Abstract

PURPOSE: To realize measurement of concentration of oxide type substrate of oxidoreductase, by using oxide type substrate as the substrate, and selecting a substance having a baser potential than the oxidation-reduction potential of coenzyme as the redox compound.
CONSTITUTION: An enzyme electrode 1 having a silver plate 3 dipped as the current collector in a buffer 2 such as phosphoric acid is communicated with a counter electrode 4 having a platinum current collector 6 dipped in a buffer 5 by way of a separator 9. The electrode 1 is further connected to a saturated calomel reference electrode 7 through a salt bridge 8. First, a redox compound, for instance, methyl viologen (oxide type) is dissolved in the buffer 2 to be turned into a reduced type at a potential of -0.5V with respect to the electrode 7. Next, a coenzyme, for instance, HADH, and an enzyme, for instance, alcohol dehydrogenase, are dissolved in the buffer 2. When the cathode current reaches the stationary value at the silver plate 3, an oxide type substrate, for instance, acetaldehyde, is added to the buffer 2, so that the concentration of the oxide type substrate may be determined from the increment of the cathod current.
