同定方法

本発明は、薬理学的活性を有する作用剤の改良された同定方法に関する。
発明の背景
過去10年間において、例えば、細胞表面リセプター、酵素または抗体のごと
き種々の細胞アクセプター分子のアゴニストまたはアンタゴニストのような薬理
学的活性を有する作用剤の製造および同定に対する需要が増大してきた。種々の
生物学的プロセスを有効に転調させうる新たな化学的部分の検索において、検索
を行うための標準的方法は、以前から存在する種々の化学的部分、例えば、天然
に存在する化合物または合成ライブラリーもしくはデータベースに存在する化合
物をスクリーニングすることである。スクリーニングすべき化学的部分の特定の
性質を試験するために設計されたアッセイ、例えば、化学的部分の特定リセプタ
ー部位への結合能を試験するリセプター結合アッセイに供することによって、以
前から存在する化学的部分の生物学的活性が決定される。
生物学的活性について非常に多くのランダムに選択された化合物をスクリーニ
ングすることに費やされる時間および経費を少なくする努力において、リード化
合物(lead compounds)の発見のための化合物のライブラリーを提供するために
いくつかの開発が行われている。分子多様性を化学的に得ることは、新規リード
構造の検索における主要な道具となっている。現在のところ、分子多様性のある
多数の化合物を化学的に得るための知られている方法は、一般的には、固相合成
の使用、詳細にはペプチドおよびペプチドライブラリーの合成および同定を包含
する。例えば、一定量のペプチドを樹脂から遊離させて可溶性形態でアッセイし
、その一方ではまだ樹脂に結合しているいくつかのペプチドを樹脂上で配列決定
で
きるような、ペプチドと樹脂との間の選択的開裂可能リンカーを得るための方法
論を開示しているLebl et al.,Int.J.Pept.Prot,Res.,41,p.210(1993)参照。さ
らに、ポリスチレンまたはポリアクリルアミド樹脂のごとき固体支持体上での直
鎖状ペプチドの合成方法を開示しているLam et al.,Nature,354,p.82(1991)およ
びWO92/00091;96ウェルのマイクロタイタープレートの配列に対応
するようにブロック上に配列された誘導体ポリスチレンピン上でのペプチド合成
を開示しているGeysen et al.,J.Immnol.Meth.,102,p.259(1987);可溶性ペプチ
ドのプールを含む抗体またはリセプター結合配列のデノボ決定のためのアプロー
チを開示しているHoughten et al.,Nature,354,p84(1991)およびWO92/09
300も参照。
ペプチド模倣物のごとき非ペプチド性有機化合物は、しばしば、ある種の酵素
のリセプターに対するアフィニティーにおいてはペプチドリガンドを上回ること
がある。高アフィニティー生物学的リガンドおよび究極的には新しく重要な薬剤
を迅速に同定するための有効な方法は、生物学的アクセプター(例えば、リセプ
ターまたは酵素)との1またはそれ以上のタイプの相互作用(例えば、水素結合
、塩結合、パイ−錯形成反応、疎水的効果等)を確立しうる種々の構造単位を含
む非ペプチド構造の迅速な構築およびスクリーニングを必要とする。しかしなが
ら、非ペプチド分子を含む合成試験化合物ライブラリーの形成およびスクリーニ
ングについての仕事はまだ揺籃期にある。この領域の一例は、固体支持体上での
ベンゾジアゼピン類の組み合わせ合成(combinatorial synthesis)に関するEll
manとBuninの仕事である(J.Am.Chem.Soc.114,10997,(1992);Chemical and Eng
ineering News,January 18,1993,33頁参照)。
歴史的には、薬理学的に活性のある作用剤の同定方法は、時間がかかり、労力
を要し、不十分なものと特徴づけられており、通常には、1のスクリーニングに
つき1の生物学的標的が関連している。さらに、方法の改善のための努力は、専
ら1のスクリーニングあたりの作用剤の数の増大および質の向上に焦点が絞られ
て来た。現在まで、生物学的標的をスクリーニングアッセイに用いる効率を高め
