PURPOSE: To prevent the deactivation and denaturation of urokinase, and to enable the reactivation of deactivated urokinase, by adding a guanine (derivative) to a urokinase-containing composition.
CONSTITUTION: The deactivation of urokinase during the purification of a crude urokinase solution obtained from human urine, or during the storage of purified urokinase powder, is prevented by adding 5×10-5W8×104 moles of a guanine dervative such as guanine and 1-methylguanine per 1 mole of protein in the urokinase. Purification of the urokinase solution is pref. carried out at a pH of 5W10, an ionic strength of 0.02W0.8 and a temperature of 0W15°C using a buffer solution containing acetic aicd, phosphoric aicd, etc. To improve the storage stability, the above stabilizer is added to the composition prior to freezing, drying etc.
