誘電泳動 技術分野
本発明は、複合試料物質、特に食品由来物質から微生物を濃縮および分離する
ための方法および装置に関するものである。細菌および酵母が好適な標的微生物
である。従来技術
食物における微生物の検出は、特に食物中毒の状況を回避することに関連した
食品製造および卸売り工業だけでなく、培養物などにおける所望の微生物を定量
するためにも関心が高まっている。
従来の検出方法は、関心のある微生物を濃縮した後に同定工程を行う培養技術
を中心とする傾向があり、たとえばEP−A−0 214 340号[ビオコン
トロール・システムス]に開示されている。この種の方法は必ずかなり時間と労
力がかかる。これは、たとえば食品の試料をたとえば工場から出荷される前に正
規の試験を行って汚染バッチを留置するような予備活動手段のための適性を低下
させる。
基礎となる一層迅速な技術が、たとえば「食品微生物学にお
ける迅速分析技術」[P.D.パテル編、ブラッキー]のような刊行物に提案さ
れている。しかしながら、たとえば電気技術および免疫学的技術を実際に使用す
るのは、試料としての食品の複雑かつ不均質な性質により大いに制限されている
。したがって、食品および他の複合試料における微生物の迅速分析を行う実用的
方法が当業界で長期にわたり待たれていることが判るであろう。本発明
本発明者等は、誘電泳動(dielectrophoresis)を用いる複
合試料(特に食物由来試料)における微生物の実用的分析方法を開発した。
誘電泳動は、特に分極した非帯電粒子における不均一な電場により誘発される
運動を特徴とする現象である[英国特許出願GB 2 071 843号(ポー
ル)]。これは、電気泳動で使用される直流フィールドでなく交流フィールドを
用いて生ずる運動である。
たとえば細菌および酵母のような微生物の運動は、誘電泳動原理を用いて操作
しうることが周知されている[たとえばアセンカー等(1990)、バイオエレ
クロケミストリー・アンド
・バイオエナージェチックス、第24巻、第203〜214頁;マルクス等(1
994)、マイクロバイオロジー、第140巻、第585〜591頁;およびホ
ークス等(1993)、マイクロビオス、第73巻、第81〜86頁参照]。さ
らに各種の細菌形態を同様に管理しうることも周知である[たとえばアーチャー
等(1993)、マイクロビオス、第73巻、第165〜172頁参照]。
さらに、粒子(特に細菌)の混合物を分離する誘電泳動装置の能力が、誘電泳
動室を通過する試料液における粒子の遅延差を用いて示されている(EO 93
/20927号(BTG))。この研究は、変化する電場を用いて特定細菌の分
離を達成することに集中しているが、液体のpHおよび導電率、並びに電圧の周
波数も遅延およびしたがって分離の順序および程度に影響を及ぼすことが認めら
れている。
しかしながら、今日までの電気泳動に関する公開された研究は、一般に比較的
単純な液体試料の使用に関するものであり、特に種々異なる誘電泳動特性を有す
る細菌の特性化および分割に関するものであった。たとえばGB 2 071
843A号では、試料をその目的につき特に増殖させた純粋培養物から
採取した。いずれの場合も、「実社会」の食物試料を試験するに際し誘電泳動は
実用されていなかった。
本発明者等は、微生物の直接的かつ積極的な誘電泳動凝集を高導電率かつ高誘
電率の食物および培地ブロスから1〜10ボルトにおける1〜20MHzの電圧
設定を用いては達成しえないこと、並びに1:10のブロスの希釈がこの状況を
修復しないことを突き止めた。しかしながら1:100〜1:100,000の
希釈は、これら周波数および電圧のパラメータを用いて或る程度凝集をもたらす
ことが判明した。しかしながら実用目的には1:1000〜1:10000の希
釈が15ボルトまでの使用を可能にする最良の結果を与え、それより高いと加熱
効果によって対流が発生する。
試料の希釈は、濃縮を意図する任意の方法を実施する前に試料における細菌の
濃度を減少させることが避けられず、したがって原理的に望ましくない。これは
特に、少なくとも細菌が存在する場合は低濃度でのみ存在しうるような試料(た
とえば食物試料)を取扱う場合である。
今回、本発明者等は、たとえば食物から得られるような複合試料を先ず最初に
希釈することなく誘電泳動により分析しうる
方法を突き止めた。
したがって本発明の第一面においては、誘電泳動の使用を特徴とする食物試料
