Abstract

PURPOSE: To analyze and quantitatively measure bile acid correctly in a short time by converting conjugated bile acid and free bile acid into fluorescent substances, thereafter separating them by liquid chromatography, and measurring the fluorescences thereof.
CONSTITUTION: Bile acids including both conjugated and free types, derived from a sample to be analyzed are esterified using as an esterification reagent, 4-bromomethyl-7-methoxy coumarin in the presence of a crown ether, and converted into bile acid esterfied compounds having fluorescences. Thereafter, the bile acid esterifed compounds are separated respectively by liquid chromatography. The thus separated bile acid esterified compounds are respectively subjected to fluometric analysis.
