ノイル−4−t−アルキルベンゼン化合物を還元
することが好ましい。
触媒の量、溶媒、反応温度、水素圧及び反応時
間(多分互いに無関係ではないけれど)に関する
要件は臨界的ではないと思われる。装入されたケ
トンの0.5ないし10重量%の範囲に及ぶ量でアル
ミナ触媒上の5%ロジウムを使用することが好ま
しい。水素添加は25ないし60℃の温度及び30ない
し300psi(1psi=0.068気圧)の圧力で好適に行
なわれる。
溶媒の使用は任意である。この水素添加は溶媒
なしに、又はエタノール、酢酸、ペンタン等の適
当な不活性溶媒中で行なわれる。挙げられた触媒
の量、与えられた温度と圧力、並びに使用された
溶媒は例示するものであり、本発明を限定する意
図はない。
別法として、このケトンの水素添加は最初に当
業者に公知の方法により(例えばNaBH4、
LiAlH4、通常の触媒還元等)カルボニル基をア
ルコールに還元すること、続いて前記のようにロ
ジウム触媒を使用して生成するベンジルアルコー
ルの水素添加により2段階で行なわれる。
チヤートの方法B及びCに従つてα・β不飽
和ケトンを飽和アルコールへ水素添加するための
水素化技術で公知の多くの方法により1−アルカ
ノイル−4−t−アルキルシクロヘキセンを新規
なアルコールに水素添加することができる。不
飽和部分の一つが他の前に水素添加される2段階
法が使用できる。しかしながら、単一段階法で両
方の二重結合を水素添加することにより1−アル
カノイル−4−t−アルキルシクロヘキセンを還
元することが好ましい。
触媒の量、溶媒、反応温度、水素圧と反応時間
(多分互いに無関係ではないけれども)に関する
単一段階法に対する要件は臨界的ではないと思わ
れる。α・β−不飽和ケトンを飽和アルコールに
転換するために多くの触媒、例えば白金、パラジ
ウム、ラネーニツケル、ロジウム、ルテニウムが
当業者に公知である。
経済的なラネーニツケルを使用することが好ま
しく、その理由はこの触媒はシス異性体が主であ
る、例えば60−85%である生成物を生ずることが
予想外に判明したからである。(シス及びトラン
ス異性体へ1−(α−ヒドロキシエチル)−4−t
−ブチルシクロヘキサンの分離はこの化合物の所
望のびやくだん香料性質が主としてシス異性体に
よつていることを示す。)更に、温度、溶媒圧、
及び触媒の量に関して条件は臨界的ではないと思
われる。
触媒の好適量は装入されたケトンの量の5−15
%である。好適な温度範囲は80ないし120℃であ
り、好適な圧力範囲は60ないし600psiである。溶
媒は使用してもよいが、水素添加は溶媒なしで進
行するので必ずしも必要ではない。溶媒を使用す
る場合には、好適な溶媒は水素添加条件下で低い
蒸気圧を有するものである。温度範囲、圧力範囲
及び触媒量のこのリストは好適な反応条件を示す
ように与えられ、そして本発明を限定する意図は
ない。与えられた範囲外の条件は殆どの場合に適
用可能である。触媒としてラネーニツケルを使用
した時に形成された生成物がシス異性体に富んで
いることは全く予想外であつた。1:1混合物は
試みられた他の方法で見出されると予想された。
比較して、ラネーニツケルを使用した時には、
生じたα−ヒドロキシアルキル−4−t−アルキ
ルシクロヘキサンはシス異性体に富み、トランス
異性体に対するシス異性体の比は約2−3対1の
範囲内である。
方法Dに従つて4−t−アルキルシクロヘキシ
ルカルボキシアルデヒドは当業者に公知の多くの
方法を使用して適当な金属アルキルR3Mと反応さ
せ得る。適当な反応不活性溶媒中の金属アルキル
の溶液へ一定の時間にわたつてこのアルデヒドを
添加することにより反応を行なわせることそして
反応の終了時に酸のようなプロトン性源で最初の
付加物を分解することが好ましい。
この金属アルキルは適切にはグリニヤール試薬
(例えばR3MgX、ここでXはCl、Br又はIであ
る)又はリチウムアルキル(例えばR3Li)であ
