Abstract

PURPOSE: A synthesis of duplex polydeoxyribonucleoside containing a structural gene useful for the insertion into a cloning vector in order to exhibit an arbitrary polypeptide.
CONSTITUTION: The parallel hydrogen bond formation and the formation of covalent bond in series are carried out by using two single-chain oligodeoxyribonucleoside fragments which are designed in such a way that each terminal fragment has the recognition side of restricted enzyme by itself or together with the fragments linked in the upward or downward stream when the terminal group parts are formed by each terminal fragment contituting the terminal group parts of the upward or downward stream side through the parallel hydrogen bond formation, each terminal fragment has at least one pair of bases adjoining the upward or downward stream side of the recognition side of restricted enzyme, and the terminal group parts are made into substantially insensible terminal group parts by the pair of bases.