ることを強調したものは、たとえあったとえも殆ど存在しない。
さらに、ヒト・ゲノムを完全に特徴づけるために努力が払われてきた。この先
取的研究は、すでに数千の新規遺伝子配列を見いだしたが、その多くは機能が不
明である。これらの遺伝子のいずれのものが薬理学的介在のための機会を提供す
るのかを確認することは、現在の方法論を用いれば多年にわたる勤勉な努力を要
するであろう。薬理学的に活性のある作用剤を効率よく同定し、ヒト・ゲノムに
ついての先取的研究から得られた情報をよりよく利用するための、改良された方
法に対する必要性が現在のところ存在する。
本発明は、知られている方法の多くの不利な点ならびに満足されていない多く
の必要な点を狙うものであり、以後より詳細に説明される本発明は、知られてい
る方法に優る多くの利点を提供する。
発明の概要
本発明は、個別にタグを付された複数の標的作用剤を同時発現させて標的ライ
ブラリーを形成し、作用剤候補プールを調製し、次いで、所望特性を有する候補
プールの作用剤を同定するアッセイにおいて標的ライブラリーおよび作用剤候補
プールを試験することを特徴とする、薬理学的に活性のある作用剤の同定方法、
ならびにかかる方法により同定される薬理学的に活性のある作用剤に関する。
また本発明は、個別にタグを付された複数の遺伝子を同時発現させて標的ライ
ブラリーを形成する方法に関する。
また本発明は、個別にタグを付された複数の遺伝子産物を同時発現させて標的
ライブラリーを形成する方法に関する。
また本発明は、個別にタグを付された複数の膜結合リセプターを同時発現させ
て標的ライブラリーを形成する方法に関する。
また本発明は、個別にタグを付された複数の発現した遺伝子を含んでなる標的
ライブラリーに関する。
また本発明は、個別にタグを付された複数の発現した遺伝子産物を含んでなる
標的ライブラリーに関する。
また本発明は、個別にタグを付された複数の発現した膜結合リセプターを含ん

発明の詳細な説明
本明細書の用語「作用剤候補のプール」は、薬理学的活性について試験される
個々の化合物、ペプチド、天然産物およびモノクローナル抗体の混合物のごとき
化学的化合物からなる群より選択される1またはそれ以上の物質の源を意味する
。組み合わせ化合物ライブラリー、組み合わせペプチドライブラリーまたはバリ
オマー(variomers)としてこれらの物質を試験してもよい。
本明細書の用語「標的ライブラリー」は、個別にタグを付された複数の発現し
た標的作用剤を意味する。
本明細書の用語「個別にタグを付された作用剤」は、個別にタグを付された遺
伝子、個別にタグを付された遺伝子産物または個別にタグを付された膜結合リセ
プターを意味する。
本明細書の用語「遺伝子産物」は、遺伝子の発現により生じる蛋白を意味する
。例えば、酵素、リセプターまたはSH2ドメインのごときドッキング蛋白を意
味する。
本明細書の用語「個別にタグを付された遺伝子」は、それぞれが標的ライブラ
リーを形成するであろう遺伝子であって、発現の前に、特定の抗体により認識さ
れうる短いペプチド配列(以後、エピトープタグという)のごとき別々のマーカ
ーで処理された遺伝子を意味する。該エピトープタグは多数製造することができ
る。さらに、いくつかの遺伝子を同じタグを用いてグループ分けすることができ
、異なるタグの組み合わせを用いてサブグループに分けることもでき、あるいは
直列タグを集合させて引き続き行う選択を可能にすることもできる。
本明細書の用語「個別にタグを付された遺伝子産物」は、それぞれが標的ライ
ブラリーを形成するであろう遺伝子産物であって、発現の前に、特定の抗体によ
り認識されうる短いペプチド配列(以後、エピトープタグという)のごとき別々
のマーカーで処理された遺伝子産物を意味する。該エピトープタグは多数製造す
ることができる。さらに、いくつかの遺伝子産物を同じタグを用いてグループ分

本明細書の用語「個別にタグを付された膜結合リセプター」は、それぞれが標
的ライブラリーを形成するであろう膜結合リセプターであって、発現の前に、特
定の抗体により認識されうる短いペプチド配列(以後、エピトープタグという)
のごとき別々のマーカーで処理され、次いで、適当なレシピエント細胞中に取り
