Description
本発明は局所軟膏剤、詳しくはカルボキシビニ
ルポリマーをゲル化剤(軟膏基剤)として利用し
た改良された局所軟膏剤に関する。
従来よりセルロース、カラヤゴム、ペクチン、
ゼラチン、コラーゲン等の天然高分子化合物、ヒ
ドロキシプロピルセルロース、CMC−Na、デキ
ストリン、グリチルリチン酸ジカリウム等の天然
高分子化合物及びその誘導体、ポリビニルアルコ
ール、カルボキシビニルポリマー、ポリビニルピ
ロリドン等の合成高分子化合物等をゲル化剤とし
て利用した医薬品は、鎭痒剤、鎭痛剤、消毒殺菌
剤、パツプ剤等として種々知られている。これら
のうちで特にカルボキシビニルポリマーは、製剤
中のアルコール含有量に影響を受けない所からゲ
ル化剤として慣用されている。しかして該カルボ
キシビニルポリマーによる製剤の増粘及びゲル化
は、製剤中に中和剤殊にトリエタノールアミン、
ジイソプロパノールアミン、トリエチルアミン等
の有機アミンを配合して上記ポリマーのカルボキ
シル基を中和することにより行なわれている〔特
公昭52−18250号公報、同54−18329号公報、同56
−10886号公報等参照〕。しかしながら上記中和剤
として用いられる有機アミンは、近年ある条件下
に試験管内及び生体内でN−ニトロソ化合物に転
化し、該N−ニトロソ化合物は魚類からサルに至
るまで多くの動物種族に対して強いガン原性を示
す発ガン性物質であることが報告された〔食衛誌
vol.15、No.6、1974年12月、第419〜433頁参
照〕。また、アメリカ食品薬品庁(FDA)は、市
販の化粧品サンプル36種類について、ニトロソア
ミンの分析を行ない、このほどその結果を発表し
た。それによれば、その半数にニトロソアミンが
検出され、最もひどい例では13万PPb(0.13%)
という値が得られた。このニトロソアミンは主と
してN−ニトロジエタノールアミンであり、
FDAの分析ではその他のニトロソアミンも付随
的に僅か発見されたといわれている。さらに、有
機アミン類は1980年発行の官報16105号及び16106
号によれば、皮膚アレルギーの原因になり得るこ
とが示されている。そのため、人体に直接使用さ
れる医薬部外品に含まれる有機アミンは、化粧品
成分として表示しなければならないもの即ちジエ
タノールアミン、ジイソプロパノールアミン、ト
リプロパノールアミン及びトリエタノールアミン
〔厚生省告示第167号参照〕の他に、モノエタノー
ルアミンが表示義務を負つている。
発明者は、従来よりゲル化剤として慣用される
上記カルボキシビニルポリマーの安全性の高い中
和剤について、有機アミンに代わるべき、数多く
の薬品について検討した。その結果、中和剤を用
いず直接、塩基性の主成分即ちジフエンヒドラミ
ン、ジブカイン、リドカイン、クロルフエニラミ
ン及びエフエドリンから選ばれた少なくとも一種
の所定量を配合するときには、カルボキシビニル
ポリマーは増粘し、軟膏剤形態の透明で極めて良
好なゲルが得られ、しかも上記塩基性化合物は、
その本来の薬効を全く阻害されることなく発揮す
るという全く新しい知見を得た。本発明はこの新
知見に基づいて完成されたものである。
即ち本発明は、メントールおよび(または)カ
ンフルと共に有効成分としてジフエンヒドラミ
ン、ジブカイン、リドカイン、クロルフエニラミ
ンおよびエフエドリンからなる群から選ばれた少
なくとも1種の塩基性化合物0.5〜3.5重量%を含
有し、且つゲル化剤としてカルボキシビニルポリ
マー1〜3重量%を配合されてなり、20℃におけ
る粘度が5万〜25万センチポイズ(B型粘度計
BHタイプ、7号ローター、4rpm使用による)の
範囲にある軟膏剤形態を有することを特徴とする
局所軟膏剤に係る。
本発明の局所軟膏剤は、従来より消炎、鎭痛剤
等の有効成分として慣用されているメントールお
よび(または)カルフルと共に、ジフエンヒドラ
ミン、ジブカイン、リドカイン、クロルフエニラ
ミンおよびエフエドリンからなる群から選ばれた
少なくとも1種の塩基性化合物の0.5〜3.5重量%
を配合させることを必須とする。上記各塩基性化
合物は、それら自体抗ヒスタミン剤、局所麻酔
剤、アドレナリン作働神経剤の有効成分として知
られるものであるが、本発明では、これを上記有
効成分としてのみならず、ゲル化剤とするカルボ
キシビニルポリマーの中和剤としても兼用する点
をその最大の特徴とし、これに基づいて従来のこ
の種ゲル化剤を用いた製品に見られる如き、発ガ
