Abstract

PURPOSE:To quantitate a hemoglobin bound to glucose by using of a liquid chromatographic filter consisting essentially of glucose(meth)acrylate as a fixed phase, and an eluate of water at 3.0-9.0pH. CONSTITUTION:To a 2l separable flask is added a mixture of 400ml of a 4wt% aqueous solution of polyvinylalcohol, 90g tetraethyleneglycol, 10g tetramethylolmethanetriacrylate and 1.5g benzoylperoxide, and further 100g toluene. Then, the mixture is stirred at 400r.p.m. for 10hr at 80 deg.C, and the reactant is cooled. After cooling, 5-20mum porous polymer is obtained and prepared for a column of chromatography. The chromatograpy is performed at 6pH by use of eluate of 0.1N phosphate buffer solution, and pH is changed to 7 immediately prior to the appearance of a big peak 5 for a standard hemoglobin to produce peaks 2-4 for a hemoglobin bound to glucose.