込まれる膜結合リセプターを意味する。該エピトープタグは多数製造することが
できる。さらに、いくつかの膜結合リセプターを同じタグを用いてグループ分け
することができ、異なるタグの組み合わせを用いてサブグループに分けることも
でき、あるいは直列タグを集合させて引き続き行う選択を可能にすることもでき
る。細胞1個につき1の標的リセプタータイプの取り込みが好ましく、このこと
は、異なる数の細胞を混合することにより標的ライブラリーの相対的成分の調節
を可能にする。この例において、この細胞混合物は標的ライブラリーを構成する
であろう。
多くのリセプターは、リガンドに対する正しい結合および機能的応答のために
、細胞膜を必要とする(例、7つの膜内外G−蛋白に結合したリセプター)。本
明細書の用語「正しい結合および機能的応答のための細胞膜を必要とする受容体
、ならびに細胞中に取り込まれる受容体」は、「膜結合リセプター」という。一
般的には、膜結合型リセプターへの標的ライブラリースクリーニングアプローチ
の適用を、本明細書に示されたようにして行うことができる。膜結合受容体をス
クリーニングする好ましい方法は、作用剤候補のプールとして1種またはそれ以
上の化学的化合物または組み合わせ化学ライブラリーを用い、化合物または組み
合わせライブラリーの合成途中の鋳型に光活性化基を付与することである。かか
る基の例は、アジ化アリールまたはベンゾフェノンである。さらに、好ましくは
、
ビオチンのごとき容易に検出可能なマーカーを鋳型中に取り入れる。かくして、
好ましくは、各化合物は光活性化基および検出基の双方を有するものである。別
法として、標的に対するリガンドが知られている場合には、光活性化タグおよび
検出タグはリガンド中に取り込まれることだけを必要とし、競争により未標識化
合物をスクリーニングすることができる。本発明のこの好ましい例において、化
合物または組み合わせライブラリーを標的ライブラリーと混合し、放射線照射し
て光活性化基を活性化する。標的ライブラリーの細胞を溶解し、標的ライブラリ
ーのエピトープタグに対して生起した空間的にコードされた抗体の配列に混合物
を添加する。マーカーを検出するための試薬で抗体のマトリックスをプローブす
る(ビオチンを検出するにはストレプトアビジン−ペルオキシダーゼを用いる)
。一定の化合物または化合物の基と相互作用する標的のスペクトルを、検出マー
カーとエピトープタグの特定の部分集合との結合により確認することができる。
必要ならば、抗体マトリックス上での脱複雑化の前に検出マーカーの取り込みに
関して標的ライブラリーをプレスクリーニングすることができる。
本明細書の用語「同時発現」とは、対象遺伝子、遺伝子産物または膜結合リセ
プターの同一性の維持を考慮せずに、多くの個別にタグを付された標的作用剤を
一緒に発現させ、精製することを意味する。
本明細書の用語「アッセイ」、「スクリーニング」または「生物学的活性に関
する試験」は、薬理学的に重要な活性に関するすべての形態の試験を包含する。
標的ライブラリーの調製に際して、管理可能な数の遺伝子、好ましくは100
未満の遺伝子から始めることができる。好ましくは、例えば、興味ある組織にお
ける発現により、または配列相同性に基づく共通のファミリー(キナーゼのごと
き)における地位により、これらの遺伝子をグループ分けする。好ましくは、エ
ピトープタグを用いて各遺伝子に個別にタグを付し、同時発現させる。
標的ライブラリーの調製に際して、管理可能な数の遺伝子産物、好ましくは1
00未満の遺伝子産物から始めることができる。好ましくは、例えば、興味ある
組織における発現により、または配列相同性に基づく共通のファミリー(SH2
ドメインのごとき)における地位により、これらの遺伝子産物をグルー
プ分けする。好ましくは、エピトープタグを用いて各遺伝子産物に個別にタグを
付し、同時発現させる。
標的ライブラリーの調製に際して、管理可能な数のリセプター、好ましくは1
00未満のリセプターから始めることができる。好ましくは、例えば、興味ある
組織における発現により、または配列相同性に基づく共通のファミリー(7つの
膜内外G−蛋白に結合した受容体のごとき)における地位により、これらの受容
体をグループ分けする。好ましくは、エピトープタグを用いて各受容体に個別に
