Abstract

PURPOSE:To determine the Hb saccharified (GHb) to grape sugar out of hemoglobin (Hb) directly by adsorbing a sample contg. the Hb in a silica gel packed column, then eluting the same with a phosphoric acid buffer soln. contg. urea or guanidine and NaCl. CONSTITUTION:A sample contg. Hb is adsorbed to the top end of a column packed with silica gel, more particularly, silica gel having carboxy alkyl groups such as carboxy methyl groups and having >=30Angstrom fine pores. On the other hand, an elutate A prepd. by dissolving NaH2OP4 and urea or guanidine in desalted water and controlling pH to 6.50 and a soln. B prepd. by dissolving NaH2PO4 and NaCl and controlling the pH to 6.50 are prepd. and A soln. A discharge valve and a soln. B discharge valve are programed to flow these solns. to the column in such a way that the soln. A and the soln. B have inverse gradient concns. and that the urine is contained from 1mol/l to 0 and NaCl from 0 to 0.2mol/l, whereby the respective fractions of the Hb are eluted. Thereby, the GHb in the Hb is separated and determined accurately and the results are used for diagnosis and treatment of diabetes.
