Abstract
PURPOSE: A compound in which p-toluenesulfonyl group is introduced on a nitrogen atom in an imidazole ring is hydrolyzed under an alkaline condition to produce a guanidine derivative used as an antiulcerative in high yield.
CONSTITUTION: N-Methyl-N'-cyano-N"-{2-[( 1-p-toluenesulfonyl-5-methyl-4-imidazolyl)methylthio]ethyl}guanidine of formulaI(Ts is formula II) is hydrolyzed under an alkaline condition with, e.g., sodium hydroxide or potassium hydroxide, to produce N-methyl-N'-cyano-N"-{2-[(5-methyl-4-imidazolyl)methylthio]ethyl} guanidine of formula III. The starting compound of formulaIis obtained by successive reactions of 4-hydroxymethyl-5-methylimidazole with p- toluenesulfonyl chloride, a compound of formula V (X is F; Y is residue of an acid) and another compound of formula VI.