タグを付し、同時発現させる。
標的ライブラリー中の標的の機能を混乱させる化合物を同定するためには、作
用剤候補のプール、好ましくは、1種またはそれ以上の化学的化合物または組み
合わせ化合物ライブラリーを、好ましくは、標準的方法により調製する。知られ
ているアッセイ方法により、または標的ライブラリーに含まれる予想活性に基づ
いて選択工程に手を加えることにより(下記実施例記載のごとく)、活性作用剤
を同定する。エピトープタグの手段により標的ライブラリーの単純化を行うこと
ができるが、質量スペクトル法のごとき分析方法を作用剤候補のプールの分析に
用いることもできる。かくして、活性作用剤の同定および個別にタグを付された
標的作用剤の機能を並行して分析する。
上記方法により同定された薬理学的に活性のある作用剤を、興味ある機能につ
いての細胞をベースにしたアッセイ、次いで、機能のスクリーニングまたは動物
モデルにおいてスクリーニングしてもよい。本明細書開示の方法により同定され
た薬理学的に活性のある作用剤も本発明の範囲に包含される。罹病している特定
の標的(遺伝子、遺伝子産物またはリセプター)を同定することは必ずしも必要
でないが、毒性の問題が生じた場合にはこの情報は潜在的に有用であり、容易に
遡及的に得ることができよう。さらに、細胞をベースにしたアッセイの組におい
て同時に評価される複数の化合物に関して、2次スクリーニンにおいても並行性
を維持する。
本発明の特に有利な態様は、発見プロセスの全局面、すなわち標的選択、発現
および精製、化合物合成、1次および2次スクリーニングにおいて並行性を維持
できることである。本発明のさらなる利点は:機能または疾病についての前知識
なしに複数の標的を同時に評価できること、もともと備わっている選択性の特徴
づけ、全く予期されない活性および疾病の徴候が明らかとなる機会が多いこと、
より多くの薬剤候補を単位時間あたり用いることによる向上した効率を包含する
。
上記並行アプローチの一般的骨格中において、標的ライブラリーをスクリーニ
ングするための多くの方法論が可能である。1のかかる方法論を実施例1に記載
する。すべてのかかる方法論は本明細書にて権利請求されている本発明の範囲内
である。
本発明のさらなる態様において、発現された個別にタグを付された標的作用剤
を用いる、標的ライブラリーのスクリーニングのための好ましい方法が提供され
、該方法においては、タグ付けをするマーカーはエピトープタグであり、該標的
ライブラリーは基材の別々の領域において空間的にコードされている(本明細書
において、「空間的にコードされた標的ライブラリー」または「SETL」という
)。本発明において使用する空間的にコードされた標的ライブラリーをi)〜iii
)により調製する:
i)抗体の組を、好ましくはモザイクパターンとして列をなして基材上に沈着
させること。それぞれの組は、標的ライブラリーの異なるエピトープタグおよび
少なくとも1つの抗体の組に対応した標的ライブラリーの各エピトープタグに指
向される。好ましくは、基材はポリビニリデンジフルオリド(PVDF)膜、ニ
トロセルロース膜、ポリスチレン層またはシリコンウエハーである。
ii)不活性蛋白での膜の抗体領域のブロッキング、次いで
iii)抗体領域への標的ライブラリーの曝露。
抗体の組は、それらが対応しているエピトープタグに選択的に結合し、そのこ
とにより、個別にタグを付された作用剤の類似のグループが膜の特定の領域に局
在化されるであろう。上記方法を繰り返し、複数の機能的に類似したSETLs
を得る。
いずれかの手段、例えば、毛細管ピペットを用いて手動で膜にスポットするこ
と、または前以て決定されている小さな沈着領域における「微小スポッティング
」
により自動化された方法(例えば、インクジェットプリンターに使用されるよう
な印刷手法を用いることによる)によって膜への抗体の沈着を行うことができる
。好ましくは、上記工程iiの前に固定化抗体を担持している膜を容器(例えば、
Merrifield合成容器、フラスコ、または好ましくはマイクロタイター96ウェル
プレート)に結合させて、各容器が全抗体領域を含むようにする。