における微生物の濃縮方法を開示する。
濃縮自身は、試料を誘電泳動フィールドに暴露させて微生物を凝集させると共
に残余の試料を除去して達成することができる。濃縮された微生物を次いでたと
えば下記するように分析することができる。
「食物試料」という用語は、培養またはたとえばキャリヤ液に懸濁させること
により予備処理されている試料を含む、ヒトもしくは動物の食品または潜在的食
品から得られる任意の液体材料を意味する。
好ましくは、この方法は細菌、たとえばサルモネラもしくはリステリア・スペ
シースの分析につき使用される。すなわち、細菌の存在につき食品素材を迅速分
析するための誘電泳動の使用は製造もしくは流通の早期段階にて食品工業におけ
る活動介入の道を開き、汚染食品が最終消費者に到達する傾向を減少させる。
この方法は、たとえば1000μS/cm2より高い初期導電率を有する試料
に特に適用しうる。この種の試料につき、こ
の方法は試料を1000μS/cm2未満の導電率まで減少させるよう処理した
後に誘電泳動を行うことをさらに特徴とする。導電率減少の好適方法は脱塩であ
る。
本発明者等は、脱塩の過程が微生物の迅速な誘電泳動分離を可能すると共に、
食物粒子からの干渉および試料の希釈に関連する欠点を最小化させることを突き
止めた。
この面の本発明の好適方法においては、導電率を500μS/cm2もしくは
それ以下まで、より好ましくは1〜450μS/cm2まで、特に好ましくは約
100〜200μS/cm2まで減少させる。
実用目的には50〜450μS/cm2の導電率が許容しうる。効率的な電気
泳動には極めて低い導電率が望ましいことも明かであるが、達成するには一層大
きい努力または長い時間を必要とし、或る程度の試料微生物の損失をもたらしう
る。したがって脱塩技術および選択される導電率範囲は、これら因子のバランス
とせねばならない。本発明者等は、GB 2 071 843A号に教示された
「10μS/cm未満」の好適範囲よりもずっと高い導電率で本発明を用いて成
功したことに注目すべきである。
用いる特定の脱塩処理は、試料から相当量の細菌を損失してはならない場合を
除き限定されない。
本発明者等により実現された便利な方法は、脱塩用ゲル[たとえばバイオ・ゲ
ルP−6DG、ビオラド・ラボラトリース社およびPD−10、ファルマシア社
]、細菌を保持する寸法の透析膜、イオン交換樹脂、逆浸透、限外濾過またはイ
オンスペシフィックバランス変化の使用を包含する。さらに、単に遠心分離(た
とえば15分間にわたる4000rev/min)を用いて試料固形分を沈澱さ
せ、次いで得られたペレットをたとえば蒸留水もしくは脱イオン水に懸濁させる
だけでもよい。
たとえばゲルのような物質を用いて試料を脱塩する方法は変化させることがで
きる。たとえばエクスクルージョン・ゲル法を用いて種々異なる寸法の物質を保
持することができ;イオン交換樹脂を用いて溶液からイオンを直接除去すること
ができ;静止、低圧力および向流の透析を用いて塩類を効果的に洗浄除去するこ
とができ;圧力ドライブの逆浸透、微小濾過、向流拡散または特異的限外濾過、
透析濾過、線状クロスフローまたはデプスフィルタ配置を用いることもでき;或
いはたとえば電気
透析もしくは限外浸透で用いるような帯電システムを使用することもできる。
迅速であると共に、ゲルが試料の微粒子特性を減少させるコースフィルタとし
て作用することによりその後の誘電泳動を単純化させうる点で、ゲル濾過の使用
が特に有利である。架橋デキストランゲルが特に効果的であると思われる。
透析カセットおよび透析室(チャンバー)は、比較的未熟な操作員でさえ使用
が簡単かつ便利である点において有利である。
連続フローシステムはさらに脱塩を単純化させ、したがって全体的な微生物濃
縮過程を単純化させうる。
上記脱塩工程の使用から生ずる利点は、全ての高導電率(たとえば>1000
μS/cm2)の複合液体試料(非食物起源のものでさえ)の誘電泳動でも得ら
れることに注目すべきである。すなわち、この種の方法およびこの方法を用いる
装置も本発明の他面を構成する。
使用する誘電泳動フィールドのパラメータは典型的には1〜15ボルトにて1
〜20MHzの周波数を含みうるが、これは20MHzより高い周波数を用いる
場合は100ボルトもしくはそれ以上まで増大させることもできる。これは、試
料の導電
率がこの種の高電圧の使用を可能にしないような従来技術の誘電泳動技術よりも
