1位でオルト置換されたアリール(または
アラルキル)基を有し、抗痙攣作用を有する新規の
イミダゾリン−2,4−ジオンおよびその製造方法
本発明は1位でオルト置換されたアリール(またはアラルキル)基を有するイ
ミダゾリン−2,4−ジオン、その製造方法および中枢神経系の疾患、特に様々
な形式のてんかんを治療する薬剤としてのその使用に関する。
アリールグリシンと尿素(Ber.10,2045(1970);Khim.Geterosikl.Soed.1
978,87)、シアン酸ナトリウム(特開平5−178802号公報)またはシア
ン酸カリウム(J.pr.Chem.1926(113),233)との反応により、ないしはN−
アシルウレタンの環化(Drug Res.1968,1(1),189)により製造することがで
きる1−アリール−イミダゾリン−2,4−ジオンは公知である。公知の方法で
は1位でオルト置換されたアリール基を有するイミダゾリン−2,4−ジオンの
製造は除外されている。
1位でオルト置換されたフェニル基を有するイミダゾリン−2,4−ジオンは
これまで記載されていなかった。
さらにベンジル基がメトキシ基によりオルト置換さ
れている1−アラルキル−イミダゾリン−2,4−ジオンが記載されている(特
開平5−178802号公報)。1−アラルキル−イミダゾリン−2,4−ジオ
ンは、相応するアラルキルグリシンまたはアラルキルグリシンエステルとシアン
酸カリウムとを反応させ、かつ引き続き塩化水素を含有する氷酢酸中で環化する
と得られる(J.het.Chem.11,(1974))。
抗痙攣作用はこれらの化合物に関して言及されていないか、または想到されて
いない。
抗痙攣作用を有する数多くの化合物が公知である。しかし今日でもまだ全ての
てんかん性の疾患を満足に治療することはできない。
従って本発明の課題は、例えば抗てんかん作用を有する薬剤として使用できる
ような、有利な薬理学的特性を有する新規の化合物を提供することである。
本発明に相応してこの新規の化合物は、一般式1:
[式中、
XはC1〜C4アルキル、トリフルオロメチルおよびn=1の場合にはハロゲン
も表し、
Yは水素、ハロゲンを表す]の1−アリール(またはアラルキル)−イミダゾ
リン−2,4−ジオンである。ただし1−(2−メチルフェニル)−イミダゾリ
ン−2,4−オンは除外する。
CH2基の数は0(1−アリール−イミダゾリン−2,4−ジオン)または1
(1−アラルキル−イミダゾリン−2,4−ジオン)である。
一般式1の化合物の例として以下のものが挙げられる:
1−(4−クロロ−2−メチル−フェニル)−イミダゾリン−2,4−ジオン
1−(2−クロロベンジル)−イミダゾリン−2,4−ジオン
1−(2,6−ジフルオロベンジル)−イミダゾリン−2,4−ジオン
1−(2,6−ジクロロベンジル)−イミダゾリン−2,4−ジオン
本発明に相応して一般式1の化合物は、一般式2:
[式中、
XはC1〜C4アルキル、トリフルオロメチル、ハロ
ゲンを表し、
Yは水素、ハロゲンを表す]の化合物とクロロスルホニルイソシアネートとを
反応させることによる新規の方法により製造することができる。
有利には反応を溶剤中、温度−20℃〜120℃で実施する。溶剤として有利
にはハロゲン化した脂肪族炭化水素、例えば塩化メチレン、クロロホルムおよび
四塩化炭素が問題となる。芳香族炭化水素例えばベンゼンおよびトルエン、なら
びに脂肪族エーテル例えばジエチルエーテル、テトラヒドロフランおよび1,4
−ジオキサンは同様に適切な溶剤である。
1−アラルキル−イミダゾリン−2,4−ジオンはJ.het.Chem.11(1974)と
同様に製造することができる。
本発明による化合物は薬学的組成物の製造に適している。薬学的組成物は本発
明による化合物を1種以上含有していてもよい。医薬品を製造するために通例の
薬学的担体および助剤を使用してもよい。
薬剤は非経口(例えば静脈内、筋肉内または皮下)で、または経口で投与して
もよい。投与形は薬学の実地で一般に公知であり、かつ通例の方法で製造するこ
とができる。
本発明による化合物は強い抗痙攣作用を有する。該化合物をマウスへのi.p.
投与により生体内で国際的に通例の標準(Pharmac.Weekblad,Sc.Ed.14,132
(1992)およびAntiepileptic Drugs,第3版、Raven Press,New York 1989)に
より、その抗痙攣作用を試験した(表1)。例えば化合物8(1−(2,6−ジ
フルオロベンジル)−イミダゾリン−2,4−ジオン)については、マウスの場
合最大電気ショックに関して36mg/kgでED50(i.p.)、s.c.ペンテ
トラゾールテストでは50mg/kgでED50、および神経毒性に関しては12
3mg/kgでNT50(i.p.)が測定された。これと比較して公知の抗てんかん
薬例えばカルバマゼピンおよびバルプロ酸カルシウムは最大の電気ショックをモ
デルでまたはペンテトラゾールテストでもたらす。

本発明による新規の化合物の製造を例に基づいて詳しく説明する。
一般式1の化合物を製造するための一般的な規定は表2に記載の例1〜9であ
る。
一般式2のN−アリールグリシンエステル0.2モルをジクロロメタン250
mlに溶解する。温度−15℃〜−20℃で少量ずつクロロスルホニルイソシア
ネート0.25モルを滴加する。反応時間2時間後、沈殿した塩化スルファモイ
ルを分離する(繰り返し置換したアリール基の場合)または塩化メチレンを留去
して乾固する(一置換されたアリール基の場合)。固体残留物を水300mlで
、80℃で1時間撹拌する。冷却後、沈殿物を分離し、かつ引き続き20%塩酸
200mlで2時間還流下に加熱する。清澄な溶液を氷浴で冷却し、かつ沈殿し
た結晶泥をエタノールから再結晶する。
一般式1の化合物を製造するための一般的な規定は表2に記載の例6〜9であ
る。
J.het.Chem.11(1974)と同様に一般式2のN−アラルキルグリシンエステル
0.2モルを33%塩酸40mlに溶解する。室温で水50mlに溶解したシア
ン酸カリウム0.25mlを徐々に滴加する。8時間後、反応材料をクロロホル
ムで抽出し、有機相を分離し、かつクロロホルムを留去する。油状の残留物を引
き続き25%塩酸100mlで2時間100℃に加熱する。冷却後固体の粗生成
物を分離し、かつエタノールから再結晶する。
The present invention relates to a novel imidazoline-2,4-dione having an aryl (or aralkyl) group substituted at the 1-position and having an anticonvulsant action, and a process for producing the same.