作用剤候補プールを他のアッセイ試薬とともに容器に分注する。検出工程後、
膜を映像化して、標的ライブラリーの個別にタグを付された作用剤の活性および
作用剤候補の効果を確認する。
後述の実施例2、3および4に示すように、有利には、リセプターまたは他の
結合蛋白、例えばSH2ドメインに関するスクリーニングアッセイを開発するた
めにSETLアプローチを用いる。
さらなる苦労なしに、当業者は、上記説明を用いて本発明を最大限に利用でき
ると確信する。それゆえ、以下の実施例は本発明の単なる説明であって、本発明
の範囲を限定するものと解してはならない。アッセイの実施例 実施例1:新規骨芽細胞キナーゼ
新たな骨の生産にかかわっている細胞タイプは骨芽細胞である。大規模配列決
定法を骨芽細胞cDNAライブラリーに適用した結果として、多くの新規な全長
のキナーゼが同定された。骨芽細胞様細胞由来の多くの全長のキナーゼのそれぞ
れに、1またはそれ以上のエピトープタグを用いて個別にタグを付す。例えば、
独自の同定タグを通常の精製タグと並べて結合させて精製工程を容易にすること
ができる。遺伝子にタグを付し、同時に受容可能宿主、例えばイー・コリ(E.co
li)またはバキュロウイルスにて発現させる。混合物としてのキナーゼ標的ライ
ブラリーのすべてのメンバーを同時に精製するためにタグを用い、独自のタグに
対する抗体を用いるウェスタンブロットにより該ライブラリーを特徴づける。エ
ピトープタグを介して標的ライブラリーの各メンバーを個別に固定化し、アイソ
トープ標識ATPとホスフェートアクセプターとの混合物を添加すること
により適当な基材を同定する。ホスホリレーションされた基材を質量スペクトル
法により同定する。
標準的方法により化合物の組み合わせライブラリーを合成する。標的ライブラ
リーは適当な基材に沿って空間的にコードされる(上述のごとく)。適当な基材
を合成して、それらがホスフェートアクセプターおよびエピトープタグの両方を
含み、それらが適切なキナーゼ標的とともに共局在化するようにする。試験化合
物をアイソトープ標識されたATPとともに各配列に添加する。配列を映像化し
て、ホスフェートが取り込まれた領域を同定する。
実施例1で得た活性作用剤を、骨形成と相関関係があるオステオカルシン放出
アッセイのごとき2次アッセイにおいてスクリーニングしてもよい。さらに、実
施例1で得た活性作用剤を、骨形成と無関係な機能についての細胞に基づくアッ
セイの組み合わせにおいてスクリーニングする。好ましくは、該細胞に基づくア
ッセイはリポーター遺伝子法を用いるものであり、例えば米国特許第5,401,
629号および第5,436,128号に記載されている。かかる方法は、リポー
ター遺伝子(例えば、ルシフェラーゼ)に結合され、適当な細胞中にトランスフ
ェクションされた興味ある種々の遺伝子(例えば、オステオカルシン)のプロモ
ーター領域を用いるものである。これを多能細胞系、例えば胚の幹細胞において
行う場合には、同じ細胞を異なるリポーター構築物とともに使用して種々の機能
(以後、「アッセイライブラリー」という)について同時にアッセイすることが
できる。よって、本発明の並行アプローチは2次アッセイのためのすべての方法
において維持され、効率が向上し、活性化合物の広範囲の特徴づけが可能となる
。次いで、好ましくは、活性作用剤を動物モデルまたはインビトロ機能アッセイ
において評価し、毒性についてスクリーニングする。実施例2:結合アッセイ
SETLアプローチを用いて、リセプターまたは他の結合蛋白、例えばSH2
ドメインについてのスクリーニングアッセイを形成することができる。SH2ド
メインに関するSETLアッセイを形成するために、該ドメインをエピトープタ
グを付した融合蛋白として発現させる。タグに対する抗体を上記のごとく膜に沈
着させる。タグを付したSH2ドメインの混合物を各ウェルに入れ、対応抗体と
結合させる。ビオチン化ホスホペプチドリガンド(各SH2標的に対するもの)
の混合物を試験化合物とともに添加する。ストレプトアビジン−ビオチン複合体
ならびに色、蛍光または増強された化学発光(ECL)を検出する試薬の添加に
よりビオチン化ペプチドリガンドの結合を検出する。膜表面上の検出試薬の活性
