Abstract

PURPOSE:To prevent delayed occurrence of gap due to contraction of filler agent, etc., by introducing specimen and moving phase solvent to a stationary phase through a sliding plug provided on the top of a separating column and also applying pressure onto the sliding plug continuously. CONSTITUTION:A sliding plug 7 is provided on the top of a separating column 1, and by introduction of specimen and moving phase solvent to a stationary phase 6 through this sliding plug 7, chromatography is conducted. During operation of this chromatography, a prescribed magnitude of pressure is applied onto the sliding plug 7 through a pressure-applying mechanism. By doing so, a filler agent which constitutes the stationary phase 6 in the separating column 1 is tightly pressed by the sliding plug 7, and therefore, a satisfactorily tight stationary phase can be constituted not by the wet-type filling process which requires complicate operation but by the dry-type filling process which can be operated easily.
