Abstract

PURPOSE: To perform a fractional determination of hemoglobin (HbA1) in a blood simply by eluting a glucosidated hemoglobin (HbA1) in order of the fraction, HbA1(a+b), HbA1C.
CONSTITUTION: A column 22 packed with a weakly acidic cation exchange resin 20 and equilibrated with anionic buffer solution 21, is used. After flowing out the buffer solution 21, a hemolytic reagent (solution A) is added into the column. Then, the HbA1(a+b) is eluted with the first eluant 23 consisting of anionic buffer solution (a buffer solution containing a phosphate and a basic salt and having 0.03W 0.07M of ionic concentration) and an eluate (solution B) is obtained. Further, the HbA1C is eluted with the second eluant 24 consisting of anionic buffer solution (pH6.4W7.7 and 0.08W0.18M of cationic concentration) and an eluate (solution C) is obtained. Then, the each absorbancy of said solutions, A, B and C is measured and each concentration of HbA1(a+b), HbA1C is calculated using formulae I and II.
