Abstract

PURPOSE: An enzymatic reaction is effected using a bacteria strain having penicillin acylase activity but losing β-lactamase activity, thus producing β-lactam antibiotic in high yield.
CONSTITUTION: The reaction between a β-lactam ring compound such as 6-aminopenicillanic acid and an α-aminoacid such as D-phenylglycine or its reactive derivative is conducted in the presence of a culture medium of the above strain, the bacterium cells or their treated product to produce a β-lactam antibiotic such as amicillin in high yield. As the bacterium strain, is used, e.g., Pseudomonas maltophilia M-14 (a mutant derived from Pseudomonas maltophilia IFO 12020). The cultivation is conducted at a pH of 3W8, preferably 6.5W7.5 and a temperature of 5W40°C. After the reaction, the objective compound is isolated and purified by extraction with an organic solvent, precipitation or isoelectric precipitation.