の映像化により個々のSH2ドメインへのリガンド結合の測定が可能となる。か
かる映像化を自動化することができる。
ヒト・src SH2ドメインを融合蛋白として発現させることによりこのア
ッセイを実際に行った。
ヒト・src SH2ドメインを含む融合蛋白を一般的配列:DET1−DE
T2−スペーサー−ek−src SH2として発現させた。DET1、DET
2、スペーサーおよびekを以下に説明する。DET1(「定義されたエピトー
プタグ」)(配列番号:1)は、ヒト・免疫不全ウイルスタイプI(HIV−1
)エンベロープ蛋白gp120(またはgp160)において見いだされる11
個のアミノ酸の配列である。HIV−1のgp120(またはgp160)の種
々のエピトープに対するモノクローナル抗体が当該分野において知られている(
例えば、米国特許第5,166,050号参照)。1の好ましい例はモノクローナ
ル抗体178.1(Thiriart et al.,J.Immunol.,143:1832-1836(1989))であり
、それを、酵母で発現されたBH10株由来のHIV−1のgp160分子(Ra
tner et al.,Nature,313:277-284(1985))でマウスを免疫することにより調製し
た。蛋白精製(アフィニティークロマトグラフィーによる)のために、そして1
78.1抗体を用いて融合蛋白を捕捉し固定化するアッセイを形成するために、
このタグを発現の検出(ウェスタンブロットによる)に使用した。DET2はニ
ッケル含有樹脂に結合するヘキサヒスチジン配列タグ(配列番号:2)であり、
精製目的に使用した。スペーサー(配列番号:3)を用いて、構築物中の必要な
位置においてBamHI制限部位を設計した。用語「−ek−」は、エンテロキ
ナーゼプロテアーゼの認識配列(配列番号:4)をいい、それは、
src SH2ドメインからのタグの最適除去を可能にし、かくして外来アミノ
酸を含まないsrc SH2ドメインが得られる。結晶像の研究には、外来アミ
ノ酸を含まないsrc SH2ドメインのほうがタグを付した蛋白よりも好まし
い。
必要な制限部位(BamHIおよびXbaI)がスペーサー−ek−src
SH2領域に隣接するようにし、そのことにより、別のスペーサー−ek−SH
2構築物が下記手順2記載のベクターのいずれか1つの中に容易に置換されてD
ET1−DET2−スペーサー−ek−SH2タグ付き蛋白を得ることができる
ように、DET1−DET2−スペーサー−ek−src SH2のそれぞれを
コードしているDNA配列を設計した。タグを付したSH2ドメイン全体がNd
eI−XbaIフラグメントとして除去されて、E.coli転写および翻訳調節配列
の下流に適当な間隔をおいて存在するNdeI部位およびXbaIに適合した下
流のクローニング部位を有する発現ベクター中に挿入されるように、DET1−
DET2−スペーサー−ek−src SH2構築物をコードしているDNA配
列も設計した。いずれの適当なベクター(例えば、pET-11a;Novagen,Inc.)も同
様に結果を生じるであろうが、本実施例に使用したベクターはE.coli発現ベクタ
ーpEA1KnRBS3であった。このベクターは、Shatzman,A.,Gross,M.およ
びRosenberg,M.,"Expression using vectors with phage lambda regulatory se
quences"(Current Protocols in Molecular Biology(F.A.Ausbelら編)中16.3
.1〜16.3.11頁)(Greene Publishing and Wiley-Interscience,N.Y.(1990
年))(以後、Ausbelらという)に記載された一連のベクターの誘導体である。
特別なベクターpEA1KnRBS3は、Bergsma et al,1991,J.Biol.Chem.266
:23204-23214に記載されている。
下記手順は、ニワトリ・srcおよびヒト・src SH2ドメインの発現を
説明する。まず、ニワトリ・src SH2ドメインをDET1−DET2−ス
ペーサー−SH2として発現させた。次いで、他方をニワトリ・srcのかわり
