Abstract

PURPOSE:To collect the recombinant DNA containing the chromosome gene of human interferon-beta from recombinant DNAs obtained by fragmentizing the whole DNA of human chromosome with restriction enzyme to a proper length, concentrating the fragments, and inserting into vector DNA. CONSTITUTION:The whole DNA of human chromosome is fragmentized to a proper length by the partial digestion with a restriction enzyme such as HaeIII, AluI, etc., and concentrated by electrophoresis, etc. as it is or after collecting the fraction having a proper length. The fraction is inserted in the DNA such as E. coli phage to obtain recombinant DNA or a human gene library which is a collection of the recombinants including the whole human genes. A recombinant DNA having a DNA exhibiting complementarity to the human interferon-beta messenger RNA is labeled with radioactive isotope, and used as an explorer to select and separate the recombinant DNA having the DNA fragment containing human interferon-beta gene, from the above library.