有利である。試料の残部(すなわち微生物を除く試料)がフィールドから除去さ
れた後、フィルードを消失させると共にキャリヤ液を微生物が凝集した領域を通
過させることにより微生物自身をキャリヤ液にて分離することができる。キャリ
ヤ液は便利には緩衝溶液、塩水または水、たとえば脱イオン水もしくは蒸留水で
ある。
本発明の好適方法を使用し、未希釈の食物および培地由来の試料(たとえば食
物ブロスおよび培養ブロス)の直接的な積極的誘電泳動を行って、全微生物(た
とえば細菌)内容物または選択された微生物成分集団を急速濃縮することができ
る。
第二面において本発明は食物試料における微生物の分析方法をも提供し、この
方法は上記したように微生物を濃縮した後に微生物を分析する工程を含む。
好ましくは微生物を分析前にフィールドから移動させる。微生物を分析すべく
使用する方法は任意特定の技術群に限定されず、誘電泳動フィールド領域(たと
えばフィールドを加えるべく使用した電極の端部)の直接的な肉眼観察を含め顕
微鏡を用いる直接的な肉眼観察のように簡単とすることができる。この
種の技術は当業者に周知されている。
第三面において本発明は承認バッチの食品の製造方法をも提供し、この方法は
食品バッチを常法で作成し、このバッチから代表的試料を採取し、上記の誘電泳
動法を用いて代表的試料における微生物を分析し、分析の結果を標準と比較し、
さらに分析が標準に合致し或いは標準を越える(上回る)場合はバッチを承認す
ることからなっている。
「代表的試料」という用語は、全体の品質を統計上有意なレベルで判定しうる
試料を意味する。使用する標準は食品の性質および分析する微生物に依存するが
、多くの場合は所定の微生物(たとえばサルモネラ)が完全に不在であることを
必要とする。
さらに他面において本発明は1000μS/cm2より高い導電率を有する複
合液体試料からの微生物の濃縮もしくは分離を行う装置をも提供し、この装置は
導電率を1000μS/cm2もしくはそれ以下の数値まで減少させるよう試料
を処置する手段と、誘電泳動フィールドを発生させる手段と、処理された試料を
誘電泳動フィールド中を通過させる手段とを備える。分離目的で装置は好ましく
は電場を消失させうるように制御す
る手段と、キャリヤ液を誘電泳動フィールドを発生させる手段を通過させて凝集
した微生物を集める手段と、通過したキャリヤ液を収集する手段とを備える。
誘電フィールドを発生させる手段は任意特定の装置に限定されず、ただし試料
を使用に際し誘電フィールド保持領域に対し通過させうるものとする。フィール
ドは便利には電極/フィールド室の電極を用いて形成され、これらは電気泳動を
生ぜしめるのでなく誘電泳動フィールドを保持するために当業界で知られたよう
に離間させる。
以下、限定はしないが実施例および図面を参照して本発明の方法および装置を
実施例につき説明する。本発明の範囲内に入る他の実施例も、これらを参照して
当業者には明かとなるであろう。図面

下記する全ての実施例に使用した装置は、種々異なる直径および間隔を有する
多数の電極を備えた。狭い曲率半径(直径)を有するものは、最も狭い電極に隣
接した凝集の増大により注目される最高のフィールドを発生した。凝集の程度は
直径1〜11μmおよび間隔30〜135μmの電極にて最高であると共に、4
2〜43μmの直径および500μmの間隔にて最小であった。微生物の凝集を
第1図に示した装置を用いて観察し、ここでイメージ分析装置(1)はカメラ(
2)を介し顕微鏡(3)から得られた微小電極列(4)における誘電泳動フィー
ルドのイメージを受入れ、次いでこれは前記列の各電極の切換えマトリックス(
5)に関数発生器(6)を介し制御信号を供給した。実施例1:誘電泳動試料に対する希釈の効果:
第2図および第3図は、緩衝ペプトン水(BPW)の導電率および誘電泳動特
性に対する希釈の効果を示す。第4図および第5図はラッパポート・バシリアジ
ス(RV)媒地に関する対応する結果を示す。実施例2:遠心分離および再懸濁による食物予備富化試料の導電率の減少および 誘電泳動:
牛肉、鶏肉または鱈の試料(10g)を90mL容積の無菌BPWに添加し、
1分間消化させ[コルワース消化装置、A.J.シュチワート・リミテッド]、
次いで37℃にて18時間培養した。培養の後、1mLづつのホモゲナイズ物を
4000rev/minにて15分間にわたり遠心分離し、ペレットを蒸留水に