ン性物質として作用するニトロソアミン類を生成
するおそれのある中和剤の使用を回避して、非常
に安全で、しかも上記塩基性化合物本来の薬効を
も充分に発揮させるに成功したものである。該塩
基性化合物の配合量は上記範囲内にあることが必
要であり、これが上記範囲を外れる場合は、所望
の軟膏剤形態の製品の収得が困難となる。
また本発明軟膏剤においてゲル化剤とするカル
ボキシビニルポリマーとしては、従来よりこの種
ゲル化剤として慣用される各種のものをいずれも
使用できる。その代表例としては例えば米国グツ
ドリツチ・ケミカル社(B、F、Goodrich
Chemical Co.)から市販される「カーボポール
(Carbopol)934、940及び941」又は日本純薬社
から市販される「ジユンロンPw−110」、和光純
薬社から市販される「ハイビスワコー103、104及
び105」を例示することができる。該カルボキシ
ビニルポリマーの配合量は、1〜3重量%の範囲
から選択され、これは上記特定の塩基性化合物に
より中和(架橋)され、製品粘度を5万〜25万セ
ンチポイズ(20℃における、B型粘度計BHタイ
プ、7号ローター、4rpm使用、以下同じ)の範
囲の適当な軟膏剤形態とする。上記カルボキシビ
ニルポリマーの配合量が上記範囲を外れる場合
は、所望の粘度範囲を有する軟膏剤形態が発現さ
れない。
本発明軟膏剤は上記特定の塩基性化合物及びカ
ルボキシビニルポリマーの夫々の所定量を配合す
る点を除いては、通常この種の軟膏剤と同様にし
て調整され、また他に適当な添加剤を添加配合す
ることができる。適当な添加剤としては例えばグ
リセリン、プロピレングリコール、ポリエチレン
グリコール、ハイドロプロピルセルロース、メチ
ルポリシロキサン、ポリビニルピロリドン及びポ
リビニルアルコール等の塗布感改良剤又は保湿
剤、ポリオキシエチレン硬化ヒマシ油
(POE20、40、80、100モル付加)、ポリオキシエ
チレンポリオキシプロピレングリコールエーテ
ル、エマレツクス640等の界面活性剤等を例示す
ることができ、之等の添加剤は通常使用される総
量として一般には製剤中に約5〜20重量%の範囲
で配合することができる。
本発明軟膏剤の調整方法は、特に限定されるも
のではないが、通常メントール及び(又は)カン
フルに他の添加剤をアルコール水溶液とした後、
これにカルボキシビニルポリマーの水溶液を加
え、更にジフエンヒドラミン、ジブカイン、リド
カイン、クロルフエラミン及びエフエドリンから
成る群から選ばれた少なくとも1種の塩基性化合
物を加え、充分に撹拌することにより行なわれ
る。ここで用いられるメントール及び(又は)カ
ンフルは、夫々d−体、l−体及びdl−体のいず
れであつてもよい。之等メントール及び(又は)
カンフル及び他の添加剤の溶媒として用いられる
アルコール類としては通常のもの例えば代表的に
はエチルアルコールが用いられるが他の低級アル
コール類例えばメタノール、イソプロパノール等
を単独で又は上記エチルアルコールと併用しても
よい。之等溶媒の使用量は有効成分化合物を溶解
できることを前提として適宜に決定できるが、通
常得られる製剤中に約30〜70重量%の範囲となる
量とするのが適当である。
かくして本発明によれば、非常に透明且つ長期
間安定で延びのある塗布感の良好な、所望粘度を

塗擦を容易にすると共に、ゲル化剤と有効成分と
する塩基性化合物との結合が離れ、該塩基性化合
物が次第に界面活性剤の浸透作用の影響も受けて
皮膚表面より吸収され、かくしてその本来の鎭
痛、消炎作用乃至は局所麻酔作用、抗ヒスタミン
作用等が長期に亘つて持続発現される。しかも本
発明軟膏剤は、従来のこの種ゲル化剤を用いた製
剤の如く中和剤としてのトリエタノールアミンや
ジイソプロパノールアミン等を配合していないた
め、之等に基づく発ガン性の危険はなく、更に皮
膚アレルギー等の危険もない。
以下本発明を更に詳細に説明するため本発明局
所軟膏剤の実施例を挙げる。

規定量を量りとり、40゜以下でかき混ぜながら溶
解する。次に、d−カンフル及びl−メントール
を添加し、均一に溶解する。さらに、液を激しく
混ぜながらジフエンヒドラミンの規定量を徐々に
加え、ゲルを形成させる。その後しばらくかき混
ぜて均一にする。これらの操作は真空下で行な
う。
かくしてPH5.6、20℃における粘度18.8×104セ
ンチポイズ、ゲル硬度(飯尾電機社M301ARカー
ドメーターによる)700ダイン/cm2の無色透明な
局所軟膏剤を得た。
