Abstract
PURPOSE:A hybrid DNA having genes of a microorganism which belongs to Thermus is introduced into the arrangement of the nucleotide to change the properties of a microorganism so that it grows at room temperature to produce selectively said heat-resistant enzyme. CONSTITUTION:A gene of a microorganism belonging to Thermus, e.g., chromosome DNA of Thermus theremophilus HB8 (ATCC-27634), and a plasmid DNA as a vector are digested with a restriction enzyme by a usual method and rebound with a ligase to prepare a hybride DNA having a gene of a microorganism belonging to Thermus in the nucleotide arrangement. The resultant recombined DNA is introduced into a microorganism which grows at room temperature such as Escherishia coli C600 (FERM-P5044) to produce a modified microorganism, which is cultured. The cells are collected, crushed, then the crushed mixture is heated to deactivate enzymes with no heat resistance, resulting in collection of said heat-resistant enzyme.