り、これらのすべては市販され、又は当業者に公
知の方法により製造できる。
溶媒は好ましくはエチルエーテル、THF等の
エーテル、又はエチルエーテル−トルエン、
THF−トルエン、THF−ベンゼン等のエーテル
−炭化水素混合物である。この工程は当業者に周
知である。
本発明の化合物は香料として用途を見出し、か
つ香水、石けん及び他の化粧品に使用できる。こ
れらは特に、絶対的ではないが、天然のびやくだ
ん油の置換物又は一部置換物として有用である。
α−及びβ−サンタロールの公知の構造に全体
的に非関連である単一環状構造体を有する、式
のこれらの化合物に天然のびやくだん油に類似し
た著しい芳香性質が付与されることは極めて驚く
べきでありかつ予想外である。
またR=Hである構造の化合物は何れにして
もびやくだんに類似せず、かつ本発明の化合物に
固有の価値ある芳香性質を全く欠いていることは
注目すべきである。
本発明の化学品の各々に共通の香料の性質はび
やくだんの特徴である。分離されたα−ヒドロキ
シアルキル−4−t−アルキルシクロヘキサンの
すべてはこの特徴を有した。この特徴はこのシリ
ーズ、1−(α−ヒドロキシエチル)−4−t−ブ
チルシクロヘキサンの低級同族体で最も有効かつ
強烈であり、そして高級同族体で分子量が増大す
るにつれて減少する。これはヒドロキシアルキル
の分子量又はt−アルキル基の大きさの何れかが
増大する場合であることが示された。15個以上の
炭素原子を有する同族体は通常の条件下でびやく
だんの特徴を検知するには弱い芳香を有すると考
えられる。
シス及びトランス異性体へ1−(α−ヒドロキ
シエチル)−4−t−ブチルシクロヘキサンの分
離は前記に概観したように、この化合物の所望の
びやくだん香料性質が主としてシス異性体による
ものであることを示す。
香料技術に使用される芳香族油、濃縮物及び化
学品と混和される時には、本発明の新規なα−ヒ
ドロキシアルキル−4−t−アルキルシクロヘキ
サンはこの調合品に天然のびやくだんの特徴を付
与する。この調合品で天然のびやくだん油に置換
し及び/又は拡大するこれらの性能が特別の利点
である。
下記の実施例は本発明のプラクチスを更に例示
するために供されるが、好適な具体化の目的のた
めのみであり、限定とて解釈されるべきでない。
特記しない限り、温度は℃であり、赤外線帯域
はcmの逆数であり、核磁気共鳴スペクトルはクロ
ロホルム−d1で行なわれ、そしてシグナルは
TMS(δ0.0)に相対的なδ単位で報告され、そ
して分子量は質量分析法により決定される。気液
クロマトグラフイーがまた生成物を分析するため
に使用された。
実施例
1−アセチル−4−t−ブチルシクロヘキセン
から1−(α−ヒドロキシエチル)−4−t−ブ
チルシクロヘキサン
A 2段階法
エチノール200ml中の1−アセチル−4−t
−ブチルシクロヘキセン70g(0.39モル)の溶
液を50psi水素圧の下でパル(Parr)水素添加
装置で木炭上の5%パラジウム0.5gで水素添
加した。理論量(0.39モル)の水素が吸収され
た時に、反応を停止し、混合物をろ過し、濃縮
し、そして減圧下蒸留してシス及びトランス−
1−アセチル−4−t−ブチルシクロヘキサン

4−t−ブチルアセトフエノンからシス及びト

−ブチルシクロヘキサン
4−t−ブチルアセトフエノン17.6g(0.10モ
ル)、アルミナ上の5%ロジウム1.3g、数滴の酢
酸及び無水エタノール10mlの混合物を、水素摂取
が止むまで室温で50psiの水素圧パル装置で水素
添加した。気液クロマトグラフイー(glc)によ
り、この水素化物は8%の1−アセチル−4−t
−ブチルシクロヘキサンを含有した:それ故に、

4−t−ブチルアセトフエノンの代りに1−
(α−ヒドロキシ−エチル−4−t−ブチルベン
ゼンに水素添加した時に同一の結果が得られた。
初期の水素添加に存在した少量の1−アセチル
−4−t−ブチルシクロヘキサンを除去するため
