Abstract

PURPOSE:To detect the ketone body in body fluid easily with good sensitivity by using an aqueous nitroprusside soln. in which yttrium salt is allowed to coexist. CONSTITUTION:An aqueous soln. contg. about 0.005-0.6M/l alkali metallic salt of nitroprusside (preferably, Na, K salt) and about 0.1-2M/l yttrium salt [e.g. YCl3, Y2(SO4)3.8H2O, etc] is used as a coloring reagent, and measurement is made at about 4-7pH. In the case of measuring, for example, ketone body (acetoacetic acid) in body fluid (urine), the above-mentioned coloring reagent is added to the liquid to be examined, and after a fixed time, its absorbancy is determined by using a spectrophotometer, with water as contrast, while the absorbancies of a standard acetoacetic acid and coloring reagent liquid are separately determined, thence the acetoacetic acid concn. in the liquid to be examined is determined by using the formula.
