Abstract

PURPOSE: To enable the mixture to be separated to each component readily and rapidly by stopping the development by a developing solvent when the mixture is separated into each component in a column made of the material that can be cut and cutting the column of the required components for each chromato tube.
CONSTITUTION: A column chromato tube 1 is made by using easy-to-mold polypropylene and glass wool is laid on the bottom thereof, thence powder sugar is filled therein. This is connected to a suction bottle 4' and petroleum ether is flowed from above while being sucked. On the other hand, spinach is acetone-extracted and the extract liquid is evaporated. The resultant black greenish solid is dispersed in petroleum ether to prepare a dispersion. Pouring this dispersion into the column and subsequent development with petroleum ether yield the chromatogram of separation state. Here, the development is suspended and the column is cut for each chromato tube in each dye part region to separately take out the powder sugar adsorbed with each dye. These are respectively extracted with ether, whereby the respective components are obtainable as pure components.
