Abstract

NEW MATERIAL:1α-Hydroxyvitamin D3 derivatives of formula I (R is H or hydroxyl).
EXAMPLE: Cholesta-5,7-diene-1α,3β-diol-hemisuccinate.
USE: They have strong vitamin D-like activity and are used as a hapten for preparing the antibody for the determination of vitamin D derivatives by the radioimmunoassay method.
PREPARATION: The reaction between a steroid derivative of formula II and succinic anhydride in the presence of a basic substance as pyridine is effected under an inert atmoshpere as nitrogen. The excess reactants are distilled off and the residue is passed over a column chromatography to form a derivative of formula III in which the 3-position is substituted. The compound of formula III is dissolved in a solvent as hydrocarbon and irradiated with ultraviolet rays at room temperature under an inert atmosphere to be converted into previtamine D3 derivative of formula IV, which is isomerized into title compound of formula I.
