Abstract

NEW MATERIAL:A hepatic basic protein (BLP), present in the liver, capable of exhibiting a molecular weight of about 35,000 by the SDS polyacrylamide gel electrophoresis and a molecular weight of about 100,000 by the thin layer gel filtration with "Sephadex G200 Superfine ") and a weakly basic isoelectric point by the isoelectric point electrophoresis.
USE: A protein specifically present in the liver effective for the diagnosis and the judgment of the recuperation of various diseases by sensing the amount of BLP in blood.
PROCESS: An extract from the liver of a rat is precipitated by ammonium sulfate having a saturation degree of 35W60%, and the BLP is then precipitated from the resultant fraction. Thus, the extract is salted out by the ammonium sulfate having a saturation degree of 50W60%, equilibrated by a weakly basic buffer solution and chromatographed by "DEAE-Sepharose ", and the unadsorbed substance is then recovered. The recovered substance is further purified by the affinity chromatography to give the aimed BLP of high purity.
