Abstract
PURPOSE:To find the concentration of pyruvic acid by utilizing POP reaction, by using the fixation enzyme membrane prepared by immobilized enzyme POP on the porous high molecule membrane which is provided on the surface of electrode as the enzyme electrode. CONSTITUTION:Platinum electrode is used for the working electrode 12 and silver chloride electrode is used for the reference electrode 13. POP bridged on acetylcellulose film by adsorption by 0.025% glutaric aldehyde, is used for the immobilized enzyme membrane 16 and the enzyme electrode 10 is made by attaching the membrane 16 on said electrodes through the oxygen permeating membrane 15 made of teflon. The electrode 10 is contacted with the sample solution and also, applying voltage of electrode is set at -0.5--0.7 constant voltage. Then, decreased quantity of the enzyme accompained by POP reaction, is detected as the variation of current of electrode and the concentration of pyruvic acid is found. Severed kinds of biochemical substance are measured because pyruvic acid is contained in the product product not only by a single reaction of the enzyme but by a conjunction of two or more reactions of the enzyme.