再懸濁させた。懸濁物における天然細菌叢の誘電泳動凝集を10KHz、100
KHz、1MHzおよび10MHzにて測定した。その結果を第6図に示す。実施例3:脱塩カラムの使用による細菌培養物の導電率の減少および誘電泳動:
約107〜108個の微生物/mLを含有するトリプトン大豆ブロス(TSB)
におけるアリコート(1mL)の細菌培養物を、予め121℃にて15分間オー
トクレーブ滅菌された市販の脱塩カラム(ビオ−ゲル、P6DG、ビオラド・ラ
ボラトリース社)を通過させた。溶出液を100μLづつのフラクションにて集
め、各フラクションにおける細菌の誘電泳動凝集を100KHzおよび10ボル
トにて測定した。その結果を第7図に示す。実施例4:脱塩カラムの使用による食物予備豊富化試料の導電率の減少および誘 電泳動:
牛肉、鶏肉および鱈の試料(10g)を90mL容積の無菌BPWに添加し、
37℃にて18時間にわたりインキュベートした。インキュベーションの後、1
mLの富化ブロスのアリコートを2mLの水和脱塩ゲル(ビオ−ゲル、P−6D
G、ビオスピン)を含有するカラムを通過させ、溶出液を100μLづつのフラ
クションにて集めた。各フラクションを導電率、全生存カウント数(トリプトン
大豆寒天:TSA)における塗沫プレート法を用いる)、並びに100KHzお
よび10ボルトにおける誘電泳動凝集につき分析した。その結果を第8、9およ
び10図に示す。実施例5:細菌培養物および食物予備富化試料の導電率の減少:脱塩カラム、透 析カセットおよび透析室の比較: 方法
ストレプトマイシンおよびナリジクス酸に対し耐性であるS.チフィムリウム
(55698)およびS.エンテリジチス(p48120)の菌株をTSBにて
1晩培養した。2.5mLの培養物を脱塩ゲル(PD10、ファルマシア社)の
カラム(1.5cm×1cm)に施し、蒸留水で溶出させた。0.5
μLの溶出液のアリコートを集め、小型導電率測定計[ホリバ・リミテッド]を
用いて導電率を測定した。各フラクションにおけるサルモネラを計数し、これは
1/4濃度のリンガー溶液でシリーズ希釈し、ストレプトマイシンおよびナリジ
クス酸を含有するTSAに0.1mL表面塗沫して測定した。各プレートを37
℃にて24時間にわたり培養した。
さらに脱塩カラムを、サルモネラの2種の抗生物質耐性菌株が接種されたココ
ナッツおよび鳥肉の予備富化培養物を用いて評価した。25gづつの各食物の試
料を225mLのBPWに添加した。次いで予備富化培養物にS.エンテリジチ
スもしくはS.チフィムリウムのいずれかの1晩培養物の10-5希釈液からの1
00μLを接種した。培養された予備富化物の試料(0.5mL)を深さ1cm
のカラムに施し、各フラクションを導電率測定およびサルモネラの計数につき上
記と同様に集めた。
透析カセット[スライド−A−ライザー、ピアス・アンド・ワリナー社]にS
.チフィムリウムが接種された1mLのTSBを満たし、500mLの蒸留水に
対し室温(RT)および4℃にて撹拌しながら透析した。100μLづつのアリ
コートを所定時間間隔にて導電率測定につき取り出した。上記のように
透析の前後でサルモネラを計数した。
両側透析室[スピン・ビオダイヤライザー、シアロメド・インコーポレーショ
ン]に0.1μmもしくは0.45μmのポリカーボネート膜を装着し、L.モ
ノサイトジーンが接種された1mLのTSBを満たし、500mLの蒸留水に対
し室温にて撹拌しながら透析した。透析の前後にリステリアを計数し、これは1
/4濃度のリンガー溶液でシリーズ希釈し、PALCAM寒天[オキソイド社]
に対し0.1mLの適する希釈物を表面塗沫して行った。
結果
PD10脱塩用ゲルを用い、9.6×107cfu/mLのS.エンテリジチ
スおよび1.2×108cfu/mLのS.チフィムリウムを含有するTSB培
養物の導電率はそれぞれ>2,000から84μS/cm2までおよび117μ
S/cm2までカラムからの第2フラクションにて5分間以内に低下した。フラ
クション2におけるS.エンテリジチスおよびS.チフィムリウムの対応の回収
率はそれぞれ100%および17%であった。
2種の血清型のサルモネラを接種したココナッツおよび
鶏の予備富化培養物の場合、富化物の導電率は脱塩前の>2,000μS/cm2
から脱塩後のフラクション2における10〜14μS/cm2まで減少した。5
.5×105cfu/mLのS.チフィムリウムを含有するココナッツおよび鳥
