Abstract
PURPOSE:To enable the quantity of callicrein to be specifically as well as highly sensitively measured, through preparation of a sample excellent in sensitivity by performing chromatography by the use of an adsorbent. CONSTITUTION:A sample solution is prepared by adding radioactive compound labeled callicrein to a sample and mixing them with each other and is charged in the upper part of a column (a) filled with silica gel equilibrated by a buffer of pH 7.0. Then, development is performed, and when there is no more supernatant, this is developed by a sample-developing buffer A, and 99% enzymes other than callicrein are adsorbed and removed. The solution having passed is developed by using a column (b) filled with fixing trypsin inhibitor in order to adsorb callicrein. After the column is washed with the sample-developing buffer A, elution is conducted by using a developing buffer B, and with the resultant eluate used as a samle the enzyme activity of urine callicrein is measured. Thereby, it is possible to highly sensitively measure an extremely small quantity of callicrein present in human urine.