肉予備富化物につき、脱塩後の回収率はそれぞれ6%および10%であった。2
.1×105S.エンテリジチスを含有するココナッツおよび鶏肉予備富化物に
つき回収率はそれぞれ57%および20%であった。
室温にてスライド−A−ライザー透析カセットはTSBの導電率を1時間後に
>2,000から530μS/cm2まで、及び6時間後に155μS/cm2ま
で減少させた。log10サルモネラカウント数は初期の8.6から6時間後の9
.0まで僅かに増大した。4℃にて脱塩の割合が僅かに低下し、最終導電率は僅
かに高くなった(第11図)。スピン・ビオダイヤライザーはスライド−A−ラ
イザーと同様な割合の脱塩を与えたが(第12図)、最終導電率は相当に低かっ
た(36μS/cm2)。脱塩の割合および最終導電率において、0.1μmの
膜と0.45μmの膜との間に差はなかった。リステリアカウント数は初期の8
.6から6時間後の8.1まで僅かに減少した。
したがって脱塩カラムは食物富化物のイオン濃度を積極的誘電泳動を可能にす
るレベル(<500μS/cm2)まで急速に減少させる手段として効果的であ
る。カラムを通過した後のサルモネラの回収率は血清型と共に変化した。透析カ
セットおよび透析室は食物予備富化物を脱塩するための簡単かつ便利な方法にな
りうるが、スライド−A−ライザーおよびスピン・ビオダイアライザーによるT
SBの脱塩の割合はゲルカラム系を用いる場合よりもずっと遅かった。磁気攪拌
機と共に使用するための組込み磁石を備えた両側透析室は透析カセットよりも大
きい導電率減少を与えた。実施例6:3mL容量の透析カセットを用いる食物予備富化試料の導電率の減少 :
方法
鶏肉およびココナッツの試料(25g)を225mL容積のBPWに秤量して
入れ、37℃で1晩インキュベートした。インキュベーションの後、富化培養物
にTSB中のS.チフィムリウムの1晩培養物の1/1000希釈物を1mL接
種した。富化培養物の試料(1mL)を透析カセットに移し、1Lの蒸留水に対
し透析した。富化培養物のアリコート(100μL)
を、カード型導電率計[ホリバ社]を用いる導電率測定のため1時間間隔で透析
カセットから取り出した。透析の前後にサルモネラカウント数を測定し、これは
マクシマム・レカバリー・ダイルーエント(MRD)における懸濁物の一連の1
0倍希釈物を作成すると共にキシロース・リジン・デスオキシコレート寒天(X
LD)に対し0.1mLを表面塗沫することにより行った。
結果
カセットは鶏肉およびココナッツ富化培養物の導電率を>2000μS/cm2
から260μS/cm2まで室温にて1時間後に減少させた。その後、脱塩の割
合は鶏肉およびココナッツ富化物の導電率をそれぞれ2時間後に160μS/c
m2および179μS/cm2まで低下させ、さらに最終的に6時間後に約100
μS/cm2に達した。透析された鶏肉およびココナッツ富化物におけるS.チ
フィムリウムの個数は顕著には変化しなかったが、それぞれ室温にて6時間後に
0.6および0.3 logサイクルで増大した。
したがって、これらカセットは誘電泳動に先立ち食物富化培養物を急速(たと
えば2〜3時間)脱塩する簡単かつ便利な方法を与え、この過程に際し細菌数の
喪失がない。実施例7:15mL容量の透析カセットを用いる食物予備富化試料の導電率にお ける減少および誘電泳動:
方法
ミンチ処理した牛肉、鶏肉、スキムドミルク粉末およびココナッツの富化培養
物を作成し、これらに実施例6に記載したようにS.チフィムリウム(S596
8)の抗生物質耐性菌株を接種した。大容積カセット(10,000)MWカッ
トオフの15mL容量[ピアス・アンド・ワリナー社]に15mLづつの富化培
養物を充填した。これら培養物を、1時間間隔で移動させながら3Lの蒸留水に
対し透析した。100μLづつの懸濁物アリコートを所定時間間隔で、透析の前
後における導電率測定およびストレプトマイシン(1mg/mL)およびナリジ
キシン酸(50μg/mL)(XLNS)を含有するキシロース・リシン寒天に
おけるサルモネラカウントのため取出した。透析の終了後、脱塩された富化培養
物の1部を誘電泳動セルに移し、光学顕微鏡および×80倍対物レンズを用い2
0MHz 20 VにてSMPにおける全微生物叢の誘電泳動分離につき観察し
た。
結果
15mL容量の透析カセットを用い鶏肉、ミンチ牛肉(MB)、スキムドミル
